In vivo quantification of neuroanatomical shape variations is possible due to recent advances in medical imaging and has proven useful in the study of neuropathology and neurodevelopment. In this paper, we apply a spherical wavelet transformation to extract shape features of cortical surfaces reconstructed from magnetic resonance images (MRIs) of a set of subjects. The spherical wavelet transformation can characterize the underlying functions in a local fashion in both space and frequency, in contrast to spherical harmonics that have a global basis set. We perform principal component analysis (PCA) on these wavelet shape features to study patterns of shape variation within normal population from coarse to fine resolution. In addition, we study the development of cortical folding in newborns using the Gompertz model in the wavelet domain, which allows us to characterize the order of development of large-scale and finer folding patterns independently. Given a limited amount of training data, we use a regularization framework to estimate the parameters of the Gompertz model to improve the prediction performance on new data. We develop an efficient method to estimate this regularized Gompertz model based on the Broyden-Fletcher-Goldfarb-Shannon (BFGS) approximation. Promising results are presented using both PCA and the folding development model in the wavelet domain. The cortical folding development model provides quantitative anatomic information regarding macroscopic cortical folding development and may be of potential use as a biomarker for early diagnosis of neurologic deficits in newborns.  相似文献

The TEX86 is a recently proposed paleothermometer through which ancient seawater temperatures of up to 120 My ago can be reconstructed. It is based on the relative distribution of glycerol dibiphytanyl glycerol tetraethers as measured by high-performance liquid chromatography/atmospheric pressure chemical ionization-mass spectrometry (HPLC/APCI-MS). The aim of this study was to examine and improve several analytical aspects in the determination of this important proxy in environmental matrices. Comparison of TEX86 analysis using single ion mode (SIM) and mass scanning (m/z 950 to 1450) detection, respectively, revealed that SIM is up to 2 orders of magnitude more sensitive and that the TEX86 can be determined with a reproducibility of +/-0.004 or +/-0.3 degrees C using this method. Comparison of TEX86 values obtained with two different HPLC/APCI-MS set-ups revealed no significant differences. In addition, analysis of TEX86 of extracts obtained by Soxhlet, ultrasonic, and accelerated high-pressure extraction techniques also showed no significant differences between the methods. Our results suggest that TEX86 analysis by HPLC/APCI-MS is robust and can be determined with analytical errors comparable to those of other temperature proxies.  相似文献

The present paper describes the development and validation of a normal-phase liquid chromatography-mass spectrometry (NP-HPLC-MS) method for the screening and quantification of vitamin E constituents in human plasma and food matrixes. Liquid-liquid extraction combined with isotope dilution was applied to extract the lipophilic target analytes. Baseline separation of alpha-tocopherylacetate, alpha-tocopherol, alpha-tocotrienol, alpha-tocopherylquinone, beta-tocopherol, gamma-tocopherol, beta-tocotrienol, gamma-tocotrienol, delta-tocopherol, and delta-tocotrienol was achieved utilizing a normal-phase amine column operated with n-hexane and 1,4-dioxane as solvents. Detection was achieved by positive-ion atmospheric-pressure chemical ionization (APCI). Key features of the method are lower limits of detection, 3-51 nmoles/L; lower limits of quantification, 8-168 nmoles/L; linearity coefficients, 0.9778-0.9989; linear ranges, 0.01-29 micromol/L; recoveries, 53-92%; accuracies, 99-103%; intraday precisions, 2-17%; interday precisions, 5-18%; and suppression values, 0-29%. Fragmentation of tocopherols was studied by tandem mass spectrometry, and a fragmentation scheme for tocotrienols/tocopherols is postulated. Neutral-loss and precursor-ion scan experiments were performed for targeted discovery of oxidation products of tocopherols in human blood and fish oil, the latter being an important food component. The presented data suggest that this method will help to expand the number of quantified/discovered vitamin E constituents detected in food products and analyzed during human/animal trials in order to give a more comprehensive picture to nutritionists about the fate of vitamin E.  相似文献

Herein, we report on the temperature‐dependent crystal structures of Ti 3 AlC 2 and Ti 3 Al 0.8 Sn 0.2 C 2 in the 373–1273 K temperature range, as determined by Rietveld analysis of high‐temperature neutron diffraction time‐of‐flight data. The compositions are 86(1) wt% Ti 3 AlC 2 and 14(1) wt% TiC 0.92(2) for the sample with no Sn , and 95(1) wt% Ti 3( Al 0.8 Sn 0.2) C 2 and 5(1) wt% Ti 2 AlC for the solid solution with Sn . The average linear volumetric thermal expansion is 8.0(2) × 10?6 K ?1 for Ti 3 AlC 2 and 8.2(5) × 10?6 K?1 for Ti 3( Al 0.8 Sn 0.2) C 2. The average linear thermal expansion in the a and c directions, respectively, are 7.6(2) × 10?6 K?1 and 8.9(2) × 10?6 K?1 for Ti 3 AlC 2. For Ti 3( Al 0.8 Sn 0.2) C 2, the respective values are 8.0(5) × 10?6 K?1 and 8.6(6) × 10?6 K?1. In the case of the solid solution, the quadratic thermal expansion coefficients are also given. Detailed bond lengths analysis shows that the thermal expansions along the a and c directions are controlled by the thermal expansions of the Ti – C , and Ti – Al bond lengths, respectively. The atomic displacement parameters (ADPs) show that the Al and Sn atoms vibrate with a higher amplitude than the Ti and C atoms. Consistent with first‐principles calculations, the ADPs of the Al/Sn site(s) in Ti 3( Al 0.8 Sn 0.2) C2 are lower than the ADPs of Al in Ti 3 AlC 2.  相似文献

