The molecular composition of a core conduction element formed by the alpha-subunit of cloned epithelial Na+ channels (ENaC) was studied in planar lipid bilayers. Two pairs of in vitro translated proteins were employed in combinatorial experiments: 1) wild-type (WT) and an N-terminally truncated alphaDeltaN-rENaC that displays accelerated kinetics (tauo = 32 +/- 13 ms, tauc = 42 +/- 11 ms), as compared with the WT channel (tauc1 = 18 +/- 8 ms, tauc2 = 252 +/- 31 ms, and tauo = 157 +/- 43 ms); and 2) WT and an amiloride binding mutant, alphaDelta278-283-rENaC. The channels that formed in a alphaWT:alphaDeltaN mixture fell into two groups: one with tauo and tauc that corresponded to those exhibited by the alphaDeltaN-rENaC alone, and another with a double-exponentially distributed closed time and a single-exponentially distributed open time that corresponded to the alphaWT-rENaC alone. Five channel subtypes with distinct sensitivities to amiloride were found in a 1alphaWT:1alphaDelta278-283 protein mixture. Statistical analyses of the distributions of channel phenotypes observed for either set of the WT:mutant combinations suggest a tetrameric organization of alpha-subunits as a minimal model for the core conduction element in ENaCs.  相似文献

Advantages of chemically prepared powders for electronic ceramics have been demonstrated for a number of multilayer capacitor (MLC) dielectrics. A cost-efficient precipitation process was developed to produce undoped or doped crystalline barium titanate powder with a narrow particle size distribution close to 0.5 μm. More complex compositions, e.g., barium-neodymium titanate, were amorphous as precipitated but could be crystallized by calcination below 1000°C. Additional compositional modifications, to adjust electrical properties or to lower sintering temperature, were accomplished by doping the surface of the powder particles using a solution coating process. Exceptional fired densities and electrical performance were obtained.  相似文献

Pressure to compress the development life cycle and reduce the duration and resources committed to testing lead to experimentation in testing at the NASA Goddard Space Flight Centerís Software Engineering Laboratory. This study investigates the trend to reduce developer testing and increasingly rely upon inspection techniques and independent functional testing to shorten the development life cycle, improve testing productivity, and improve software quality.An approach is developed to conduct this comparison. In particular, the problem faced by software researchers, having a comprehensive characterization of software projects so similar types may be identified for comparative studies, is addressed using expert opinion.  相似文献

The closure of ungrafted sacrococcygeal pilonidal sinus excisional wounds was studied in 15 patients. Wound punch biopsies were taken on a regular basis, and histologic sections were made. To document changes, computer-assisted morphometric image analysis was employed. Initial average wound depth was 37.8 +/- 4.6 mm, and complete closure (0 wound depth) was reached by 68 days. Wound contraction contributed 88 percent to wound closure, whereas the deposition of scar only contributed 12 percent. Maximum cells density within granulation tissue was reached by day 18. Myofibroblasts, identified by alpha-smooth muscle actin immunostaining, first appeared on day 11. Unlike those observed in laboratory animals, myofibroblasts were a minor cell population of granulation tissue, never exceeding 10 percent of the cells. The pattern of collagen fiber organization was documented by polarized light microscopy of Sirius red-stained sections. Early granulation tissue collagen fibers demonstrated a fine greenish birefringence, whereas more mature granulation tissue collagen fibers were thicker, displaying orange-yellowish birefringence. Myofibroblasts were associated exclusively with thicker collagen fibers, whereas fibroblasts were associated with both fine and thick collagen fibers. It is proposed that human wound contraction involves a volume change whereby normal dermal and adipose tissues are pulled into the defect by forces generated within fibroblasts.  相似文献

Previous research and this research indicate that the mechanical melting for poly(vinylidene chloride) copolymers (PVDC) is complex. Mechanical melting is defined as the melting (or devitrification) of a polymer when a significant portion of the thermal energy originates from a mechanical energy dissipative process. PVDC mechanically melted on a moving metal surface at temperatures of the test instrument that were considerably lower than the differential scanning calorimetry (DSC) onset melting temperature. PVDC formulated with low levels of high density polyethylene (HDPE), however, melted at metal temperatures near the DSC onset melting temperature. Two different mechanical melting mechanisms are proposed to explan the data, and the frictional data are discussed with respect to solids conveying in a single-screw, plasticating extruder.  相似文献

