In this work, FT-NIR spectroscopy was employed to determine iodine value (IV) and fatty acids (FA) content of pig fat samples, through the combined use of signal preprocessing, multivariate calibration, and variable selection methods. In particular, the main focus was on the use of variable selection methods, both in order to improve the predictive performance of the calibration models, and to identify relevant wavelengths that could be subsequently used for the development of simple, fast, and cheap hand-held devices, able to measure IV and FA content directly on the fat without the need of any sample pretreatment. Firstly, for each property of interest, partial least squares (PLS) multivariate calibration models were calculated considering the whole spectral range and testing different signal preprocessing methods. Then, once chosen the optimal signal preprocessing method, a two-step variable selection procedure was applied. In the first step, the interval-PLS variable selection algorithm was used to calculate a set of calibration models, whose outcomes were considered altogether in the second step, in order to select the optimal calibration model. The variable selection procedure allowed to lower the number of spectral variables retained by the model, and often led to an increase of the performance in prediction of the external test set samples.  相似文献

Solar plants that use molten salts as heat transfer fluid need careful control to avoid the freezing of the salt in the pipes; if such a problem occurs, a diagnostic instrument to localize where is the frozen salt plug and to determine its length is useful. If the salt contains potassium (as is the case of the most common mixture used in solar plants, NaNO3/KNO3 60/40% by weight), the gamma decay of the natural unstable isotope 40K can be exploited to detect the frozen salt in a non-invasive way. Simulations and experimental results regarding the detectability of such plugs with different masses/lengths are presented.  相似文献

Prophages account for most of the genetic diversity among strains of a given bacterial species, and represent a latent source for the generation of virulent phages. In this work, a set of 30 commercial, collection and dairy-isolated Lactobacillus casei group strains were used. A species-specific PCR assay allowed a reclassification, mainly of strains previously considered Lactobacillus casei, into either Lactobacillus paracasei or Lactobacillus rhamnosus. All the strains were induced with mitomycin C, allowing direct recovering of phage DNA in 25 cases, which corroborates the widely occurrence of lysogeny on Lactobacillus genomes, including probiotic strains of Lactobacillus casei group. Ten out of 11 commercial strains studied contained prophages, evidencing the potential risks of their use at industrial scale. Strains were also induced by treatment with different concentrations of hydrogen peroxide but, however, this agent was not able to evidence a prophage release for any of the strains tested. According to a RAPD-PCR fingerprinting with M13, 1254 and G1 primers, most of the commercial strains presented a high degree of homology and, regarding BglII- and BamHI-restriction profiles of phage DNA, six of them harboured the same prophage. Surprisingly, both Lactobacillus paracasei ATCC 27092 and Lactobacillus paracasei ATCC 27139 shared a second prophage with both an INLAIN collection and a commercial Lactobacillus paracasei strains, whereas two collection strains shared a third one. On the other hand, mitomycin C-inducible prophages were detected only on about a half of the strains isolated from dairy products, which had (with only one exception) from moderate to high correlation coefficients according to RAPD-PCR fingerprinting. After induction, supernatants were filtered and tested against nine Lactobacillus strains of the set sensitive to previously assayed virulent phages, allowing isolation of two new virulent phages: ф iLp1308 and ф iLp84. Predicting the fate of microorganisms in the environment is increasingly warranted, especially for pathogenic strains. A major habitat of Escherichia coli, which encompasses commensal as well as pathogenic strains, is the gastrointestinal tract with conditions very different from the environment it encounters after shedding from the host or during cultivation in the laboratory. We developed two relevant growth conditions representative of intestinal (host-associated) and external (postshedding) environments to investigate the surficial properties and behaviors of a diverse subset of E. coli feedlot isolates. Surficial properties may determine an isolate's physical fate. A pronounced increase in cell hydrophobicity and concomitant biofilm mass formation was observed for isolates grown under external conditions. Isolates that exhibited the highest surface hydrophobicity also formed visible suspended aggregates under external conditions. Other than hydrophobicity, flagella-mediated motility was determinant in affecting E. coli biofilm formation under external conditions, with all four nonmotile E. coli isolates characterized as thin-biofilm formers. The majority (88%) of Ag43+ (outer membrane protein, antigen 43) isolates formed thick biofilms, whereas the majority (75%) of Ag43- isolates formed thin biofilms. The tested E. coli O157:H7 strain behaved differently from the environmental E. coli isolates: it displayed a low electrostatic charge, a small decrease in hydrophobicity upon shifts to external conditions, and very little biofilm formation. On the other hand, the commonly used laboratory strain E. coli K-12 displayed low hydrophobicity both intestinally and externally, but it formed significant biofilm mass under external conditions. Clearly, various E. coli strains manifest significant variability in surficial behavior. This variability is further modulated by growth conditions. The interacting strain-inherent and cultivation-dependent effects on surficial behavior may have broad consequences for the fate and ecology of pathogenic and commensal E. coli strains.  相似文献

