The early-age hydration (≤48 h) of a series of self-compacting concretes and corresponding mortars and one traditionally vibrated concrete and mortar is monitored in a continuous way using ultrasonic testing and isothermal calorimetry. The mixtures differ in type of mineral addition, superplasticizer, cement, cement-to-powder ratio and water-to-powder ratio. The influence of these different mixture compositions on the kinetics of the hydration during the first days of the hydration is characterized by the heat production rate q and the evolution of the p-wave velocity, which is a consequence of the microstructural changes. The variations in the acceleration caused by mineral additions and the deceleration caused by superplasticizers lead to a significantly different behavior. Separating the impact of each of the affecting factors is not always possible due to their combined actions. The nature of the acceleration due to limestone additions and the deceleration caused by polycarboxylate ether superplasticizers can be distinguished clearly, but cannot be quantified. The correlation between the ultrasonic and isothermal calorimetric results is investigated based on parameters related to the start and the end of the setting and reveals the meaningfulness of these parameters when assessing the hydration of self-compacting mixtures with continuous ultrasonic techniques.  相似文献

We report on the results of an experimental study established to optimize the design of microfabricated flow distributors for use in pressure-driven separations and reactions in flat-rectangular channels. For this purpose, the performance of a wide variety of possible flow distributor designs etched in glass/silicon wafers was compared, using CCD camera detection to study the shape and variance of the bands eluting from them. The best performance was obtained with radially interconnected distributors with a diverging inlet section and filled with diamond-shaped pillars, oriented perpendicular to the main flow direction and with a high transversal over axial aspect ratio. It was found that the best distributor designs start with a diverging section containing some 10-12 subsequent rows of high aspect ratio pillars (with a transversal width making up 10-15% of the final channel width) and with a divergence angle selected such that the sloped side-walls run parallel with the sides of the diamond-shaped pillars. After this zone, one or more regions with pillars with a smaller aspect ratio should be provided to increase the number of exit points. To prevent the formation of dead zones in these subsequent zones, so-called distributor wedges can be used to prevent the formation of any dead zones in the wake of the large aspect ratio pillars of the preceding section.  相似文献

In the current styrene production process the distillation of the close-boiling ethylbenzene/styrene mixture to obtain an ethylbenzene impurity level of 100 ppm in styrene accounts for 75–80% of the energy requirements. The future target is to reach a level of 1–10 ppm, which will increase the energy requirements for the distillation even further. Extractive distillation is a well-known technology to separate close-boiling mixtures up to high purities. The objective of this study was to investigate whether extractive distillation using ionic liquids (ILs) is a promising alternative to obtain high purity styrene. Three ILs were studied: [3-mebupy][B(CN)4], [4-mebupy][BF4], and [EMIM][SCN]. Extractive distillation with sulfolane and the current conventional distillation process were used as benchmark processes. The IL [4-mebupy][BF4] is expected to outperform the other two ILs with up to 11.5% lower energy requirements. The operational expenditures of the [4-mebupy][BF4] process are found to be 43.2% lower than the current distillation process and 5% lower than extractive distillation with sulfolane extractive distillations. However, the capital expenditures for the sulfolane process will be about 23% lower than those for the [4-mebupy][BF4] process. Finally, the conclusion can be drawn from the total annual costs that all studied extractive distillation processes outperform the current distillation process to obtain high purity styrene, but that the ILs evaluated will not perform better than sulfolane.  相似文献

Flat-panel X-ray detectors for fluoroscopy represent a modern imaging equipment that is being implemented in paediatric cardiac catheterisation laboratories. Infants and children represent a group of patients with a high radiosensitivity. A survey of 273 (126 diagnostic and 147 therapeutic) paediatric catheterisations was performed to investigate the radiation doses delivered by the new X-ray system. Statistical parameters (75th, 50th and 25th percentiles) of dose-area product (DAP) and fluoroscopy time are reported for patients divided into six age groups: 0-30 d, >1-12 m, >1-3, >3-5, >5-10 and >10-15 y. For accurate risk estimation, effective dose (E) has been determined for all patients using the PCXMC software. For diagnostic procedures, the third quartile of E ranges from 11.3 mSv for newborns to 7 mSv for children of 10-15 y. Therapeutic procedures are more complex than diagnostic. Consequently, the third quartile of E is 22.6 mSv (0-30 d), 18.6 (>1-12 m), 13.3 (>1-3 y), 21.5 (>3-5 y), 17.8 (>5-10 y) and 34.1 mSv (>10-15 y). Dose conversion factors, which relate the DAP and E, have been estimated for each age group. The results of this study may serve as a first step in the optimisation process, in order to make full use of the dose reduction potential of flat-panel systems.  相似文献

It is shown that all C m and C s expressions ever established in the literature are intrinsically fully identical and can be derived from a simple parallel plug flow model. Analytically solving this model yields C m- and C s-term expressions that exactly correspond to the expressions obtained via either the method of moments (general rate model) or via Giddings' nonequilibrium theory. This holds for open-tubular as well as packed and monolithic columns. From this equivalence, a set of convenient "translation" expressions could be established, enabling a swift transition between the different C m- and C s-term expressions that currently coexist in literature. The link with the parallel plug flow model provides a good physical insight into the assumptions underlying the general rate model (e.g., a plug flow in the flow-through pores) and in the physical meaning of the different parameters involved in it. The parallel plug flow model also allows us to illustrate the difference between the zone and the phase retention factor and between the intraparticle diffusion coefficient and the effective diffusion coefficient used in the general rate model. These differences are subtle, but as they can have a large impact, they constitute important potential sources of confusion currently obscuring the modeling of HPLC columns.  相似文献

