In order to analyze the possible relationship between metabolic rate and oxidative stress, OF1 female mice were rendered hyper- or hypothyroid for 4-5 weeks by administration of 0.0012% L-thyroxine (T4) or 0.05% 6-n-propyl-2-thiouracil (PTU), respectively, in their drinking water. Treatment with T4 resulted in increased basal metabolic rate measured by oxygen consumption and liver cytochrome oxidase activity without altering the glutathione redox system. Endogenous lipid peroxidation, sensitivity to lipid peroxidation and fatty acid unsaturation were decreased in the hyperthyroid group. Hypothyroidism also decreased phosphatidylcholine and cardiolipin fatty acid unsaturation but not in total lipids, and thus lipid peroxidation was not altered. Cardiolipin, a mainly mitochondrial lipid, was the most profoundly altered fraction by both hyper- and hypothyroidism. It is suggested that the lipid changes observed in hyperthyroid animals can protect them against an increased oxidative attack to tissue lipids due to their increased mitochondrial activities.  相似文献

The effect of composition, storage conditions and temperature over the rheological properties of dough-like materials prepared within a mixing-rheometer was studied using different egg albumen protein/glycerol ratios: 50/50, 60/40 and 65/35. As protein/glycerol ratio increases from 50/50 to 65/35, the mechanical spectra display a dramatic increase (i.e. 15,000-fold in the storage modulus at 1 rad/s), as a greater degree of structuration is achieved. An evolution from a predominantly viscous behaviour into a predominantly elastic one is also observed (i.e. the loss tangent shifts from 1.9 to 0.3 at 1 rad/s).When the viscoelastic properties were studied along ageing, a relaxation related minimum was found at relatively short times, which could define the optimal condition for bioplastic processing (e.g. injection moulding). Freezing also plays an important role by inhibiting ageing and strongly retarding the evolution of G′ and G″ along ageing time after thawing. When samples are submitted to a thermal treatment up to 90 °C, a minimum is always found both in G′ and G″ that takes place at lower temperature as the protein/glycerol ratio increases (from 62 to 49 °C).  相似文献

Carbonaceous particulate matter (PM) was collected by both thermal precipitation and glass fiber filters from specific combustion sources and examined by transmission electron microscopy (TEM) and field emission scanning electron microscopy (FESEM). PM from various natural gas burner combustion regimes (NGPM), burning tires (TPM), a heavy duty diesel truck (DPM), wood burning (WPM), candle burning (CPM), along with commercial black carbon (BC) powder and multiwall carbon nanotube (MWCNT) aggregates material exhibited similarities in PM microstructure, and average primary spherule diameters. The spherule structures consisted of curved graphene fragments and intercalated polycyclic aromatic hydrocarbons (PAH). PAH concentrations were measured and compared for these PMs, including PAH diagnostic ratios. An analysis of PAH carcinogenicity either as a carcinogenicity index or a potency equivalency factor from literature data indicated that only the TPM PAH content would pose any significant long term, carcinogenicity threat.  相似文献

Lung cancer (LC) and chronic obstructive pulmonary disease (COPD) commonly coexist in smokers, and the presence of COPD increases the risk of developing LC. Cigarette smoke causes oxidative stress and an inflammatory response in lung cells, which in turn may be involved in COPD and lung cancer development. The aim of this study was to identify differential proteomic profiles related to oxidative stress response that were potentially involved in these two pathological entities. Protein content was assessed in the bronchoalveolar lavage (BAL) of 60 patients classified in four groups: COPD, COPD and LC, LC, and control (neither COPD nor LC). Proteins were separated into spots by two dimensional polyacrylamide gel electrophoresis (2D-PAGE) and examined by matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF/TOF). A total of 16 oxidative stress regulatory proteins were differentially expressed in BAL samples from LC and/or COPD patients as compared with the control group. A distinct proteomic reactive oxygen species (ROS) protein signature emerged that characterized lung cancer and COPD. In conclusion, our findings highlight the role of the oxidative stress response proteins in the pathogenic pathways of both diseases, and provide new candidate biomarkers and predictive tools for LC and COPD diagnosis.  相似文献

