An ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method was developed for the determination of fluoroquinolones–ciprofloxacin (CIPRO), danofloxacin (DANO), enrofloxacin (ENRO) and sarafloxacin (SARA)–in aquaculture products, specifically salmon, shrimp and tilapia. After initial sample extraction with an acidic acetonitrile solution, the extract was diluted with dichloromethane and centrifuged, then an aliquot was concentrated and applied to a C18 solid-phase extraction cartridge and concentrated for a second time. The resultant residue was dissolved in acetonitrile, diluted with water, and then further defatted with hexane. The fluoroquinolone residues were determined by UPLC with an HSS T3 C18 reverse-phase column using an ammonium hydroxide-formic acid buffer in an acetonitrile gradient with MS/MS detection using multiple reaction monitoring. Average recoveries for salmon tissue ranged from 73% for DANO to 95% for SARA, for shrimp from 71% for DANO to 109% for SARA, and from 62% for DANO to 111% for SARA in tilapia, fortified at the 1.0 ng g?1 level. Standard curves were linear between 0.002 and 0.5 ng injected for all compounds. Detection limits of 0.2 ng g?1 for CIPRO, DANO, ENRO, and SARA were easily obtainable. The operational errors, interferences, and recoveries for fortified samples demonstrate that this described method is suitable for routine use in a regulatory programme. The recommended method is simple, rapid, specific and reliable for the routine monitoring of fluoroquinolone residues in aquatic species such as salmon, tilapia and shrimp.  相似文献

Blends of bisphenol-A polycarbonate (PC) and polystyrene (PS) prepared by screw extrusion and solution casting have been investigated with weight fractions of PC in the blends varying from 0.95 to 0.05. From the measured glass transition temperatures (Tg) and specific heat increments (ΔCp) at the Tg, the polystyrene appears to dissolve more in the PC phase than does the PC in the PS phase. The blend appears to be near eqilibrium under extrusion conditions so that the polymer–polymer interaction parameter of PC/PS blends was calculated and found to be 0.038±0.004 for extruded blends at 250°C. Scanning electron microscopy supports the conclusion that the compatibility increases more in the region of PS-rich compositions than in the regions of PC-rich compositions of the PC/PS blends.  相似文献

Fractalkine (CX3CL1) acts as a chemokine as well as a regulator of iron metabolism. Fractalkine binds CX3CR1, the fractalkine receptor on the surface of monocytes/macrophages regulating different intracellular signalling pathways such as mitogen-activated protein kinase (MAPK), phospholipase C (PLC) and NFκB contributing to the production of pro-inflammatory cytokine synthesis, and the regulation of cell growth, differentiation, proliferation and metabolism. In this study, we focused on the modulatory effects of fractalkine on the immune response and on the iron metabolism of Escherichia coli and Pseudomonas aeruginosa lipopolysaccharides (LPS) and Staphylococcus aureus lipoteichoic acid (LTA) activated THP-1 cells to get a deeper insight into the role of soluble fractalkine in the regulation of the innate immune system. Pro-inflammatory cytokine secretions of the fractalkine-treated, LPS/LTA-treated, and co-treated THP-1 cells were determined using ELISArray and ELISA measurements. We analysed the protein expression levels of signalling molecules regulated by CX3CR1 as well as hepcidin, the major iron regulatory hormone, the iron transporters, the iron storage proteins and mitochondrial iron utilization. The results showed that fractalkine treatment alone did not affect the pro-inflammatory cytokine secretion, but it was proposed to act as a regulator of the iron metabolism of THP-1 cells. In the case of two different LPS and one type of LTA with fractalkine co-treatments, fractalkine was able to alter the levels of signalling proteins (NFκB, PSTAT3, Nrf2/Keap-1) regulating the expression of pro-inflammatory cytokines as well as hepcidin, and the iron storage and utilization of the THP-1 cells.  相似文献

