This paper presents a new metallurgical model for the ausferritic transformation of ductile cast iron. The model allows predicting the evolution of phases in terms of the chemical composition, austenitization and austempering temperatures, graphite nodule count, and distribution of graphite nodule size. The ferrite evolution is predicted according to the displacive growth mechanism. A representative volume element is employed at the microscale to consider the phase distributions, the inhomogeneous austenite carbon content, and the nucleation of ferrite subunits at the graphite nodule surface and at the tips of existing ferrite subunits. The performance of the model is evaluated by comparison with experimental results. The results indicate that the increment of the ausferritic transformation rate, which is caused by increments of austempering temperature and graphite nodule count, is adequately represented by this model.

Skeletal muscles represent 40% of body mass and its native regenerative capacity can be permanently lost after a traumatic injury, congenital diseases, or tumor ablation. The absence of physiological regeneration can hinder muscle repair preventing normal muscle tissue functions. To date, tissue engineering (TE) represents one promising option for treating muscle injuries and wasting. In particular, hydrogels derived from the decellularized extracellular matrix (dECM) are widely investigated in tissue engineering applications thanks to their essential role in guiding muscle regeneration. In this work, the myogenic potential of dECM substrate, obtained from decellularized bovine pericardium (Tissuegraft Srl), was evaluated in vitro using C2C12 murine muscle cells. To assess myotubes formation, the width, length, and fusion indexes were measured during the differentiation time course. Additionally, the ability of dECM to support myogenesis was assessed by measuring the expression of specific myogenic markers: α-smooth muscle actin (α-sma), myogenin, and myosin heavy chain (MHC). The results obtained suggest that the dECM niche was able to support and enhance the myogenic potential of C2C12 cells in comparison of those grown on a plastic standard surface. Thus, the use of extracellular matrix proteins, as biomaterial supports, could represent a promising therapeutic strategy for skeletal muscle tissue engineering.  相似文献

Aβ(1-42) peptide is a neurotoxic agent strongly associated with the etiology of Alzheimer’s disease (AD). Current treatments are still of very low effectiveness, and deaths from AD are increasing worldwide. Huprine-derived molecules have a high affinity towards the enzyme acetylcholinesterase (AChE), act as potent Aβ(1-42) peptide aggregation inhibitors, and improve the behavior of experimental animals. AVCRI104P4 is a multitarget donepezil-huprine hybrid that improves short-term memory in a mouse model of AD and exerts protective effects in transgenic Caenorhabditis elegans that express Aβ(1-42) peptide. At present, there is no information about the effects of this compound on human erythrocytes. Thus, we considered it important to study its effects on the cell membrane and erythrocyte models, and to examine its protective effect against the toxic insult induced by Aβ(1-42) peptide in this cell and models. This research was developed using X-ray diffraction and differential scanning calorimetry (DSC) on molecular models of the human erythrocyte membrane constituted by lipid bilayers built of dimyristoylphosphatidylcholine (DMPC) and dimyristoylphosphatidylethanolamine (DMPE). They correspond to phospholipids representative of those present in the external and internal monolayers, respectively, of most plasma and neuronal membranes. The effect of AVCRI104P4 on human erythrocyte morphology was studied by scanning electron microscopy (SEM). The experimental results showed a protective effect of AVCRI104P4 against the toxicity induced by Aβ(1-42) peptide in human erythrocytes and molecular models.  相似文献

Improving the phytochemical composition of broccoli sprouts by elicitation   总被引:1,自引:0,他引:1  
In order to increase the concentrations of bioactive compounds in broccoli sprouts, exogenous elicitors: methionine, tryptophan, chitosan, salicylic acid (SA) and methyl jasmonate (MeJA) were sprayed at different concentrations during germination. The effects of the elicitors on vitamin C, phenolic compounds and glucosinolate levels were assessed in 3, 5 and 7-days old seedlings. The application of 200, 300 μM SA and 0.01% chitosan induced increases, by 26%, 18% and 54%, respectively in the content of vitamin C in 5 days old broccoli sprouts. Flavonoid concentration was also increased by 31% and 33% after 10 μM MeJA and 100 μM SA treatments, respectively in 7 days old broccoli sprouts. In contrast, methionine or tryptophan solutions did not positively affect the vitamin C or the phenolic compounds in the sprouts. The individual classes of glucosinolates respond differently to the elicitor treatment. Methionine, a sulphur-containing amino acid, applied at 5 mM, significantly increased, by 32%, the levels of aliphatic glucosinolates. However, indole glucosinolates presented a significant response to the induction with tryptophan, SA or MeJA treatments. Tryptophan at 10 mM concentration augmented by 1.8-fold the indole glucosinolate concentration in 7 days old broccoli sprouts, and the treatments of 100 μM SA and 25 μM MeJA also induced significantly higher amounts of indole glucosinolates (by 33% and 51%, respectively) in 7 days old treated broccoli sprouts. Therefore, the application of elicitors could be a useful tool for improving the bioactive metabolites in broccoli sprouts for fresh consumption or functional ingredients for nutraceuticals foods.  相似文献

A novel sensor array using seven room-temperature ionic liquids (ILs) as sensing materials and a quartz crystal microbalance (QCM) as a transducer was developed for the detection of organic vapors at ambient and elevated temperatures. Ethanol, dichloromethane, benzene, and heptane were selected as representative gas analytes for various kinds of environmental pollutants and common industrial solvents. The QCM/IL sensors responded proportionately and reversibly to the organic vapor concentrations (i.e., ethanol, heptane, and benzene) in the gas phase from 0 to 100% saturation at room and elevated temperatures (e.g., 120 degrees C) but deviated from this linear relationship at high concentrations for dichloromethane, a highly volatile compound. Linear discriminant analysis was used to analyze the sensing patterns. Excellent classifications were obtained for both known and unknown concentrations of vapor samples. The correct classifications were 100% for known concentration samples and 96% for samples with unknown concentrations. Thermodynamics and ATR-FT-IR studies were conducted to understand specific molecular interactions, the strength of the interaction between ILs and organic vapors, and the degree of ordering that takes place upon dissolution of the vapors in ILs. The different response intensity of the QCM/IL sensors to the organic vapors depends on the different solubilities of organic vapors in ILs and varying molecular/ion interactions between each organic vapor and IL. The diverse set of IL studied showed selective responses due to structural differences. Therefore, a sensor array of ILs would be able to effectively differentiate different vapors in pattern recognitions, facilitating discrimination by their distinctive patterns in response to organic vapors in both room and high temperatures.  相似文献
