1H spin-lattice relaxation experiments have been performed for a series of ionic liquids including bis(trifluoromethanesulfonyl)imide anion and cations of a varying alkyl chain length: triethylhexylammonium, triethyloctylammonium, decyltriethylammonium, dodecyltriethylammonium, triethyltetradecylammonium, and hexadecyltriethylammonium. The relaxation studies were carried out in abroad frequency range covering three orders of magnitude, from 10 kHz to 10 MHz, versus temperature. On the basis of a thorough, quantitative analysis of this reach data set, parameters characterizing the relative, cation-cation, translation diffusion (relative diffusion coefficients and translational correlation times), and rotational motion of the cation (rotational correlation times) were determined. Relationships between these quantities and their dependence on the alkyl chain length were discussed in comparison to analogous properties of molecular liquids. It was shown, among other findings, that the ratio between the translational and rotational correlation times is smaller than for molecular liquids and considerably dependent on temperature. Moreover, a comparison of relative and self-diffusion coefficients indicate correlated translational dynamics of the cations.  相似文献

A multiplex PCR assay was developed for the detection and differentiation of the Yersinia enterocolitica and Yersinia pseudotuberculosis isolates in both pure bacterial cultures and pig tonsils. The assay was based on the amplification of the ail, inv, yadA, and ystB genes. The PCR products, corresponding to the ail gene and the plasmid-borne yadA gene or only one product corresponding to the ail gene, were detected in Y. enterocolitica 4 biotype isolates. All of the Y. pseudotuberculosis isolates (n=6) tested gave a positive PCR reaction for the inv gene. For all tested Y. enterocolitica 1A biotype isolates (n=31), one product corresponding to the ystB gene was observed. The multiplex PCR assay was used to detect Y. enterocolitica and Y. pseudotuberculosis strains in pig tonsil samples obtained from 80 slaughtered pigs from three different herds. The presence of at least one of the specific PCR amplification products of ail-, ystB-, yadA-, and inv-specific sequences was observed in 11 samples (13.75%). These results of the multiplex PCR assay were compared with the results of conventional, microbiological testing. Y. enterocolitica isolates were cultured from only 3 (3.75%) of the 80 pig tonsils examined. The multiplex PCR assay was shown to be an efficient tool for differentiation between the pYV plasmid-bearing Y. enterocolitica isolates, the plasmidless Y. enterocolitica isolates, the Y. enterocolitica biotype 1A isolates, and the Y. pseudotuberculosis isolates with and without the pYV plasmid in naturally contaminated pig tonsils. This indicates that this assay is useful to control food processing and track the source of contamination.  相似文献

BACKGROUND: The effect of Brochothrix thermosphacta on the quality of meat and meat products is of vital importance in connection with Regulation EC/178/2002 extending the definition of unsafe foodstuffs to encompass all those which are unfit for human consumption. This study aimed to determine the prevalence of B. thermosphacta in meat and meat products packaged under different conditions and to estimate the effect of B. thermosphacta strains on product quality based on their protein and lipid degradation activity. RESULTS: B. thermosphacta was absent in only two of 132 samples. All other samples were contaminated with this bacterium (101 to 109cfu g?1 meat and 102 to 108cfu g?1meat product). In products stored under high‐oxygen atmosphere Brochothrix cells accounted for almost 100% total mesophilic count (TMC) and below 50% TMC in oxygen‐free atmosphere. While the tested B. thermosphacta strains did not show any proteolytic activity, most of them displayed lipolytic activity at 25 °C and some even at 4 °C. CONCLUSION: B. thermosphacta is commonly present in meat and meat products packaged in different ways. This bacterium can display lipolytic activity also at refrigeration temperature. Its over‐proliferation can be inhibited through vacuum packaging or packaging under a modified atmosphere with reduced oxygen content. Copyright © 2011 Society of Chemical Industry  相似文献

The effect of succinylation on native rapeseed albumin was studied using polyacrylamide gel electrophoresis (PAGE), isoelectric focusing, viscometry, turbidimetry and by investigation of some functional properties (heat-induced aggregation, emulsifying and foaming properties). The protein can be quickly modified with a relatively small excess of succinic anhydride. A 98% succinylation of the protein amino groups could be attained. The PAGE patterns point out a relatively high heterogeneity of the succinylated protein. The isoelectric focusing shows, however, a concentration of the succinylated protein fractions in a near pI range (pH 4.45 to 4.60). The intrinsic viscosity [η] of the native albumin drops after a 50% succinylation from 0.15 dl· g?1 to 0.06–0.07 dl. g1. Increasing the degree of succinylation to 88% increases [η] to 0.09 dl· g?1. The native albumin possesses excellent whipping properties and high foam stability. Succinylation did not improve the foam capacity but decreases the foam stability. The emulsifying properties could not be improved by succinylation. The heat-induced aggregation of the rapeseed albumin in dilute solution is inhibited by a moderate or high succinylation.  相似文献