Fast real-time PCR TaqMan assays were developed and validated for species identification in dairy products. Based on the amplification of 12S rRNA and cytB partial genes of mitochondrial DNA, the methods were demonstrated to be sensitive, fast, and species-specific for Bos taurus, Ovis aries, Bubalus bubalis, and Capra hircus. The limit of detection calculated was lower than 1%, and the efficiency was reported to be higher than 96% in every assay. An internal amplification control was used to detect possible false negatives. The method was validated by means of laboratory-prepared samples mixing different species. Moreover, 18 commercial dairy samples were analyzed by both real-time PCR and isoelectric focusing, the official European Union reference method. The 4 TaqMan assays were confirmed to be a useful tool for milk and dairy product authentication.  相似文献

The goal of this paper is to describe a method to solve a class of time optimal control problems which are equivalent to finding the sub-Riemannian minimizing geodesics on a manifold M. In particular, we assume that the manifold M is acted upon by a group G which is a symmetry group for the dynamics. The action of G on M is proper but not necessarily free. As a consequence, the orbit space M/G is not necessarily a manifold but it presents the more general structure of a stratified space. The main ingredients of the method are a reduction of the problem to the orbit space M/G and an analysis of the reachable sets on this space. We give general results relating the stratified structure of the orbit space, and its decomposition into orbit types, with the optimal synthesis. We consider in more detail the case of the so-called K?P problem where the manifold M is itself a Lie group and the group G is determined by a Cartan decomposition of M. In this case, the geodesics can be explicitly calculated and are analytic. As an illustration, we apply our method and results to the complete optimal synthesis on S O(3).  相似文献

The genetic diversity among 15 fig accessions (Ficus carica L), belonging to 9 renowned cultivars of the Calabrian fig collection, and 24 unidentified genotypes of figs also located in the Calabrian region of Southern Italy, was investigated by random amplified polymorphic DNA (RAPD) analysis. The genetic similarity values were calculated for the 39 samples, and a dendrogram was elaborated by cluster analysis according to the UPGMA algorithm. The generated DNA fragments grouped the samples into two main clusters of RAPD profiles. Most of the 24 unknown samples, coming from the Luzzi area, formed a unique cluster with high degree of genetic similarity. This indicates that it is possible to distinguish, at DNA level, the fig trees with an already well‐known potential to produce figs suitable for the drying process and possibly to specify cultivars with suitable features for industrial transformation. BACKGROUND: Rabbit meat has excellent nutritive properties. The purpose of this study was to characterize rabbit meat and offal; in particular, the lipid fraction was studied in order to evaluate total and positional fatty acid (FA) compositions of triacylglycerol (TAG) and phosphatidylcholine (PC) fractions. Eight samples of weaned and eight of fattened rabbits were considered. RESULTS: Fattened rabbit meat contained slightly higher protein percentage content (P < 0.05) in comparison to weaned (20.1% versus 18.0%). Calcium content was higher in meat than in offal, unlike sodium, iron, zinc, manganese and copper. The cholesterol content in offal was much higher than in meat. FA profiles of total lipid showed a high percentage of unsaturated fatty acids and an n‐6/n‐3 ratio of 10.3 for fattened rabbit meat. Stereospecific analysis of TAG and PC was carried out on an eight‐sample pool of each meat and offal from weaned and fattened rabbits. In all samples the sn‐2‐position was prevalently esterified with oleic and linoleic acids in TAG, with polyunsaturated fatty acids in PC. CONCLUSION: Lipids from rabbit meat presented higher content of monounsaturated FA and lower n‐6/n‐3 ratio in comparison to offal, which was characterized by higher cholesterol and mineral levels. An observational study was designed to assess the bioaccumulation of polychlorodibenzodioxins (PCDD) and polychlorodibenzofurans (PCDF), dioxin-like polychlorobiphenyls (DL-PCB), and 13 selected polybromodiphenylethers (PBDE) in autochthonous pigs reared in the Nebrodi Park of Sicily (Italy). Perirenal fat and liver samples were drawn from animals representative of three different outdoor farming systems and from wild pigs and then analyzed for the chemicals mentioned previously. The highest concentrations of PCDD + PCDF and DL-PCB were detected in the fat (0.45 and 0.35 pg World Health Organization toxicity equivalents [WHO-TE] per g of fat base [FB], respectively) and livers (12.7 and 3.28 pg WHO-TE per g FB) of the wild group, whereas the free-ranging group showed the lowest levels (0.05 and 0.03 pg WHO-TE per g FB in fat and 0.78 and 0.27 pg WHO-TE per g FB in livers). The sum of PBDE congeners was highest in wild pigs (0.52 ng/g FB in fat and 5.64 ng/g FB in livers) and lowest in the farmed group (0.14 ng/g FB in fat and 0.28 ng/g FB in livers). The contamination levels in fat and livers of outdoor pigs had mean concentration values lower than those levels reported for intensively indoor-farmed animals. In wild pigs, bioaccumulation was associated with their free grazing in areas characterized by bush fires. The results of this study aid to emphasize the quality of the environment as a factor to guarantee food safety in typical processed pig meat products, specifically from outdoor and extensive Nebrodi farming systems.  相似文献