We apply multivariate error-propagation analysis to color-signal transformations. Results are given that indicate how linear, matrix, and nonlinear transformations influence the mean, variance, and covariance of color-measurements and color-images. Since many signal processing paths include these steps, the analysis is applicable to color-measurement and imaging systems. Expressions are given that allow image noise or error propagation for a spectrophotometer, colorimeter, or digital camera. In a computed example, error statistics are propagated from tristimulus values to CIELAB coordinates. The resulting signal covariance is interpreted in terms of CIELAB error ellipsoids and the mean value of color-difference measures, and . The application of this analysis to system design is also illustrated by relating a tolerance to equivalent tristimulus-value error statistics. © 1997 John Wiley & Sons, Inc. A new nondestructive technique, confocal scanning laser microscopy (CSLM), is described that is used to characterize the topography and morphology of polymer coatings. The topography of the coating can be determined even when the coating is completely opaque. When the coating is not completely opaque, CSLM has the distinct advantage of also being able to distinguish between the coating surface and the substrate, thus enabling coating thickness to be determined over a wide range of areas. In this study CSLM was successfully applied to poly(2-vinylpyridine) coatings formed on mild steel substrates by in situ electropolymerization. Satisfactory morphological details were obtained for areas ranging from 200 × 200 μm to 4 × 4 mm. Quantitative measurements of the coating thickness and the surface roughness distribution were also carried out. Although several other nondestructive techniques for coating morphological analyses are available, CSLM has unique advantages in being able to provide simultaneous qualitative and quantitative information on coating surfaces as well as measurements over a wide range of surface areas. A comparison of CSLM with other popularly used methods is provided and the characteristics and limitations of the various techniques are discussed. © 1998 John Wiley & Sons, Inc. In this contribution, four new compounds synthesized from 4-hydroxycoumarin and tyramine/octopamine/norepinephrine/3-methoxytyramine are characterized spectroscopically (IR and NMR), chromatographically (UHPLC-DAD), and structurally at the B3LYP/6-311++G*(d,p) level of theory. The crystal structure of the 4-hydroxycoumarin-octopamine derivative was solved and used as a starting geometry for structural optimization. Along with the previously obtained 4-hydroxycoumarin-dopamine derivative, the intramolecular interactions governing the stability of these compounds were quantified by NBO and QTAIM analyses. Condensed Fukui functions and the HOMO-LUMO gap were calculated and correlated with the number and position of OH groups in the structures. In vitro cytotoxicity experiments were performed to elucidate the possible antitumor activity of the tested substances. For this purpose, four cell lines were selected, namely human colon cancer (HCT-116), human adenocarcinoma (HeLa), human breast cancer (MDA-MB-231), and healthy human lung fibroblast (MRC-5) lines. A significant selectivity towards colorectal carcinoma cells was observed. Molecular docking and molecular dynamics studies with carbonic anhydrase, a prognostic factor in several cancers, complemented the experimental results. The calculated MD binding energies coincided well with the experimental activity, and indicated 4-hydroxycoumarin-dopamine and 4-hydroxycoumarin-3-methoxytyramine as the most active compounds. The ecotoxicology assessment proved that the obtained compounds have a low impact on the daphnia, fish, and green algae population.  相似文献

Using a wedged and antireflection-coated diamond heatspreader, a continuously tunable semiconductor disk laser with intracavity second-harmonic generation (SHG) is demonstrated. Output powers of > 600 mW tunable over 10 nm around 530 nm are obtained. Finite-element modeling shows that the use of a diamond heatspreader for thermal management - in contrast to substrate thinning approaches - permits power scaling across the 670-2300-nm range of these lasers. Using a green laser as an exemplar, this paper details the issues involved in translating this spectral coverage to the ultraviolet and visible via SHG. Polarization and wavelength selection are discussed and the adopted approaches presented. Almost 1 W of second-harmonic light at 530 nm is demonstrated, with an efficiency of 11% with respect to the incident pump power.  相似文献