New substitutional, limited solid solution of the formula Y8-yYbyV2O17 where 0 < y ≤ 2.0 was synthesized by the high-temperature reaction in the air from mixtures of α-Y8V2O17 with Yb8V2O17. The new Y8-yYbyV2O17 were characterized by powder XRD, DTA–TGA, IR, UV–vis-DRS and SEM methods. The structure, thermal stability, unit cell volume, band gap energies as well as the position of the IR absorption bands in the spectrum of Y8-yYbyV2O17 were determined. The solid solution was established to have the same structure as β-Y8V2O17 and it was found that with increasing concentration of Yb3+ ions in its crystal lattice the volume of its unit cell decreases along with its energy gap decreasing to 2.4 eV (for y = 2.0), while the IR absorption bands are shifted towards higher wavenumbers. The morphologies of the high temperature polymorph of Y8V2O17 and solid solution were analyzed.  相似文献

There are two main possibilities to substitute conventional wood material in the panel production: the use of wood of fast-growing trees plantations and/or the use of alternative ligno-cellulosic raw materials. Especially particleboards produced from the ‘‘energetic’’ willow present the same or better mechanical parameters compared to particleboards conventionally produced mainly from soft woods. Increasing bio-fuel production, mostly from rape, will cause an escalation of the rape cultivation area and, as a result, will augment the amount of waste (rape straw) to be utilized. The aim of this study was to verify the friction coefficient of the new board types in tribological contact with tools surfaces. The investigations were performed in situ. The results of the friction coefficient related to the tools surfaces as well as lingo-cellulose materials are presented.  相似文献

The 5‐substituted 2‐thiouridines (R5S2Us) present in the first (wobble) position of the anticodon of transfer RNAs (tRNAs) contribute to accuracy in reading mRNA codons and tuning protein synthesis. Previously, we showed that, under oxidative stress conditions in vitro, R5S2Us were sensitive to hydrogen peroxide (H2O2) and that their oxidative desulfuration produced 5‐substituted uridines (R5Us) and 4‐pyrimidinone nucleosides (R5H2Us) at a ratio that depended on the pH and an R5 substituent. Here, we demonstrate that the desulfuration of 2‐thiouridines, either alone or within an RNA/tRNA chain, is catalyzed by cytochrome c (cyt c). Its kinetics are similar to those of Fenton‐type catalytic 2‐thiouridine (S2U) desulfuration. Cyt c/H2O2‐ and FeII‐mediated reactions deliver predominantly 4‐pyrimidinone nucleoside (H2U)‐type products. The pathway of the cyt c/H2O2‐peroxidase‐mediated S2U→H2U transformation through uridine sulfenic (U‐SOH), sulfinic (U‐SO2H), and sulfonic (U‐SO3H) intermediates is confirmed by LC–MS. The cyt c/H2O2‐mediated oxidative damage of S2U‐tRNA may have biological relevance through alteration of the cellular functions of transfer RNA.  相似文献

The mechanism(s) by which zinc is transported into cells has not been identified. Since zinc uptake is inhibited by reducing the temperature, zinc uptake may depend on the movement of plasma membrane micoenvironments, such as endocytosis or potocytosis. We investigated the potential role of potocytosis in cellular zinc uptake by incubating normal and acrodermatitis enteropathica fibroblasts with nystatin, a sterol-binding drug previously shown to inhibit potocytosis. Zinc uptake was determined during initial rates of uptake (10 min) following incubation of the fibroblasts in 50 micrograms nystatin/mL or 0.1% dimethylsulfoxide for 10 min at 37 degrees C. The cells were then incubated with 1 to 30 microM 65zinc. Michaelis-Menten kinetics were observed for zinc uptake. Nystatin inhibited zinc uptake in both the normal and AE fibroblasts. Reduced cellular uptake of zinc was associated with its internalization, not its external binding. In normal fibroblasts, nystatin significantly reduced the K(m) 56% and the Vmax 69%. In the AE fibroblasts, nystatin treatment significantly reduced the Vmax 59%, but did not significantly affect the K(m). The AE mutation alone affected the Vmax for cellular zinc uptake. The control AE fibroblasts exhibited a 40% reduction in Vmax compared to control normal fibroblasts. We conclude that nystatin exerts its effect on zinc uptake by reducing the velocity at which zinc traverses the cell membrane, possibly through potocytosis. Furthermore, the AE mutation also affects zinc transport by reducing zinc transport.  相似文献

A novel transduction chemistry for preparing optical anion-selective polymeric films that respond reversibly and selectively to chloride ion activity is demonstrated. The chloride sensors are prepared by casting thin (5-10 microm) plasticized PVC films containing indium(III) octaethylporphyrin hydroxide, along with optimized levels of a lipophilic tetraphenylborate salt, onto glass slides. When bathed in low-pH buffered solutions void of chloride, the porphyrin species spontaneously forms a hydroxide ion-bridged dimer, with the added lipophilic borate species serving as the counteranion for this complex. The maximum for the Soret absorption band of this dimeric species is shifted to 390 nm, from 410 nm for the initial monomeric porphyrin. Increases in chloride ion levels in the bathing solution results in chloride extraction and ligation to the In(III) center, and concomitant breaking of the dimer into monomeric porphyrin species, yielding a decrease in absorbance at 390 nm and an increase in optical signal at 410 nm. Under optimized conditions, optical selectivity coefficients toward chloride over a wide range of other anions (NO3-, ClO4-, SCN-, SO4(2-), F-, Br-, H2PO4-) are measured to be < 10(-3). Of all anions tested, only salicylate yields a slightly greater response than chloride. This selectivity is shown to be adequate for reversible and accurate sensing of chloride levels in diluted serum samples.  相似文献