The experiments reported here were designed to test the suggestion of many researchers that selective attention to visual features of a prey can account for search-image effects. In 3 experiments pigeons ate wheat and vetch grains presented on multicolored and gray gravel trays. In Experiment 1 search-image effects were evident when grains were cryptic but not when they were conspicuous. Experiment 2 demonstrated that search images can be activated when the grains encountered are either cryptic or conspicuous but that search images affect search performance only when the grains are cryptic. Experiment 3 demonstrated that search images are short-term in nature: A 3-min delay between successive encounters with a type of grain disrupted an activated search image. The discussion addresses how these results further develop a model in which search images are viewed as selective attention to visual features of a prey. The effects of conditioning on the endomysial fraction of bovine meat were investigated. Solubility studies on endomysial connective tissue and analysis of insoluble endomysial fractions remaining after conditioning were carried out. Yields of soluble endomysial fractions represented, on average, 94·5% of total extracted endomysial material for unconditioned muscles compared with 97·5% for conditioned muscles. Soluble endomysial fractions contained, on average, 0·13% collagen from unconditioned muscles and 0·22% collagen from conditioned muscles. The main peptide components observed on analysis of insoluble endomysial fractions after CN Br digestion were derived from types I and III collagen. Changes observed on the peptide maps, evident as the appearance of a number of new bands from conditioned samples, appeared to be muscle specific. The amount of type III collagen relative to type I decreased on conditioning, indicating that endomysial type III collagen was preferentially destroyed during conditioning. Two-dimensional polyacrylamide gel electrophoresis maps revealed new peptide material in the molecular weight range 40 000 to 50 000 on conditioning. These data provide direct evidence of the effect of proteolytic processing on endomysial collagen during conditioning.  相似文献

Despite the possible benefits of implementing healthcare information technologies, successful implementation of effective healthcare information technology is constrained by cultural and regulatory concerns and technical obstacles encountered when establishing or upgrading an organisation's enterprise infrastructure. In this paper, we advance Ross' four‐stage model of enterprise architecture maturity as a valuable IT resource for helping healthcare organisations sustain a competitive advantage. We use partial least squares (PLS) structural equation modelling to analyse survey data from 164 US hospitals at different stages of EA maturity. Our results provide evidence that enterprise architecture maturity directly influences the effectiveness of hospitals' IT resources for achieving strategic goals. Further, enterprise architecture maturity indirectly influences the effectiveness of IT resources when IT alignment is incorporated as a mediating variable. We discuss the implications of our findings for research and practice and suggest opportunities for future research.  相似文献

Lithium zinc silicate glasses are of interest for the preparation of moderately high thermal expansion glass-ceramics which are suitable for sealing to a number of nickel-based superalloys. The effect of composition, in particular the variation of nucleating species, on the crystallization behaviour of a number of these glasses has been examined using differential thermal analysis, X-ray diffraction, and electron microscopy. Various crystal phases have been identified, including cristobalite, quartz, tridymite and 0 Li2ZnSiO4. In addition, most of the glass-ceramics also contain an unidentified phase which may be related to the-series of lithium zinc silicates. Heat-treatment schedules have been derived on the basis of these results in order to produce a number of glass-ceramic materials. The resultant thermal expansion characteristics of the glass-ceramics have been monitored using dilatometry, and expansions in the range 12.3 to 17.1×10–6° C–1 (20 to 460° C), have been obtained, depending on the precise glass composition and heat-treatment schedule employed. In addition, the mechanical properties of a number of selected samples have been monitored, employing a biaxial flexure technique.  相似文献

Polystyrene (PS)-polyether sulphone (PES) polymer blend thin films were prepared for examination in a scanning transmission electron microscope. The microstructures observed in 75 wt % PS-25 wt % PES films consisted of spherical inclusions, ranging from 0.2 to 1.2m in diameter. X-ray spectrometric analysis in the microscope revealed that the inclusions were PES-rich, while the matrix contained only PS. Attention in this paper is paid to the contrast in the annular dark-field detector (ADF) images from these thin films. This image contrast has a complicated dependence on both the angular range subtended by the dark-field detector and mass-thickness variations within the films. On microscopes with appropriate lens controls which permit the acceptance angle of the ADF detector to be varied, it becomes possible actually to reverse the contrast between the two phases.  相似文献