BACKGROUND: To ameliorate the detrimental effects of freezing and thawing, blends of κ‐carrageenan (κ‐C) and xanthan gum (XG) were added to frozen/thawed mashed potatoes (F/TM potatoes). Product was tested by textural and rheological measurements, other quality parameters and sensory analyses. The effects of variation in levels of κ‐C (1.5–4.5 g kg?1) and XG (0.5–2.5 g kg?1) concentrations were studied. RESULTS: Instrumental measurements and sensory analyses indicated that 4.5 g kg?1κ‐C and 2.5 g kg?1 XG provided better thickening. κ‐C provided the appropriate texture, while XG imparted creaminess to the product, which could be associated with an increase in the amount of XG–water interactions. Addition of κ‐C had a major impact on textural properties, gel strength and viscoelastic behaviour, whereas XG influenced steady properties, colour, water‐holding capacity (WHC) and overall acceptability (OA). CONCLUSION: The OA of F/TM potatoes was mostly improved by addition of cryoprotectant blends, which is ascribed to improvement of F/TM potatoes texture by retarding starch retrogradation, increasing WHC, and enhancing the principal characteristics determining consumer acceptance. Creaminess was the most crucial factor for OA of the products. Addition of both polysaccharides at a low concentration (each cryoprotectant at 1.5 g kg?1) is recommended. The present study characterises garden strawberry leaf extract and reports its effect on shelf life and quality characteristics of fish oil. Radical scavenging capacity of the extract in DPPH? and ABTS?+ assays was equivalent to 1207 and 1579 μmol g?1 of trolox equivalents, respectively, total phenolic content was 257 mg g?1 of gallic acid equivalents. Phenolic and volatile constituents were analysed by liquid chromatography/mass spectrometry (LC/MS) and gas chromatography/mass spectrometry (GC/MS). The extract was added to fish oil and its oxidation was followed during 42 days: peroxide, p‐anisidine values and hexanal concentration were lower in the samples with extract, particularly at the end of storage. The effect of extract on fatty acid composition was not significant, however the changes in the percentages of some individual acids were observed. Fish oil containing 5% of extract had lower levels of lipid oxidation in comparison to other samples. The results indicate that strawberry leaves could be a potential source of natural antioxidants.  相似文献

The objective of this study was to investigate instrumental–sensory relationships of some texture scales using argentine foods as references. Textural characteristics of these foods were instrumentally investigated by the texture profile analysis technique. Principal component analysis (PCA) was used to describe the main attributes of the food samples. High Pearson’s correlation coefficients were found between hardness and fracturability (r = 0.94; P < 0.0001), hardness and gumminess (r = 0.71; P < 0.0001) and springiness and cohesiveness (r = 0.85; P < 0.0001). PCA identified two significant principal components, which accounted for 81.2% of the variance in the instrumental data. Additionally, a trained panel described the texture characteristics of the food samples according to the standard reference scales. The correlation curves showed nonlinear relationships (R2 between 85.6% and 99.9%) which were used to predict sensory attributes of other food samples. Some texture attributes like hardness and fracturability were accurately predicted by mechanical properties, while others like cohesiveness and adhesiveness were less representative.  相似文献

External and internal quality parameters were measured in oranges (Citrus sinensis (L.) Osbeck cv. “Powell Summer Navel”) during on-tree ripening and at harvest using near-infrared reflectance (NIR) spectroscopy. The performance of two NIRS instruments was evaluated: a handheld microelectromechanical system spectrophotometer working in the 1,600- to 2,400-nm range, and a diode array visible–NIR spectrophotometer working in the 380- to 1,700-nm range. Spectra and analytical data were used to construct MPLS prediction models for quantifying weight, size (equatorial and axial diameters), color (L*, a*, b*, C*, h*, and color index), texture (firmness and maximum penetration force), yield (pericarp thickness, juice weight, and juice content), and chemical parameters (soluble solids content, pH, titratable acidity, and maturity index). Both instruments yielded promising results for on-tree and at-harvest quality measurements, but models constructed using the diode array instrument provided greater predictive capacity, particularly for fruit size (equatorial and axial diameters) and total soluble solids content. Subsequent evaluation of the LOCAL algorithm revealed that it increased the predictive capacity of models constructed for all the main parameters tested. These results confirm that noninvasive NIRS technology can be used to simultaneously evaluate external and internal quality parameters in intact oranges both during on-tree ripening and at harvest, thus making it easier for farmers to monitor the ripening process and also to optimize harvest timing in order to meet the demands of the citrus-fruit industry.  相似文献

Human motor performance, speed and variability are highly susceptible to emotional states. This paper reviews the impact of the emotions on the motor control performance, and studies the possibility of improving the perceived skill/challenge relation on a multimodal neural rehabilitation scenario, by means of a biocybernetic controller that modulates the assistance provided by a haptic controlled robot in reaction to undesirable physical and mental states. Results from psychophysiological, performance and self assessment data for closed loop experiments in contrast with their open loop counterparts, suggest that the proposed method had a positive impact on the overall challenge/skill relation leading to an enhanced physical human–robot interaction experience.  相似文献
