The aim of this work was to investigate in which phases of ripening of Parmigiano Reggiano cheese lactic acid bacteria aminopeptidases present in cheese extract could be involved in release of free amino acids and to better understand the behavior of these enzymes in physical-chemical conditions that are far from their optimum. In particular, we evaluated 6 different substrates to reproduce broad-specificity aminopeptidase N, broad-specificity aminopeptidase C, glutamyl aminopeptidase A, peptidase with high specificity for leucine and alanine, proline iminopeptidase, and X-prolyl dipeptidyl aminopeptidase activities releasing different N-terminal amino acids. The effects of pH, NaCl concentration, and temperature on the enzyme activities of amino acid β-naphthylamide (βNA)-substrates were determined by modulating the variables in 19 different runs of an experimental design, which allowed the building of mathematical models able to assess the effect on aminopeptidases activities over a range of values, obtained with bibliographic data, covering different environmental conditions in different zones of the cheese wheel at different aging times. The aminopeptidases tested in this work were present in cell-free Parmigiano Reggiano cheese extract after a 17-mo ripening and were active when tested in model system. The modeling approach shows that to highlight the individual and interactive effects of chemical-physical variables on enzyme activities, it is helpful to determine the true potential of an amino-peptidase in cheese. Our results evidenced that the 6 different lactic acid bacteria peptidases participate in cheese proteolysis and are induced or inhibited by the cheese production parameters that, in turn, depend on the cheese dimension. Generally, temperature and pH exerted the more relevant effects on the enzymatic activities, and in many cases, a relevant interactive effect of these variables was observed. Increasing salt concentration slowed down broad-specificity amino-peptidase C, glutamyl aminopeptidase A, proline iminopeptidase, and peptidase with high specificity for leucine and alanine. Interestingly, this variable did not affect broad-specificity aminopeptidase N and positively affected X-prolyl dipeptidyl aminopeptidase. The models elaborated varying pH, temperatures, and salt concentration and were a useful, low cost, and fast tool to understand the role of the main peptidases in the different phases of cheese ripening in relation to the major environmental factors influencing enzyme activity. 相似文献

BACKGROUND: Variations in the contents of phytochemicals with biological activity in broccoli could originate as a result of genetic and environmental factors. An understanding of the effects of growth conditions on the bioactive compounds in broccoli is essential for improving its quality and nutritive value. Using salinity (40 mmol L?1 NaCl), and foliar sprayed compounds (methionine, tryptophan and chitosan) as different stress conditions, broccoli developed in soilless culture in the greenhouse was analysed for biologically active phytochemicals (glucosinolates, caffeoyl‐quinic, ferulic and sinapic derivatives and vitamin C). RESULTS: The application of elicitors during head formation could be beneficial for the enrichment in phytochemicals in broccoli. Management practices for increasing a given phytochemical (e.g., glucoraphanin or glucobrassicin) may be related to a decreased level of natural antioxidants (hydroxycinnamic acids). Growing broccoli hydroponically in the greenhouse in winter (Mediterranean climate) needs the supporting treatment of abiotic stress during development (i.e., NaCl, elicitors). CONCLUSION: The use of hydroponic growth conditions for broccoli and the application of stress factors (elicitors) at head induction and during development may serve the purpose of enhancing its nutritional quality to deliver a health‐promoting food. Storage hydropower plants, which are an important component of energy production in Switzerland, can lead to hydro- and thermopeaking, affecting river habitats and organisms. In this study, we developed an approach for integrating water temperature simulations into a habitat model to assess the impact of both hydro- and thermopeaking on the availability of suitable fish habitats. We focused on the habitat requirements of juvenile brown trout (Salmo trutta) in a semi-natural braided floodplain along the Moesa River (Southern Switzerland) in early summer. First, we defined different scenarios (with and without hydropeaking) based on the local hydrological and meteorological conditions. Second, we used a two-dimensional depth-averaged hydro- and thermodynamic model to simulate the spatial distributions of water depth, flow velocity, and water temperature. Third, we applied generalized preference curves for juvenile brown trout to identify hydraulically suitable habitats, and developed a new index to assess the availability of thermally suitable habitats. Finally, we quantified the extent to which hydraulically and thermally suitable habitats overlap in space and time. During both base and peak flow phases, most of the hydraulically and thermally suitable habitats are located in the side channels. High flow conditions combined with strong cold-thermopeaking lead to a higher thermal heterogeneity. However, disconnected habitats originate in the dewatering zone, increasing the risk of stranding as well as thermal stress. By helping to better understand the effects of thermopeaking on the availability of fish habitats, our approach could contribute to the design and evaluation of ecological restoration in hydropeaking rivers. 相似文献

