The aim of this study is to prepare silanized polymeric nanoparticles for DNA isolation. Polymeric p(HEMA)-IMEO-PBA nanoparticles around 85.7 nm diameter, was obtained by surfactant free emulsion polymerization for DNA isolation. Synthesized nanoparticles for characterization studies were realized scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), and Zeta-size. Surface area, average particle size and size distribution were also performed. The surface area of synthesized silanized polymeric nanoparticles was 2460 m2/g. Synthesized polymeric nanoparticles were silanized with 3-(2-imidazoline-1-yl)propyl (triethoxysilane) (IMEO). After that, phenylboronic acid (PBA) which is DNA specific ligand were covalently binded to silanized polymeric nanoparticles. The amount of DNA adsorbed onto the p(HEMA)-IMEO-PBA nanoparticles first increased and then reached a saturation value at around 14.0 mg/mL of DNA concentration. The maximum adsorption was 672.41 mg/g silanized polymeric nanoparticles in the optimum adsorption medium. The maximum DNA adsorption was achieved at 4 °C. The overall recovery of DNA was calculated as 95%. In repetitive adsorption–desorption circles, it is observed not being important decrease in DNA adsorption capacities. The results were shown that silanized polymeric nanoparticles can be a good alternative for DNA isolation.  相似文献

Immobilized metal ion affinity chromatography (IMAC) is a useful method for adsorption of proteins that have an affinity for transition metal ions. In this study, poly(hydroxyethyl methacrylate-methacryloyl-l-tryptophan) (PHEMATrp) nanoparticles were prepared by surfactant free emulsion polymerization. Then, Cu(II) ions were chelated on the PHEMATrp nanoparticles to be used in lysozyme adsorption studies in batch system. The maximum lysozyme adsorption capacity of the PHEMATrp nanoparticles was found to be 326.9 mg/g polymer at pH 7.0. The nonspecific lysozyme adsorption onto the PHEMA nanoparticles was negligible. In terms of protein desorption, it was observed that adsorbed lysozyme was readily desorbed in medium containing 1.0 M NaCl. The results showed that the metal-chelated PHEMATrp nanoparticles can be considered as a good adsorbent for lysozyme purification.  相似文献

In this study, poly(2-hydroxyethyl methacrylate–glycidylmethacrylate) [poly(HEMA–GMA)] cryogels were prepared by radical cryocopolymerization of HEMA with GMA as a functional comonomer and N,N′-methylene-bisacrylamide (MBAAm) as a crosslinker. Iminodiacetic acid (IDA) functional groups were attached via ring opening of the epoxy group on the poly(HEMA–GMA) cryogels and then Zn(II) ions were chelated with these structures. Characterization of cryogels was performed by FTIR, SEM, EDX and swelling studies. These cryogels have interconnected pores of 30–50 μm size. The equilibrium swelling degree of Zn(II) chelated poly(HEMA–GMA)-IDA cryogels was approximately 600%. Zn(II) chelated poly(HEMA–GMA)-IDA cryogels were used in the adsorption of alcohol dehydrogenase from aqueous solutions and adsorption was performed in continuous system. The effects of pH, alcohol dehydrogenase concentration, temperature, and flow rate on adsorption were investigated. The maximum amount of alcohol dehydrogenase adsorption was determined to be 9.94 mg/g cryogel at 1.0 mg/mL alcohol dehydrogenase concentration and in acetate buffer at pH 5.0 with a flow rate of 0.5 mL/min. Desorption of adsorbed alcohol dehydrogenase was carried out by using 1.0 M NaCI at pH 8.0 phosphate buffer and desorption yield was found to be 93.5%. Additionally, these cryogels were used for purification of alcohol dehydrogenase from yeast with a single-step. The purity of desorbed alcohol dehydrogenase was shown by silver-stained SDS–PAGE. This purification process can successfully be used for the purification of alcohol dehydrogenase from unclarified yeast homogenates and this work is the first report about the usage of the cryogels for purification of alcohol dehydrogenase.  相似文献

A novel synthetic polymer selective for p-nitrophenylpalmitate was synthesized by molecular imprinting technique. We have combined the principle of molecular imprinting with the ability of histidine, glutamic acid and serine to form a catalytic cavity that can promote the catalytic degradation of p-nitrophenyl palmitate.For the creation of such catalytic sites we first synthesized appropriate monomers and used p-nitrophenyl palmitate as a template to synthesize the imprinted polymers and the binding characteristics of the polymers were evaluated. The optimum pH was determined by evaluating different pH values and the hydrolytic activity of synthetic lipase was evaluated in the framework of Micheaelis–Menten kinetics. In addition, the values of maximal rate: Vm (0.68 mM/min) and Michaelis–Menten constant, Km (1.4 × 10?2 mM) were obtained from Lineweaver–Burk plots for the imprinted polymeric catalyst.  相似文献

