HPLC analysis of Echium plantagineum seed oil shows a complex triacylglycerol (TAG) profile. TAG species were separated on an analytical scale by HPLC and their fatty acid (FA) composition is reported. GLC analyses showed that some TAG fractions reached a stearidonic acid (SDA, 18:4n‐3) percentage significantly higher than that in the original oil. TAG separation on a bigger scale was also essayed, by means of a gravimetric normal‐phase chromatographic column, using silver ion‐silica gel as stationary phase. Gradient elution with solvents of increasing polarity was applied, allowing the separation of valuable TAG species containing γ‐linolenic acid (GLA, 18:3n‐6), α‐linolenic acid (ALA, 18:3n‐3) and SDA as the main constituents (more than 85% of the total FA). An enzymatic hydrolysis reaction showed the distribution of FA in the isolated species of TAG. SDA was the major FA in the sn‐2 position (more than 50% of total FA), followed by ALA (19%) and GLA (18.5%). 相似文献
The conformations of two synthetic pentapeptides with antimicrobial activity and their 4-fluorophenylalanine (Pff)-containing analogues (ArXArXAr-NH(2); Ar=Phe, Pff; X=Lys, Arg) have been studied. NMR experiments carried out both in aqueous fluoroalcohol solutions and SDS micelles permitted their interactions with membrane-like environments to be explored. WaterLOGSY experiments and Mn(2+)-based paramagnetic probes were also applied to assess their orientations with respect to the SDS micelles. In addition, pulse-field gradient (PFG) diffusion NMR spectroscopy studies were conducted, under different experimental conditions (i.e., concentration, temperature) to characterize the possible changes in the peptides' aggregation states as a putative critical factor for their antimicrobial activity. Finally, molecular dynamics simulations on a variety of conformations showed the intrinsic flexibility of these peptides in both aqueous solutions and membrane-mimetic systems. 相似文献
The aim of the present study was to establish an aquatic biomonitoring network for the Galician Environmental Specimen Bank (BEAG) (NW Spain). For this, a sampling system was designed that comprised of 121 points distributed throughout Galician rivers, from which samples of water and of three species of bryophytes were collected. The results obtained allowed selection of 74 sampling points and 2 species (Fontinalis antipyretica Hedw. and F. squamosa Hedw.) as the most suitable for use in future BEAG sampling surveys. The two species selected showed a strong similarity in their capacity to accumulate the 17 elements determined (Al, As, Ca, Cd, Co, Cr, Cu, Fe, Hg, K, Mg, Mn, Na, Ni, Pb, Se and Zn), as well as in their ecological preferences. Furthermore, the levels of contamination of epicontinental waters were lower than those observed in previous surveys. 相似文献
The kinetics which can be used to describe thermal softening of potato tissue by water cooking and pressure steaming were established and the temperature dependence of rheological and textural properties, using six different instrumental objective methods as firmness indicators, was determined within the temperature range of 100–122 °C. In water cooking at 100 °C and in steaming at 100, 112 and 117 °C, the thermal softening was best expressed by one simple pseudo first-order kinetic mechanism, which was considered to be the response to the changes in the pectic material in the cell wall and interlamellar region. Tension energy was the best rheological property for establishing the kinetics of softening of potato by water cooking, whereas maximum shear force was the best method of representing the tissue softening by steaming, using first-order models. Steaming at 122 °C caused overcooking of the tissue, even for short times. At this high temperature, rheological parameters (compression, tension and creep compliance) and textural properties from Texture Profile Analysis (TPA) fitted second-order models. When second-order models were used, the Arrhenius equation was significant for chewiness, indicating that this textural property can be useful for detecting overcooking caused by prolonged pressure steaming in the experimental temperature range. 相似文献
The effect of natural content of tannins and other phenolic compounds in faba bean seeds hull on the infestation by some store pests was investigated. The hull, particularly the content of phenolic compounds, does not influence the infestation and reproduction of Tribolium destructor and Trogoderma granarium. More sensitive to phenols seem to be the species Sitophilus oryzae and Oryzaephilus surinamensis. These results do not markedly confirm the checking effect of phenols on the degree of the infestation of faba beans by stored pests. 相似文献