The objective of this study was to determine the incorporation of conjugated linoleic acid (CLA) into triacylglycerols (TAG) and phospholipids (PL) of tissues and plasma, and to interpret the role of dietary‐derived vaccenic acid (VA) in increasing the tissue content of CLA (c9,t11) and the influence on the fatty acid profile. We fed five groups of rats semi‐purified diets with varying levels of CLA and VA: control butter with low CLA (c9,t11) and VA; control butter added 5% CLA (c9,t11); control butter added 5% Tonalin [equal amount of CLA (c9,t11) and CLA (t10,c12)]; control butter added 5% VA; butter with high CLA (c9,t11) and VA (H‐CLA), for 3 weeks. The highest incorporation of CLA (c9,t11) was found in adipose tissue, and the lowest was observed in liver. Low intake of CLA (c9,t11) combined with high intake of VA resulted in a higher incorporation of CLA (c9,t11) in tissues due to the conversion of VA to CLA (c9,t11), compared to feeding CLA (c9,t11) without VA. However, in enterocytes, the proportion of CLA (c9,t11) was low after feeding VA, indicating no or only a minor conversion of VA to CLA (c9,t11) in the intestine. The incorporation of CLA (t10,c12) into TAG from plasma and tissues was generally much lower than that of the CLA (c9,t11) isomer, except in the enterocyte TAG, which had similar proportions of the two isomers.  相似文献

We present a modern, light portable device specifically designed for environmental samples for cryogenic transmission-electron microscopy (cryo-TEM) by on-site cryo-plunging. The power of cryo-TEM comes from preparation of artifact-free samples. However, in many studies, the samples must be collected at remote field locations, and the time involved in transporting samples back to the laboratory for cryogenic preservation can lead to severe degradation artifacts. Thus, going back to the basics, we developed a simple mechanical device that is light and easy to transport on foot yet effective. With the system design presented here we are able to obtain cryo-samples of microbes and microbial communities not possible to culture, in their near-intact environmental conditions as well as in routine laboratory work, and in real time. This methodology thus enables us to bring the power of cryo-TEM to microbial ecology.  相似文献

Objective

The heavy metals lead (Pb) and mercury (Hg) are ubiquitous environmental pollutants with high neurotoxic potential. We aimed to compare perinatal Pb and Hg concentrations and to explore the potential association between Pb and Hg exposure and newborn anthropometry.

Study design

Pregnant women were recruited in 2005 at the General Hospital Vienna for participation in this longitudinal study. Pb and Hg concentrations were measured in maternal blood and hair, placenta, cord blood, meconium, and breast milk of 53 mother-child pairs by CV-AAS, GF-AAS, and HPLC-CV-ICPMS. We conducted bivariate analyses and categorical regression analysis (CATREG) to evaluate the determinants of Pb and Hg exposure, and of infant anthropometry.

Results

Median Pb and total Hg contents were low, i.e., 25 μg/L (maternal blood-Pb), 13 μg/L (cord blood-Pb), 0.7 μg/L (maternal blood-Hg), and 1.1 μg/L (cord blood-Hg). Hg levels in maternal and fetal tissues were frequently correlated (r > 0.3, P < 0.05, respectively). Regarding Pb, only maternal blood and cord blood concentrations correlated (P = 0.043). Cord blood levels indicated higher Hg exposure but lower Pb exposure relative to maternal blood contents. Adjusted CATREG models indicated the significant predictors of birth length (placenta-Pb, gestational length, meconium-Pb), birth weight (placenta-Pb, gestational length, maternal blood-Pb), and head circumference (maternal education, maternal height). Besides one significant correlation between maternal hair Hg and birth length, the mercury levels were not associated with newborn anthropometry.

Conclusions

Our data implicate that different modes of action may exist for placentar transfer of Pb and Hg as well as that low Pb exposure levels can result in lower birth weight. The findings related to newborn anthropometry need to be confirmed by the examination of larger study groups. Further research is needed to clarify the mechanisms of Pb and Hg transfer via the placenta, and to explore how prenatal Pb exposure is related to intrauterine growth.  相似文献

The ‘people and performance’ model asserts that performance is a sum of employee ability, motivation and opportunity (AMO). Despite extensive evidence of this people–performance link within manufacturing and many service sectors, studies within the construction industry are limited. Thus, a recent research project set out to explore the team deployment strategies of a large construction company with the view of establishing how a balance could be achieved between organisational strategic priorities, operational project requirements and individual employee needs and preferences. The findings suggested that project priorities often took precedence over the delivery of the strategic intentions of the organisation in meeting employees’ individual needs. This approach is not sustainable in the long term because of the negative implications that such a policy had in relation to employee stress and staff turnover. It is suggested that a resourcing structure that takes into account the multiple facets of AMO may provide a more effective approach for balancing organisational strategic priorities, operational project requirements and individual employee needs and preferences more appropriately in the future.  相似文献