ABSTRACTA convergence enhancement technique known as the integral balance approach is employed in combination with the Generalized Integral Transform Technique (GITT) for solving diffusion or convection-diffusion problems in physical domains with subregions of markedly different materials properties and/or spatial scales. GITT is employed in the solution of the differential eigenvalue problem with space variable coefficients, by adopting simpler auxiliary eigenproblems for the eigenfunction representation. The examples provided deal with heat conduction in heterogeneous media and forced convection in a microchannel embedded in a substrate. The convergence characteristics of the proposed novel solution are critically compared against the conventional approach through integral transforms without the integral balance enhancement, with the aid of fully converged results from the available exact solutions. 相似文献

The environmental performance of hydrogen production via indirect gasification of poplar biomass was evaluated following a Life Cycle Assessment approach. Foreground data for the study were provided mainly from process simulation. The main subsystems and processes that contribute to the environmental impacts were identified. Thus, poplar production and direct emissions to air from the processing plant were found to be the main sources of environmental impact. Abstract: Broccoli, (Brassica oleracea var. Italica), is generally recognized as a nutritive food rich in natural antioxidants, including vitamins and phenolics compounds. Phytochemical composition of 3 different purple sprouting broccoli varieties according with their production cycle: EEP (Extra Early), EP (Early), and LP (Late) were determined. The 3 cultivars tested were grown under the same conditions in an experimental field (SE Spain) where they were not previously cultivated since these cultivars are typical of cold, Northern‐European areas. EP variety showed greater growth according to the photosynthetic rate and C assimilation. However, it was not possible to obtain inflorescences for variety LP. The health‐promoting compounds (vitamin C, phenolic compounds, and glucosinolates) were higher in purple broccolis than in traditionally grown green broccolis and showed differences according to the variety. With respect to the use of broccoli byproducts as source of antioxidants, any variety would be suitable although the leaves of EP variety seems the richest in total phenolic acids, whereas the amount of anthocyanins was higher in LP variety, followed by EP and EEP varieties. Under our experimental conditions, the levels of glucosinolates were higher in purple broccoli than in green cultivars and also higher than other purple broccolis grown under different climate conditions. Therefore, the results reported data for production of the EEP and EP varieties in the Mediterranean climate with potential to obtain vegetables with improved nutritional quality. Practical Application: The results of this study reported data of the health‐promoting nutrients and natural antioxidants of EEP, EP, and LP purple sprouting broccoli varieties grown under the Mediterranean climate. The vitamin C, phenolic compounds, and glucosinolates in these purple varieties were higher than in traditionally grown green broccolis and other purple broccolis grown under different climate conditions. 相似文献

Hydropeaking dam operation and water extractions for irrigation have been broadly stated as alterations to natural flow regimes, which have also been noticed in the Biobío Watershed, in Central Chile, since 1996. In the Biobío River, most of native fish species are endemic and very little is known about them. Their ecological and social values have never been estimated, and there is lack of information about their habitat preferences. Furthermore, changes on fish habitat availability due to natural and/or man‐made causes have not been evaluated. In this study, eight native fish species, in a representative reach of the Biobío River, were studied and their preferred habitats were surveyed and characterized. A hydrodynamic model was built and linked to the fish habitat simulation model CASiMiR. Fuzzy rules and fuzzy sets were developed for describing habitat preference of the native fish species. CASiMiR was then used to simulate how physical habitat conditions vary due to flow control (i.e. upstream dams). Results show how overall habitat quality, expressed as weighted usable area (WUA) and hydraulic habitat suitability (HHS), changes and fluctuates due to the dam operation and how the daily hydropeaking is influencing quantity, quality and location of different habitats. The study suggests that the analysed fish are highly susceptible to flow control, as dams are currently operated, and fish habitat improvement suggestions are proposed. In literature, it is well know that the Smoothed Particle Hydrodynamics method can be affected by numerical noise on the pressure field when dealing with liquids. This can be highly dangerous when an SPH code is dynamically coupled with a structural solver. In this work a simple procedure is proposed to improve the computation of the pressure distribution in the dynamics of liquids. Such a procedure is based on the use of a density diffusion term in the equation for the mass conservation. This diffusion is a pure numerical effect, similar to the well known artificial viscosity originally proposed in SPH method to smooth out the shock discontinuities. As the artificial viscosity, the density diffusion used here goes to zero increasing the number of particles recovering consistency and convergence of the final numerical scheme adopted. Different artificial density diffusion formulas have been studied, paying attention to prevent unphysical changes of the flows. To show the improvements of the new scheme proposed here, a suitable set of examples, for which reference solutions or experimental data are available, has been tested. 相似文献
