Mercury is a severe environmental pollutant with neurotoxic effects, especially when exposed for long periods. Although there are several evidences regarding mercury toxicity, little is known about inorganic mercury (IHg) species and cerebellum, one of the main targets of mercury associated with the neurological symptomatology of mercurial poisoning. Besides that, the global proteomic profile assessment is a valuable tool to screen possible biomarkers and elucidate molecular targets of mercury neurotoxicity; however, the literature is still scarce. Thus, this study aimed to investigate the effects of long-term exposure to IHg in adult rats’ cerebellum and explore the modulation of the cerebellar proteome associated with biochemical and functional outcomes, providing evidence, in a translational perspective, of new mercury toxicity targets and possible biomarkers. Fifty-four adult rats were exposed to 0.375 mg/kg of HgCl2 or distilled water for 45 days using intragastric gavage. Then, the motor functions were evaluated by rotarod and inclined plane. The cerebellum was collected to quantify mercury levels, to assess the antioxidant activity against peroxyl radicals (ACAPs), the lipid peroxidation (LPO), the proteomic profile, the cell death nature by cytotoxicity and apoptosis, and the Purkinje cells density. The IHg exposure increased mercury levels in the cerebellum, reducing ACAP and increasing LPO. The proteomic approach revealed a total 419 proteins with different statuses of regulation, associated with different biological processes, such as synaptic signaling, energy metabolism and nervous system development, e.g., all these molecular changes are associated with increased cytotoxicity and apoptosis, with a neurodegenerative pattern on Purkinje cells layer and poor motor coordination and balance. In conclusion, all these findings feature a neurodegenerative process triggered by IHg in the cerebellum that culminated into motor functions deficits, which are associated with several molecular features and may be related to the clinical outcomes of people exposed to the toxicant.  相似文献

α-Amylases are known to be multidomain proteins, i.e., the molecules consist of several folding units. Each α-amylase is believed, however, to have a catalytic domain consisting, of a barrel of eight parallel α-strands surrounded by eight α-strands. with an extra helix inserted after the sixth γ-strands. The α-strands and helices alternate along the polypeptide chain and are linked together by irregular loops. Amino acid residues situated on the loops joining the C-terminal end of each α-strand to the N-terminal end of the following helix make up the active site of the enzymes. A similar structure has been found in cyclodextrin glucanotransfcrases and it is now believed that such a (α/α)8-barrel also constitutes the catalytic domain of enzymes active on α-1.6-glucosidic bonds, and of enzymes with dual specificity for both α-1.4- and α-1.6- bonds. Knowledge of the three-dimensional structure of α-amylases and cyclodextrin glucanotransferase has made possible identification of structural features important for enzymic activity and specificity. By analogy, some general conclusions are reached concerning pullulanase, isoamylase. oligo-1,6-glucosidase, neopullulanase and branching enzymes.  相似文献

The reaction of nitrosubstituted aryl fluorides 1 with cyclic β-diketones 2 proceeds at 20–80°C in the presence of bases, such as KOH, KF or NaOEt, leading to the aryl ethers 3a – m . Depending on the base the reaction of dimedone 2a or 1,3-cyclohexanedione 2b with 2,4-dinitrofluorobenzene 1a or 4-fluoro-3-nitrobenzonitrile 1b furnishes C- and/or O-arylated products. Upon heating of 3g , e and 3m at 40–100°C in DMF/K2CO3, the C-arylated ketones 4a – c are formed in good yields. Starting from 3a we obtained the chromenedione 5 under the same conditions.  相似文献