Molecular imprinted polymers are artificial, template-made materials with the ability to recognize and to specifically bind the target molecule. The aim of this study is to prepare supermacroporous cryogel with embedded bilirubin-imprinted particles which can be used for the selective removal of bilirubin from human plasma. N-methacryloyl-(l)-tyrosinemethylester (MAT) was chosen as the pre-organization monomer. In the first step, bilirubin was complexed with MAT and the bilirubin-imprinted poly(hydroxyethyl methacrylate-N-methacryloly-(l)-tyrosine methyl-ester) [MIP] monolith was produced by bulk polymerization. MIP monolith was smashed and the particles ground and sieved through 100 μm sieves. In the second step, the supermacroporous poly(hydroxyethyl methacrylate) (PHEMA) cryogel with embedded MIP particles [PHEMA/MIP composite cryogel] was produced by free radical polymerization initiated by N,N,N′,N′-tetramethylene diamine (TEMED) and ammonium persulfate (APS) pair in an ice bath. After that, the template (i.e., bilirubin) molecules were removed using sodium carbonate and sodium hydroxide. Compared with the PHEMA cryogel (0.2 mg/g polymer), the bilirubin adsorption capacity of the PHEMA/MIP composite cryogel (10.3 mg/g polymer) was improved significantly due to the embedded MIP particles into the polymeric matrix. The relative selectivity coefficients of PHEMA/MIP composite cryogel for bilirubin/cholesterol and bilirubin/testosterone were 8.6 and 4.1 times greater than the PHEMA cryogel, respectively. The PHEMA/MIP composite cryogel could be used many times without decreasing the bilirubin adsorption capacity significantly.  相似文献

The aim of this study is to investigate in detail the feasibility of poly(2‐hydroxyethyl methacrylate‐N‐methacryloyl‐(L )‐histidine methyl ester), PHEMAH membranes for purification of immunoglobulin G (IgG) from human plasma. PHEMAH membranes were prepared by photo‐polymerization technique. Then, Zn2+, Ni2+, Co2+, and Cu2+ ions were chelated directly on the PHEMAH membranes. Elemental analysis assay was performed to determine the nitrogen content and polymerized MAH was calculated as 168.5 μmol/g. The nonspecific IgG adsorption onto the plain PHEMA membranes was negligible (about 0.25 mg/mL). A remarkable increase in the IgG adsorption capacities were achieved from human plasma with PHEMAH membranes (up to 68.4 mg/mL). Further increase was observed with the metal‐chelated PHEMAH membranes (up to 118 mg/mL). The metal‐chelate affinity membranes allowed the one‐step separation of IgG from human plasma. The binding range of metal ions for surface histidines from human plasma followed the order: Cu2+ > Ni2+ > Zn2+ > Co2+. Adsorbed IgG was eluted using 250 mM EDTA with a purity of 94.1%. IgG molecules could be repeatedly adsorbed and eluted with the metal‐chelated PHEMAH membranes without noticeable loss in their IgG adsorption capacity. © 2011 Wiley Periodicals, Inc. Poly(hydroxyethyl methacrylate) (PHEMA) nanobeads with an average size of 300 nm in diameter and with a polydispersity index of 1.156 were produced by a surfactant free emulsion polymerization. Specific surface area of the PHEMA nanobeads was found to be 996 m2/g. Imidazole containing 3-(2-imidazoline-1-yl)propyl(triethoxysilane) (IMEO) was used as a metal-chelating ligand. IMEO was covalently attached to the nanobeads. PHEMA-IMEO nanobeads were used for the removal of copper(II) ions from aqueous solutions. To evaluate the degree of IMEO loading, the PHEMA nanobeads were subjected to Si analysis by using flame atomizer atomic absorption spectrometer and it was estimated as 973 µmol IMEO/g of polymer. The PHEMA nanobeads were characterized by transmission electron microscopy and fourier transform infrared spectroscopy. Adsorption equilibrium was achieved in about 8 min. The adsorption of Cu2+ ions onto the PHEMA nanobeads was negligible (0.2 mg/g). The IMEO attachment into the PHEMA nanobeads significantly increased the Cu2+ adsorption capacity (58 mg/g). Adsorption capacity of the PHEMA-IMEO nanobeads increased significantly with increasing concentration. The adsorption of Cu2+ ions increased with increasing pH and reached a plateau value at around pH 5.0. Competitive heavy metal adsorption from aqueous solutions containing Cu+, Cd2+, Pb2+ and Hg2+ was also investigated. The adsorption capacities are 61.4 mg/g (966.9 µmol/g) for Cu2+; 180.5 mg/g (899.8 µmol/g) for Hg2+; 34.9 mg/g (310.5 µmol/g) for Cd2+ and 14.3 mg/g (69 µmol/g) for Pb2+. The affinity order in molar basis is observed as Cu2+ > Hg2+ > Cd2+ > Pb2+. These results may be considered as an indication of higher specificity of the PHEMA-IMEO nanobeads for the Cu2+ comparing to other ions. Consecutive adsorption and elution operations showed the feasibility of repeated use for PHEMA-IMEO nanobeads.  相似文献

