Ultrafine Gold nanoparticles (Au NPs) functionalized with various biomolecules constitute an alternative to antibodies as anti-amyloidogenic agents. However, generating stable ultrafine Au NPs with high surface activity is challenging. Here, the capacity of phosphate groups in biomolecules is used to stabilize Au NPs. The characteristics of Au NPs decorated with adenosine mono-, di-, and tri-phosphate are compared as well as adenosine and peptide nucleic acid-containing adenosine as controls. Among them, ATP-Au NPs are found to be superior having small size (2–4 nm) and stability (for several months) when analysed by spectroscopy and electron microscopy. Spectroscopy analysis also revealed that each ATP-stabilized Au NP is decorated with 7–8 molecules of ATP. ThT binding analysis and TEM imaging showed that the ATP-Au NPs efficiently prevented amyloid fibril formation in vitro by Aβ-42, α-Synuclein as well as by the Glucosylceramide metabolite, and disaggregated their pre-formed fibrils. NMR analysis revealed the interaction of the ATP-Au NPs with the amyloid fibrils. The ATP-Au NPs are safe toward cultured SH-SY5Y cells and when co-incubated with α-Synuclein amyloids inhibited their cytotoxicity and readily enter the cells to inhibit formation of amyloid fibrils within them. The results indicates the pharmacological potentials of ATP decorated Au NPs.  相似文献

The navigational tasks of computing time to impact and controlling movements in specific range are addressed here. We show how time to impact can be obtained via the Mellin-based correlator, and we introduce the concept of fractional based correlators. The fractional Fourier/Mellin based correlators can be used in detecting or controlling specific range of movements. Also, both the conventional and the fractional-based correlators can be easily implemented optically in lenses, thus providing correlation images directly at image acquisition time. Lenses are considered cost-effective and therefore, the optical correlators are optimal in both senses, speed and cost.  相似文献

Antimicrobial packaging can preserve and increase shelf life of free preservatives food products. Active materials present in the packaging material can migrate, in a controlled manner, to the food surface, avoiding bacterial and fungal proliferation and keeping the food product edible for longer periods of time. Essential oils (EO) are natural antimicrobial agents that can be released to the headspace with no direct contact between the package and the food. To minimize loses of EO during high heat melt processing, a three stages process was implemented and tested. Antimicrobial films were prepared by melt mixing a variety of polyethylene copolymers in the presence of organo‐modified montmorillonite nano clay (NC) and thymol, an EO present in oregano and thyme. A controlled EO desorption from films can be achieved by changing the polymer crystallinity and polarity. As the crystallinity increased, the thermal stability of the EO during the extrusion process improved. The addition of NC affects the structure and homogeneity of the crystals. The combination of high polymer crystallinity and chemical affinity between EO and NC increased the thermal stability of the EO during film processing, enabling to control the desorption rate. The effect of multilayer structure based on varied densities and polarities was also studied. Increasing the polarity of the outer layers in multilayered film reduced the EO desorption rate as a result of chemical interactions between the polymer and the EO. Peptide assemblies are ideal components for eco‐friendly optoelectronic energy harvesting devices due to their intrinsic biocompatibility, ease of fabrication, and flexible functionalization. However, to date, their practical applications have been limited due to the difficulty in obtaining stable, high‐performance devices. Here, it is shown that the tryptophan‐based simplest peptide cyclo‐glycine‐tryptophan (cyclo‐GW) forms mechanically robust (elastic modulus up to 24.0 GPa) and thermally stable up to 370 °C monoclinic crystals, due to a supramolecular packing combining dense parallel β‐sheet hydrogen bonding and herringbone edge‐to‐face aromatic interactions. The directional and extensive driving forces further confer unique optical properties, including aggregation‐induced blue emission and unusual stable photoluminescence. Moreover, the crystals produce a high and sustained open‐circuit voltage (1.2 V) due to a high piezoelectric coefficient of 14.1 pC N?1. These findings demonstrate the feasibility of utilizing self‐assembling peptides for fabrication of biointegrated microdevices that combine high structural stability, tailored optoelectronics, and significant energy harvesting properties.  相似文献

