Lipofilling (LF) is a largely employed technique in reconstructive and esthetic breast surgery. Over the years, it has demonstrated to be extremely useful for treatment of soft tissue defects after demolitive or conservative breast cancer surgery and different procedures have been developed to improve the survival of transplanted fat graft. The regenerative potential of LF is attributed to the multipotent stem cells found in large quantity in adipose tissue. However, a growing body of pre-clinical evidence shows that adipocytes and adipose-derived stromal cells may have pro-tumorigenic potential. Despite no clear indication from clinical studies has demonstrated an increased risk of cancer recurrence upon LF, these observations challenge the oncologic safety of the procedure. This review aims to provide an updated overview of both the clinical and the pre-clinical indications to the suitability and safety of LF in breast oncological surgery. Cellular and molecular players in the crosstalk between adipose tissue and cancer are described, and heterogeneous contradictory results are discussed, highlighting that important issues still remain to be solved to get a clear understanding of LF safety in breast cancer patients. 相似文献

Extracellular vesicles (EVs) are a family of particles/vesicles present in blood and body fluids, composed of phospholipid bilayers that carry a variety of molecules that can mediate cell communication, modulating crucial cell processes such as homeostasis, induction/dampening of inflammation, and promotion of repair. Their existence, initially suspected in 1946 and confirmed in 1967, spurred a sharp increase in the number of scientific publications. Paradoxically, the increasing interest for EV content and function progressively reduced the relevance for a precise nomenclature in classifying EVs, therefore leading to a confusing scientific production. The aim of this review was to analyze the evolution of the progress in the knowledge and definition of EVs over the years, with an overview of the methodologies used for the identification of the vesicles, their cell of origin, and the detection of their cargo. The MISEV 2018 guidelines for the proper recognition nomenclature and ways to study EVs are summarized. The review finishes with a “more questions than answers” chapter, in which some of the problems we still face to fully understand the EV function and potential as a diagnostic and therapeutic tool are analyzed. 相似文献

Evidence suggests that dietary long chain polyunsaturated fatty acids (LCPUFAs), and particularly those belonging to the n-3 family, may influence the brain fatty acid profile and, thereby, the biosynthesis of endocannabinoids in rodents. However, the doses used are usually quite high and not comparable with human intake. Recently, we have shown that relatively low doses of dietary n-3 LCPUFAs (4 weeks), in the form of either fish or krill oil, balanced for EPA and DHA content, and against a control diet with no EPA and DHA and similar contents of oleic, linoleic and α-linolenic acids, lower the concentrations of the endocannabinoids, anandamide (AEA) and 2-arachidonoylglycerol (2-AG), in the visceral adipose tissue, and of AEA in the liver and heart, of obese Zucker rats. This, in turn, is associated with lower levels of arachidonic acid in membrane phospholipids and with amelioration of some metabolic syndrome parameters. We investigated here whether in Zucker rats, under the same conditions, fish and krill oil are also able to influence LCPUFA and endocannabinoid profiles in brain. Only krill oil was able to increase significantly DHA levels in brain phospholipids, with no changes in arachidonic acid. DHA increase was associated with lower levels of 2-AG in the brain, whereas AEA and its congeners, N-palmitoylethanolamine and N-oleoylethanolamine, were unchanged. We conclude that, despite the strong impact of dietary n-3 fatty acid on endocannabinoid levels previously observed in peripheral tissues, in the brain only 2-AG is affected by dietary krill oil, suggesting that the beneficial effect of the latter on the metabolic syndrome is mostly exerted by modifying peripheral endocannabinoids. Nevertheless, possible effects of dietary krill oil in the brain through modification of 2-AG levels deserve further investigation. 相似文献

In this paper the performances of an electronic nose based on metalloporphyrin‐coated quartz microbalance sensors and of an experienced panel of seven human assessors in the evaluation of gases derived from degradation reactions in tomatoes are presented and discussed. The performances are measured in terms of the capability of both systems to distinguish between samples of different quality coming from conventional and organic production systems. The study deals with the application of pattern recognition techniques based on either multivariate statistical methods (PCA, GPA) or artificial neural networks using a self‐organising map (SOM). The response pattern of the sensor array and the sensory data are analysed and compared using these methods. We report on the deposition of soft matter thin films by Matrix Assisted Pulsed Laser Evaporation (MAPLE). In particular,

This study focuses its attention on the boundary between the growth and no growth of three strains ofSalmonella enteritidis , Bacillus cereus and Staphylococcus aureus in the presence of growth controlling factors such as temperature, pH, water activity (Aw) and ethanol concentration. Preliminarly, the minimal values of pH, Awand temperature, and the maximum ethanol concentrations allowing the growth of the considered micro-organisms were determined. The calculation of these values enabled the use of logistic model to evaluate the growth/no growth boundary for the bacteria in relation to the considered independent variables. The location of the growth/no-growth boundaries for S. enteritidis and Staph. aureus were strongly affected, at the same ethanol concentration, by temperature, pH and Aw. Among the considered species, Staph. aureus was endowed with the highest sensitivity to low pH values whileB. cereus's growth/no growth interface, was quite unaffected by the combination of the stresses, when the physico–chemical conditions were above the minimum for growth. The effects of temperature, Awand ethanol on the limitation of growth of the considered species were not merely additive. It was possible to identify the combinations of such factors preventing the growth of Salmonella enteritidis, Staph. aureus and B. cereus. 相似文献

