Ultraviolent crosslinking is a key experimental step in the numerous protocols that have been developed for capturing and dissecting RNA–protein interactions in living cells. UV crosslinking covalently stalls dynamic interactions between RNAs and the directly contacting RNA-binding proteins and enables stringent denaturing downstream purification conditions needed for the enrichment and biochemical analysis of RNA–protein complexes. Despite its popularity, conventional 254 nm UV crosslinking possesses a set of intrinsic drawbacks, with the low photochemical efficiency being the central caveat. Here we show that genetically encoded photoreactive unnatural amino acids bearing a dialkyl diazirine photoreactive group can address this problem. Using the human iron regulatory protein 1 (IRP1) as a model RNA-binding protein, we show that the photoreactive amino acids can be introduced into the protein without diminishing its RNA-binding properties. A sevenfold increase in the crosslinking efficiency compared to conventional 254 nm UV crosslinking was achieved using the diazirine-based unnatural amino acid DiAzKs. This finding opens an avenue for new applications of the unnatural amino acids in studying RNA–protein interactions.  相似文献

Using metallographic, diffraction and thermal analysis examinations the base centered orthorhombic Al4(Cr,Fe) phase reported by Sui et al. (Philos Mag Lett 1995;71:139) was found to be a ternary extension of the Al–Cr η-phase. Mysis diluviana is one of the most abundant zooplankton by biomass in the Laurentian Great Lakes of North America, a predator of other zooplankton and an important prey for fishes. Studies of long-term trends in Lake Michigan have shown 2005–2016 densities to be 50–80% lower than 1990s densities, but these observations have been based on annual monitoring that is either spatially or seasonally limited. We combined Lake Michigan Mysis data from three annual programs and the 2015 Cooperative Science and Monitoring Initiative to achieve broad spatial coverage during spring, summer, and fall of 2015 and broad depth coverage during spring 2016. Lake-wide, annual density and biomass were 82 (SE: 10) Mysis/m2 and 200 (SE: 36) mg dry mass/m2. Density and biomass estimates were highest offshore, generally higher in the north basin, and seasonally highest in summer. Annual lake-wide averages for depths >30 m were better captured by seasonally-extensive annual programs than spatially-extensive annual programs, although spring sampling may bias annual values low. Mysis cohorts grew 0.026 mm/d (age-0) and 0.007 to 0.027 mm/d (age-1). Annual mortality was 81–98%. Reproduction was fall-spring and seasonal lake-wide estimates ranged from 0.6 to 19.1% females brooding, 13–20 embryos/brood, and 3–46 embryos/m2. Annual production (423 mg dry mass/m2/yr, SE: 31) was lower than all but one previous estimate from lakes Michigan, Huron, and Ontario. While Mysis tend to persist, low Mysis production may be a concern for prey fishes that feed on Mysis.  相似文献

Ice-templating is a well-established processing route for porous ceramics. Because of the structure/properties relationships, it is essential to better understand and control the solidification microstructures. Ice-templating is based on the segregation and concentration of particles by growing ice crystals. What we understand so far of the process is based on either observations by optical or X-ray imaging techniques, or on the characterization of ice-templated materials. However, in situ observations at particle-scale are still missing. Here we show that confocal microscopy can provide multiphase imaging of ice growth and the segregation and organization of particles. We illustrate the benefits of our approach with the observation of particles and pore ice in the frozen structure, the dynamic evolution of the freeze front morphology, and the impact of PVA addition on the solidification microstructures. These results prove in particular the importance of controlling both the temperature gradient and the growth rate during ice-templating.  相似文献

Two near-β Ti-10V-3Fe-3Al and Ti-10V-2Fe-3Al alloys were produced by blended elemental powder metallurgy using hydrogenated titanium and V-Fe-Al master alloy powders. The distributions of the alloying elements were investigated at different stages of transformation of the heterogeneous powder compacts into the final homogeneous alloy product. The influence of iron content on chemical homogenization, densification, microstructure, and mechanical properties of as-sintered alloys was discussed with respect to the fast diffusion mobility of iron in titanium. It was concluded that a 1 pct increase in Fe content, as the alloying element with the fastest diffusivity in titanium, has a positive effect on densification. However, this also results in some grain coarsening of the final material. The attained mechanical properties were comparable with those of cast/wrought near-beta titanium alloys.  相似文献

The use of air‐hardening steels in the manufacture of automobile body components shortens the corresponding process chain by means of eliminating the immersion or gas quenching operation since hardening occurs in still air. To choose the optimal forming and heat‐treatment parameters, the development of the microstructure needs to be considered as a consequence of the thermomechanical production processes. The object of this work is to investigate the effects of forming and the subsequent heat‐treating on the changes of the microstructural parameters for the air‐hardening steel LH800. For this purpose, specimens were processed under different forming and heat‐treating conditions and subsequently examined by means of SEM and mechanically tested using tensile tests. Based on this, an empirical equation was derived which describes the influence of both the martensite's bundle thickness and the original austenite's grain size on the yield stress. In this way, microstructural parameters can be identified which lead to good mechanical properties. The measured correlation can thus be used for modeling the LH800 steel's forming and heat‐treatment processes.  相似文献

Color bind: We have developed a probe TMR-para-MG that switches its fluorescence emission upon binding to a fluorogen-activating protein (FAP). We report on the experimental analysis of the charge transport through single-molecule junctions of the open and closed isomers of photoswitching molecules. Sulfur-free diarylethene molecules are developed and studied via electrical and optical measurements as well as density functional theory calculations. The single-molecule conductance and the current-voltage characteristics are measured in a mechanically controlled break-junction system at low temperatures. Comparing the results with the single-level transport model, we find an unexpected behavior of the current-dominating molecular orbital upon isomerization. We show that both the side chains and end groups of the molecules are crucial to understand the charge transport mechanism of photoswitching molecular junctions.  相似文献