We have measured the density of plasma (electron concentration) in discharge maintained in inhomogeneous argon flow under the action of high-power pulsed radiation of gyrotron (frequency, 0.67 THz; power 40 kW; pulse duration, 20–30 μs) in a range of background gas pressures in the discharge chamber from 10–3 to 300 Torr. The electron concentration at low pressures (10–3 to 7 Torr) was determined using Starkeffect induced broadening of the Hα atomic emission line (656.3 nm) of hydrogen present in discharge as a small impurity in residual gases. The maximum observed Stark broadening of the Hα line corresponded to a plasma density on the order of 2 × 1016 cm–3, which exceeded the critical value for the given frequency of radiation sustaining the discharge. At background pressures above 7 Torr, the plasma density was estimated from analysis of the spatiotemporal patterns and waveforms of discharge glow in the visible spectral range. These estimations gave electron concentrations on the level of (1–2) × 1015 cm–3.  相似文献

Self-assembled peptide hydrogels represent the realization of peptide nanotechnology into biomedical products. There is a continuous quest to identify the simplest building blocks and optimize their critical gelation concentration (CGC). Herein, a minimalistic, de novo dipeptide, Fmoc-Lys(Fmoc)-Asp, as an hydrogelator with the lowest CGC ever reported, almost fourfold lower as compared to that of a large hexadecapeptide previously described, is reported. The dipeptide self-assembles through an unusual and unprecedented two-step process as elucidated by solid-state NMR and molecular dynamics simulation. The hydrogel is cytocompatible and supports 2D/3D cell growth. Conductive composite gels composed of Fmoc-Lys(Fmoc)-Asp and a conductive polymer exhibit excellent DNA binding. Fmoc-Lys(Fmoc)-Asp exhibits the lowest CGC and highest mechanical properties when compared to a library of dipeptide analogues, thus validating the uniqueness of the molecular design which confers useful properties for various potential applications.  相似文献

Human factors practitioners (HFPs) play many different roles in the design, creation, operation and maintenance of engineered systems. Less well known are the methods which are aimed at helping with the early stages of design, which are more systems-oriented and often involve questions of the concept of operation in which the engineered system will be fielded. Emerging from the field of cognitive engineering, these methods, including simulation, cognitive work analysis, cognitive task analyses and hierarchical task analysis, will be important as autonomous systems become increasingly capable. Even the most capable systems will continue to interact with humans, and it is at these interfaces between humans and engineered systems that HFP will continue to be needed. This paper describes recent work to leverage these methods to inform concepts of operation in aviation and space, machine learning algorithms and goal-oriented human–machine collaboration.  相似文献

The current study set to examine the effects of simulator use in driving instruction on newly licensed drivers, comparing the road safety knowledge and reported intended behavior, as well as the actual driving performance of new drivers. Participants consisted of 280 newly licensed driver, of which 140 whose drivers license training included additional simulator-based lessons, and 140 drivers whose training precluded simulator-based lessons. All drivers answered questionnaires pertaining to their intended safe driving behaviors (according to Ajzen's (2000) theory of planned behavior), and to their traffic safety knowledge. Of the initial sample, 40 drivers received actual driving performance evaluation by an expert driving instructor, as well as by in-vehicle data recorders (IVDRs). We assumed that safer drivers report safer driving intentions, demonstrate greater traffic safety knowledge, evaluated as safer drivers by the driving instructor, and display lower and stable driving parameters on the IVDRs. We hypothesized that theoretical driving studies combined with practical training on simulators will elevate the safety level of novices driving. Hierarchical regression analyses on driving intentions indicated that drivers who did not receive simulator-based lessons demonstrated safer driving intentions compared to drivers who received simulator-based lessons. This pattern possibly indicating the drivers who received simulator-based lessons felt more confident in their driving abilities compared to drivers who did not receive simulated training. No significant difference was found in traffic safety knowledge, or in the evaluation of the expert driving instructor. IDVR data comparisons indicated drivers who received simulator-based lessons braked more often and were less prone to headway events, suggesting a more responsive driving style. These findings do not point to any significant advantage or disadvantage of the current simulator-based driving training over other driving training methods.  相似文献