We modified microporous polyamide hollow fibers by acid hydrolysis to amplify the reactive groups and subsequent binding of Cibacron Blue F3GA. Then, we loaded the Cibacron Blue F3GA‐attached hollow fibers with different metal ions (Cu2+, Ni2+, and Co2+) to form the metal chelates. We characterized the hollow fibers by scanning electron microscopy. The effect of pH and initial concentration of human serum albumin (HSA) on the adsorption of HSA to the metal‐chelated hollow fibers were examined in a batch system. Dye‐ and metal‐chelated hollow fibers had a higher HSA adsorption capacity and showed less nonspecific protein adsorption. The nonspecific adsorption of HSA onto the polyamide hollow fibers was 6.0 mg/g. Cibacron Blue F3GA immobilization onto the hollow fibers increased HSA adsorption up to 147 mg/g. Metal‐chelated hollow fibers showed further increases in the adsorption capacity. The maximum adsorption capacities of Co2+‐, Cu2+‐, and Ni2+‐chelated hollow fibers were 195, 226, and 289 mg/g, respectively. The recognition range of metal ions for HSA from human serum followed the order: Ni(II) > Cu(II) > Co(II). A higher HSA adsorption was observed from human serum (324 mg/g). A significant amount of the adsorbed HSA (up to 99%) was eluted for 1 h in the elution medium containing 1.0M sodium thiocyanide (NaSCN) at pH 8.0 and 25 mM ethylenediaminetetraacetic acid at pH 4.9. Repeated adsorption–desorption processes showed that these metal‐chelated polyamide hollow fibers were suitable for HSA adsorption. © 2002 Wiley Periodicals, Inc. The aim of this study was to investigate in detail the performance for removal of heavy metal ions of beads composed of poly(2‐hydroxyethyl methacrylate) (pHEMA) to which N‐methacryloylhistidine (MAH) was copolymerized. The metal‐complexing ligand MAH was synthesized by using methacryloyl chloride and histidine. Spherical beads with an average size of 150–200 μm were obtained by the radical suspension polymerization of MAH and HEMA conducted in an aqueous dispersion medium. Owing to the reasonably rough character of the bead surface, p(HEMA‐MAH) beads had a specific surface area of 17.6 m2/g. The synthesized MAH monomer was characterized by NMR; p(HEMA‐MAH) beads were characterized by swelling studies, FTIR and elemental analysis. The p(HEMA‐MAH) beads with a swelling ratio of 65%, and containing 1.6 mmol MAH/g, were used in the adsorption/desorption experiments. Adsorption capacity of the beads for the selected metal ions, i. e., Cu(II), Cd(II), Cr(III), Hg(II) and Pb(II), were investigated in aqueous media containing different amounts of these ions (10–750 mg/L) and at different pH values (3.0–7.0). Adsorption equilibria were established in about 20 min. The maximum adsorption capacities of the p(HEMA‐MAH) beads were 122.7 mg/g for Cu(II), 468.8 mg/g for Cr(III), 639.4 mg/g for Cd(II), 714.1 mg/g for Pb(II) and 1 234.4 mg/g for Hg(II). pH significantly affected the adsorption capacity of MAH incorporated beads. The chelating beads can be easily regenerated by 0.1 M HNO3 with high effectiveness. These features make p(HEMA‐MAH) beads a potential candidate for heavy metal removal at high capacity.  相似文献

Molecularly imprinted polymer beads (PIBs) and non-imprinted (control) polymer beads (NIBs) have been prepared from methacryloylhistidine-Co2+, -Ni2+, and -Zn2+ monomers and applied as catalyst in the hydrolysis of paraoxon which is an organophosphate ester and used as a pesticide. The PIBs were prepared by a molecular surface imprinting technique. The catalytic performance of these polymers having Co2+, Ni2+ and Zn2+ ions in their catalytic active centers was evaluated according to the enzyme kinetic model of Michaelis–Menten and their activities were compared to each other. PIBs always showed higher catalytic activities than NIBs analogues. In addition, the imprinted polymers of methacryloylhistidine-Co2+, -Ni2+, or -Zn2+ complexes provided hydrolysis rate enhancements by a factor of 356, 241, and 95, respectively, compared to the rate in non-catalyzed media that contains only buffer.  相似文献