Probabilistic microbial modeling using logistic regression was used to predict the boundary between growth and no growth of Saccharomyces cerevisiae at selected incubation periods (50 and 350 h) in the presence of growth-controlling factors such as water activity (a(w); 0.97, 0.95, and 0.93), pH (6.0, 5.0, 4.0, and 3.0), and potassium sorbate (0, 50, 100, 200, 500, and 1,000 ppm). The proposed model predicts the probability of growth under a set of conditions and calculates critical values of a(w), pH, and potassium sorbate concentration needed to inhibit yeast growth for different probabilities. The reduction of pH increased the number of combinations of a(w) and potassium sorbate concentration with probabilities to inhibit yeast growth higher than 0.95. With a probability of growth of 0.05 and using the logistic models, the critical pH values were higher for 50 h of incubation than those required for 350 h. With lower a(w) values and increasing potassium sorbate concentration the critical pH values increased. Logistic regression is a useful tool to evaluate the effects of the combined factors on microbial growth. 相似文献
The susceptibility of Escherichia coli, Salmonella enteritidis, Saccharomyces cerevisiae, and Zygosaccharomyces bailii to binary and ternary mixtures of potassium sorbate (KS), vanillin (V), and citral (C) was evaluated according to the Berenbaum experimental design, in laboratory media. For some V/C combinations, KS inhibitory concentrations were determined in agarized melon and mango purées by the spiral gradient endpoint (SGE) method. In laboratory media, inhibitory antimicrobial combinations were generally additives. For the yeasts, some synergistic effects were observed. All Berenbaum mixtures which resulted inhibitory in laboratory media were confirmed in the fruit purées. When the SGE method was used, several inhibitory ternary mixtures were found. The lowest inhibitory KS concentrations, estimated for a given V/C combination, corresponded to the bacteria assayed in melon purée. Z. bailii was not inhibited at any condition. Some synergistic antimicrobial combinations (595 ppm V + 251 ppm C + 8 ppm KS in melon and 280 ppm V + 123 ppm C + 8 ppm KS in mango purées) could be useful to achieve a desired inhibitory effect in fruit purées while reducing their concentrations. 相似文献

Results are presented of the effect of pre-treatments before freezing followed by long and short-term frozen storage (12 months at –18 °C and 24 days suffering temperature fluctuations between –18 °C and –12 °C) on quality parameters of raspberry. Pre-treatments were carried out with calcium, low methoxyl pectin, a combined solution, and results compared with untreated control fruits. Kramer shear, back extrusion, compression, and multiple penetration tests were used to measure rheological behavior. One-hundred mM CaCl2 reduced the long and short-term frozen storage induced loss of firmness. For long-term storage at –18 °C, a softening of the tissue became evident between 3 and 12 months and at each date test the stored fruits were firmer than those without storage. For short-term storage with fluctuations, the loss of firmness was evident between 0 and 24 days, and at all the testing dates the stored fruits were softer than those without storage. Results evidenced a higher cell damage in the short-term frozen storage. Coefficients of softening per day suffering fluctuation were determined, the highest values being given by Kramer shear energy and back extrusion maximum force (>1%). Short-term frozen storage affected physical and physico-chemical characteristics, increasing the saturation (r) and the anthocyanins and decreasing the ascorbic acid of the raspberries. In both storage conditions, pre-treatments reduced the drip loss, which correlated best with the Kramer shear energy. Panelists detected mainly time effect on the sensory firmness. For long-term, sensory firmness and juiciness gave the highest correlations with back extrusion maximum force, while for the short-term, sensory firmness and drip loss gave the highest correlations with the Kramer shear energy. SEM revealed different degrees of mechanical damage to structure, which accounted for rheological behavior of the fruits. 相似文献
