Fecal coliform removal in wastewater treatment plants studied by plate counts and enzymatic methods

Twelve wastewater treatment plants (WWTPs) were sampled in France and Belgium in 1999 and 2000 in order to estimate the fecal coliform (FC) removal efficiency of various types of treatment. Only one of these WWTPs was equipped with a specific step to eliminate microorganisms (UV disinfection preceded by sand filtration). FC abundance was measured in raw and treated sewage by plate counts on selective medium and rapid beta-D-glucuronidase (GLUase)-based assays. Removal of culturable FC was the most efficient in treatments with high retention time (activated sludge process with nitrification and denitrification, lagooning), in biofiltration and in the treatment with a tertiary disinfection step. GLUase activity measurements showed the same removal pattern as plate counts except for UV disinfection, where no reduction of GLUase activity was measured. Specific loads of culturable FC and GLUase activity, i.e. daily amounts of culturable FC or GLUase activity in sewage per inhabitant-equivalent, were calculated in raw and treated wastewater for the different WWTPs.     

Beyond Heparinization: Design of Highly Potent Thrombin Inhibitors Suitable for Surface Coupling

During extracorporeal circulation, when blood comes in contact with artificial surfaces, patients receive a standard treatment with anticoagulants to avoid blood coagulation. Dialysis patients in particular are systemically treated with heparin up to four times a week, causing a high burden for the body. For potential anticoagulant modification of external materials, such as dialysis equipment, a series of highly potent thrombin inhibitors was developed. All inhibitors share the general formula arylsulfonyl‐P3‐Pro‐4‐amidinobenzylamide, where P3 is glycyl or a trifunctional amino acid residue in L ‐configuration. Among this series, several derivatives inhibit thrombin with K_i values of less than 1 nM . Specificity measurements revealed that this inhibitor type is highly specific for thrombin with negligible activity against related trypsin‐like serine proteases. X‐ray analysis of the most potent analogue in complex with thrombin demonstrated that the N‐terminal arylsulfonyl group occupies the aryl binding site, whereas the P3 side chain is directed into the solvent and therefore is well suited for further coupling. Based on their in vitro profile, these inhibitors are suitable candidates for the development of hemocompatible materials with anticoagulant properties.

The environmental impact assessment (EIA) system in Uzbekistan was inherited from the USSR and is centred on state environmental review, a procedure for reviewing the impacts of proposed activities by environmental authorities. This is proving to be a less effective tool of environmental protection in the new political and economic conditions. Since the collapse of the USSR, the Uzbekistani EIA system has undergone gradual reform but the national EIA system remains almost identical to the ‘socialist’ system. At the same time, some elements are similar to those of EIA systems used in developed countries. The shortcomings of the system include unclear screening provisions and lack of public participation. The paper analyses strengths, weaknesses, opportunities and threats for the development of EIA in Uzbekistan, taking into account various political and economic factors.     

Heterologous Expression and Structural Characterisation of a Pyrazinone Natural Product Assembly Line

Through a number of strategies nonribosomal peptide assembly lines give rise to a metabolic diversity not possible by ribosomal synthesis. One distinction within nonribosomal assembly is that products are elaborated on an enzyme‐tethered substrate, and their release is enzyme catalysed. Reductive release by NAD(P)H‐dependent catalysts is one observed nonribosomal termination and release strategy. Here we probed the selectivity of a terminal reductase domain by using a full‐length heterologously expressed nonribosomal peptide synthetase for the dipeptide aureusimine and were able to generate 17 new analogues. Further, we generated an X‐ray structure of aureusimine terminal reductase to gain insight into the structural details associated with this enzymatic domain.     

Broken symmetries and thermodynamic limit in quantum antiferromagnets

We study how the Néel order breaks the symmetries of the hamiltonian in quantum antiferromagnet. The construction of a quantum wave packet that mimics the classical groundstate implies the existence of an extensive number of states: 1) with an energy scale much smaller than the spin-waves excitations (magnons); 2) with well defined symmetries.For the case of the triangular lattice, we find that all necessary properties to obtain a Néel state are well verified, already on periodic samples as small as 21 spins. For this system, this is a new and strong evidence of Néel order in the thermodynamic limit.