Photobacteriosis is a septicaemic bacterial disease affecting several marine species around the globe, resulting in significant economic losses. Although many studies have been performed related to the pathogen virulence and resistance factors, information regarding the host defence mechanisms activated once an infection takes place is still scarce. The present study was designed to understand innate immune responses of farmed juvenile gilthead seabream (Sparus aurata) after Photobacterium damselae subsp. piscicida (Phdp) infection. Therefore, two groups of seabream juveniles were intraperitoneally injected with 100 µL of PBS (placebo) or 100 µL of exponentially growing Phdp (1 × 106 CFU/mL; infected). The blood, plasma, liver, and head kidney of six fish from each treatment were sampled immediately before infection and 3, 6, 9, 24 and 48 h after infection for the broad screening of fish immune and oxidative stress responses. Infected animals presented marked anaemia, neutrophilia and monocytosis, conditions that are correlated with an increased expression of genes related to inflammation and phagocytic activity. Similar studies with different fish species and bacteria can be useful for the definition of health biomarkers that might help fish farmers to prevent the occurrence of such diseases.  相似文献

Some say that all diseases begin in the gut. Interestingly, this concept is actually quite old, since it is attributed to the Ancient Greek physician Hippocrates, who proposed the hypothesis nearly 2500 years ago. The continuous breakthroughs in modern medicine have transformed our classic understanding of the gastrointestinal tract (GIT) and human health. Although the gut microbiota (GMB) has proven to be a core component of human health under standard metabolic conditions, there is now also a strong link connecting the composition and function of the GMB to the development of numerous diseases, especially the ones of musculoskeletal nature. The symbiotic microbes that reside in the gastrointestinal tract are very sensitive to biochemical stimuli and may respond in many different ways depending on the nature of these biological signals. Certain variables such as nutrition and physical modulation can either enhance or disrupt the equilibrium between the various species of gut microbes. In fact, fat-rich diets can cause dysbiosis, which decreases the number of protective bacteria and compromises the integrity of the epithelial barrier in the GIT. Overgrowth of pathogenic microbes then release higher quantities of toxic metabolites into the circulatory system, especially the pro-inflammatory cytokines detected in osteoarthritis (OA), thereby promoting inflammation and the initiation of many disease processes throughout the body. Although many studies link OA with GMB perturbations, further research is still needed.  相似文献

Friedel-Crafts acetylation of phenanthrene ( 1a ) in sym-tetrachloroethane yields mixtures of 2-, 3- and 9-acetylphenanthrenes ( 2a, 3a, 4 ). The distribution of isomers is found to depend strongly upon the method of mixing the reagents. Acetylation of 9-chlorophenanthrene ( 1b ), performed by a variety of methods and solvents, led mainly to 3-acetyl-9-chlorophenanthrene ( 3b ) (≥85%). Previously unreported 2-acetyl-9-chlorophenanthrene ( 2b ) was found to form up to a maximum 11% in nitrobenzene.  相似文献

Studies on the Synthesis of Polyethers from Anhydropolyols . By reaction of 1,4:3,6-dianhydro-D-sorbitol with base and αω-dihaloalkanes mono and dialkyl derivatives are obtained. The use of trans-1,4-dichloro-2-butene leads to oligomers up to the heptamer which are isolated and structurally assigned. Polymerisations with super acids of substituted tetrahydrofuranes such as the 1,4-anhydro-2,3-di-O-alkyl-D-erythritols lead to functionalized poly(oxytetramethylenes). By detailed studies of reaction parameters optimized conditions are achieved, and by polymer analyses the linear structures of these polyethers are demonstrated.  相似文献

Petroleum-derived solvents commonly used in membrane fabrication are often hazardous and toxic, so the investigation of safer alternatives is important. In this study, two low-hazard solvents, methyl 5-(dimethylamino)-2-methyl-5-oxopentanoate (Rhodiasolv® PolarClean) and gamma-valerolactone (GVL), were investigated as sole solvents and as cosolvents to cast polysulfone membranes via nonsolvent induced phase inversion. Normalized viscosity was introduced as an indicator of dope solution homogeneity and was used to compare the required time of mixing to achieve full dissolution of the polymer in the different solvents/solvent mixtures. All dope solutions made with low-hazard solvents were found to be more viscous than those made with traditional solvents, which meant additional mixing time was needed, and that fabricated membranes were morphologically different. With respect to operation, membranes cast from dope solutions containing equal amounts of PolarClean and GVL displayed the most similar flux curves and solute rejection to those made using the traditional solvent tested.  相似文献