Ultraviolet B (UVB) radiation acts as a strong apoptotic trigger in many cell types, in tumor and normal cells. Several studies have demonstrated that UVB-induced cell death occurs through the generation of reactive oxygen species. The consequent oxidative stress includes the impairment of cellular antioxidants, the induction of DNA damage and the occurrence of apoptosis. In this review, we investigated UVB apoptotic action in various cell models by using ultrastructural, molecular and cytofluorimetric techniques. Myeloid leukemia HL-60, T-lymphoblastoid Molt-4 and myelomonocytic U937 human cells, generally affected by apoptotic stimuli, were studied. Human chondrocytes and C2C12 skeletal muscle cells, known to be more resistant to damage, were also considered. All of them, when exposed to UVB radiation, revealed a number of characteristic apoptotic markers. Membrane blebbing, cytoplasm shrinkage and chromatin condensation were detected by means of electron microscopy. DNA cleavage, investigated by using agarose gel electrophoresis and TUNEL reaction, was observed in suspended cells. Differently, in chondrocytes and in skeletal muscle cells, oligonucleosomic DNA fragmentation did not appear, even if a certain TUNEL positivity was detected. These findings demonstrate that UVB radiation appears to be an ideal tool to study the apoptotic behavior. 相似文献

The thermal behavior of pure ZrO2 and four ZrO2-based organic-inorganic hybrids (OIHs) containing increasing amount (6, 12, 24 and 50?wt%) of poly(ε-caprolactone) (PCL) (named Z, ZP6, ZP12, ZP24 and ZP50 respectively) has been studied by simultaneous thermogravimetry (TG) and differential scanning calorimetry (DSC). The FTIR analysis of the gas mixture evolved at defined temperatures from the samples submitted to the TG experiments identified the mechanism of each thermally activated process. The obtained results suggest that the inorganic matrix of the OIHs prepared by this method exerts a stabilizing effect on the polymer, in particular for poor-PCL hybrid materials. In fact, the different thermal behavior of the ZP50 sample suggests that the polymer is not entirely bonded to the -OH groups of the zirconia matrix due to their saturation. For this reason a part of PCL is not affected by the stabilizing effect of the matrix and is subjected to thermal degradation. Finally, by observing their thermal behavior it was possible to select the most suitable temperatures for thermal pretreatment: 400, 600 and 1000?°C. The structural analysis by X-ray diffraction (XRD) revealed that at 400?°C the materials are amorphous, while at 600?°C they are mostly tetragonal, and the content of the tetragonal phase decreases with increasing the amount of PCL in the OIHs. All the materials treated at 1000?°C are monoclinic, but their crystallinity decreases with increasing the PCL content. 相似文献

In this paper we investigate the surface chemistry, including surface contaminations, of SnO2 nanowires deposited on Ag-covered Si substrate by vapor phase deposition (VPD), thanks to x-ray photoelectron spectroscopy (XPS) in combination with thermal desorption spectroscopy (TDS). Air-exposed SnO2 nanowires are slightly non-stoichiometric, and a huge amount of C contaminations is observed at their surface. After the thermal physical desorption (TPD) process, SnO2 nanowires become almost stoichiometric without any surface C contaminations. This is probably related to the fact that C contaminations, as well as residual gases from air, are weakly bounded to the crystalline SnO2 nanowires and can be easily removed from their surface. The obtained results gave us insight on the interpretation of the aging effect of SnO2 nanowires that is of great importance for their potential application in the development of novel chemical nanosensor devices. 相似文献

Proliferating thyroid cells are more sensitive to UV-C radiations than quiescent cells. The effect is mediated by nuclear phosphatidylcholine and sphingomyelin metabolism. It was demonstrated that proton beams arrest cell growth and stimulate apoptosis but until now there have been no indications in the literature about their possible mechanism of action. Here we studied the effect of protons on FRTL-5 cells in culture. We showed that proton beams stimulate slightly nuclear neutral sphingomyelinase activity and inhibit nuclear sphingomyelin-synthase activity in quiescent cells whereas stimulate strongly nuclear neutral sphingomyelinase activity and do not change nuclear sphingomyelin-synthase activity in proliferating cells. The study of neutral sphingomyelinase/sphingomyelin-synthase ratio, a marker of functional state of the cells, indicated that proton beams induce FRTL-5 cells in a proapoptotic state if the cells are quiescent and in an initial apoptotic state if the cells are proliferating. The changes of cell life are accompanied by a decrease of nuclear sphingomyelin and increase of bax protein. 相似文献