The necessity for early interaction between the embryo and the oviductal and/or uterine environment in the horse is reflected by several striking differences between equine embryos that develop in vivo and those produced in vitro. Better understanding of the salient interactions may help to improve the efficiency of in vitro equine embryo production. In an initial experiment, cleavage-stage in vitro-produced (IVP) equine embryos were transferred into the uterus of recipient mares that had ovulated recently to determine whether premature placement in this in vivo environment would improve subsequent development. In a second experiment, an important element of the uterine environment was mimicked by adding uterocalin, a major component of the endometrial secretions during early pregnancy, to the culture medium. Intrauterine transfer of cleavage-stage IVP equine embryos yielded neither ultrasonographically detectable pregnancies nor day 7 blastocysts, indicating that the uterus is not a suitable environment for pre-compact morula stage horse embryos. By contrast, exposure to uterocalin during IVP improved capsule formation, although it did not measurably affect the development or expression of a panel of genes known to differ between in vivo and in vitro embryos. Further studies are required to evaluate whether uterocalin serves purely as a carrier protein or more directly promotes improved capsule development.  相似文献

It is demonstrated that the kinetic plot representation of experimental plate height data can also account for practical constraints on the column length, the peak width, the viscous heating, and the mobile-phase velocity without needing any iterative solution routine. This implies that the best possible kinetic performance to be expected from a given tested support under any possible set of practical optimization constraints can always be found using a directly responding calculation spreadsheet template. To show how the resulting constrained kinetic plots can be used as a powerful design and selection tool, the method has been applied to a series of plate height measurements performed on a number of different commercial columns for the same component (butyl-parabene) and mobile-phase composition. The method, for example, allows one to account for the fact that the advantageous solutions displayed by the silica monolith and 5 microm particle columns in the large plate number range of the free kinetic plot are no longer accessible if applying a maximal column length constraint of Lmax = 30 cm. In the plate number range that remains accessible, the investigated sub-2 mum particle columns in any case perform (at least for the presently considered parabene separation) better than the 3.5 mum particle columns or silica monolith, especially if considering the use of system pressures exceeding 400 bar. The constrained kinetic plot method can also be used to select the best-suited column length from an available product gamma to perform a separation with a preset number of plates. One of the optimization results that is obtained in this case is that sometimes a significant gain in analysis time can be obtained by selecting a longer column, yielding the desired plate number at a larger velocity than that for a shorter column.  相似文献

Novel surface plasmon resonance-based biosensor assays for the bioeffect-related screening of chemicals with thyroid-disrupting activity are described. Two thyroid transport proteins (TPs), thyroxine binding globulin (TBG) and recombinant transthyretin (rTTR), were applied in an inhibition assay format in a Biacore 3000 using CM5 biosensor chips coated with l-thyroxine (T4), the main hormone of the thyroid system. Assay conditions were optimized for the natural thyroid hormones, and known thyroid disruptors and structurally related compounds were selected as model compounds to be tested in both assays for their relative potency (RP) compared to T4. The chosen compounds were halogenated phenols, halogenated bisphenols, bisphenol A, 3,5-dichlorobiphenyl, and its hydroxylated metabolite 4-hydroxy-3,5-dichlorobiphenyl (4-OH PCB 14). The TBG-based assay was highly specific for T4, and the rTTR-based assay was sensitive toward several compounds, the highest sensitivity (RP = 4.4) being obtained with 4-OH PCB 14, followed by tetrabromobisphenol A (RP = 1.5) and tetrachlorobisphenol A (RP = 0.75). For the bioeffect-related screening of known and identification of possible new thyroid disruptors, the TPs-based biosensor assays were more sensitive (IC(50) of 13.7 +/- 1.3 and 8.6 +/- 0.7 nM for the rTTR and the TBG-based assay, respectively), easier to perform, and faster alternatives (10 min/sample) than the currently used methods such as radioligand binding assays and immunoprecipitation-HPLC.  相似文献

The Gram-negative bacterium Bordetella pertussis is the causative agent of a respiratory infection known as whooping cough. Previously developed whole-cell pertussis vaccines were effective, but appeared to be too reactogenic mainly due to the presence of lipopolysaccharide (LPS, also known as endotoxin) in the outer membrane (OM). Here, we investigated the possibility of reducing endotoxicity by modulating the LPS levels. The promoter of the lpxC gene, which encodes the first committed enzyme in LPS biosynthesis, was replaced by an isopropyl β-D-1-thiogalactopyranoside (IPTG)-inducible promoter. The IPTG was essential for growth, even when the construct was moved into a strain that should allow for the replacement of LPS in the outer leaflet of the OM with phospholipids by defective phospholipid transporter Mla and OM phospholipase A. LpxC depletion in the absence of IPTG resulted in morphological changes of the cells and in overproduction of outer-membrane vesicles (OMVs). The reduced amounts of LPS in whole-cell preparations and in isolated OMVs of LpxC-depleted cells resulted in lower activation of Toll-like receptor 4 in HEK-Blue reporter cells. We suggest that, besides lipid A engineering, also a reduction in LPS synthesis is an attractive strategy for the production of either whole-cell- or OMV-based vaccines, with reduced reactogenicity for B. pertussis and other Gram-negative bacteria.  相似文献
