Myocardial infarction (MI) is one of the leading causes of heart-related deaths worldwide. Following MI, the hypoxic microenvironment triggers apoptosis, disrupts the extracellular matrix and forms a non-functional scar that leads towards adverse left ventricular (LV) remodelling. If left untreated this eventually leads to heart failure. Besides extensive advancement in medical therapy, complete functional recovery is never accomplished, as the heart possesses limited regenerative ability. In recent decades, the focus has shifted towards tissue engineering and regenerative strategies that provide an attractive option to improve cardiac regeneration, limit adverse LV remodelling and restore function in an infarcted heart. Acellular scaffolds possess attractive features that have made them a promising therapeutic candidate. Their application in infarcted areas has been shown to improve LV remodelling and enhance functional recovery in post-MI hearts. This review will summarise the updates on acellular scaffolds developed and tested in pre-clinical and clinical scenarios in the past five years with a focus on their ability to overcome damage caused by MI. It will also describe how acellular scaffolds alone or in combination with biomolecules have been employed for MI treatment. A better understanding of acellular scaffolds potentialities may guide the development of customised and optimised therapeutic strategies for MI treatment. 相似文献

Two-thirds of differentiated thyroid cancer (DTC) patients with distant metastases would be classified as radioactive iodine-refractory (RAIR-DTC), evolving into a poor outcome. Recent advances underlying DTC molecular mechanisms have shifted the therapy focus from the standard approach to targeting specific genetic dysregulations. Lenvatinib and sorafenib are first-line, multitargeted tyrosine kinase inhibitors (TKIs) approved to treat advanced, progressive RAIR-DTC. However, other anti-angiogenic drugs, including single targeted TKIs, are currently being evaluated as alternative or salvage therapy after the failure of first-line TKIs. Combinatorial therapy of mitogen-activated protein kinase (MAPK) and phosphoinositide 3-kinase (PI3K) signalling cascade inhibitors has become a highly advocated strategy to improve the low efficiency of the single agent treatment. Recent studies pointed out targetable alternative pathways to overcome the resistance to MAPK and PI3K pathways’ inhibitors. Because radioiodine resistance originates in DTC loss of differentiation, redifferentiation therapies are currently being explored for efficacy. The present review will summarize the conventional management of DTC, the first-line and alternative TKIs in RAIR-DTC, and the approaches that seek to overcome the resistance to MAPK and PI3K pathways’ inhibitors. We also aim to emphasize the latest achievements in the research of redifferentiation therapy, immunotherapy, and agents targeting gene rearrangements in advanced DTC. 相似文献

With reference to a real industrial application of process control, some considerations are discussed concerning the accuracy of methods for auto-tuning of proportional, integral and derivative factor (PID). In particular, a theoretical–experimental approach is described, that allows to evaluate the adequateness of new methods for auto-tuning of PID, able to significantly reduce the time duration for auto-tuning with respect to traditional ones. This result has been achieved by using suitable techniques of experimental data processing, based on neural-networks algorithms, set for this specific application. Amineboranes of the type L-BH3 (L = NH3; tert-BuNH2; Me2NH; Me3N) and sodium cyanotrihydroborate(III) (NaBH3CN) have been tested as derivatization reagents in the generation of volatile hydrides and elemental mercury following aqueous phase reaction with ionic species of Hg(II), As(III), As(V), Sb(V), Sb(III), Bi(III), Se(IV), Se(VI), Te(IV), and Te(VI). Continuous flow generation atomic absorption spectrometry coupled with a flameless quartz tube atomizer (T = 25 degrees C) and a miniature argon-hydrogen diffusion flame atomizer were employed for the detection of mercury vapors and volatile hydrides, respectively. All of the reductants were able to reduce Hg(II) to the elemental state, giving sensitivities comparable to NaBH4 reduction. Under reaction conditions giving maximum sensitivity for hydride generation with NaBH4, only some amineboranes are able to produce volatile hydrides from all the elements. No evidence of hydride formation was observed from the Se(VI) and Te(VI). In general, the reducing power decreased in the order NaBH4 > H3N-BH3 > tert-BuNH2-BH3 > NaBH3CN > or = Me2HN-BH3 > Me3N-BH3. In comparison with THB, amineboranes and NaBH3CN allowed, in general, a better control of interference effects of Fe(III), Ni(II), Co(II), and Cu(II). Application to determination of mercury in certified reference material is reported. The most likely mechanism of reaction of borane complexes in chemical vapor generation is based on the direct action of hydrogen bound to boron. 相似文献

The present review deals with the morphological features of the removal of apoptotic cells by liver. The engulfment of cells undergoing apoptosis can be considered a specialized form of phagocytosis, playing a major role in the general tissue homeostasis in physiological and pathological conditions. In fact, defects of phagocytosis of apoptotic cells might have deleterious consequences for neighboring healthy cells, i.e., pathogenesis of inflammatory disease or dysregulation of the immune system. Phagocytosis of apoptotic cells by liver is a complex phenomenon, involving multiple molecular mechanisms of recognition (i.e., lectin-like receptors and receptors for externalized phosphatydilserine) of both parenchymal (hepatocytes) and nonparenchymal (Kupffer and endothelial cells) liver cells, often operating in cooperation. The data discussed in the present review are drawn from studies of phagocytosis of apoptotic cells in the liver, carried out with in vivo and in situ adhesion experiments as well as in vitro assays. Our results indicate that the three main liver cell types (hepatocytes, Kupffer, and endothelial cells) are able to recognize and internalize apoptotic cells by means of specific receptors (galactose and mannose-specific receptor; receptor for phosphatydilserine) and by cytoskeletal reorganization that favors the engulfment of the apoptotic cells. The "flags" for the identification of apoptotic cells by the liver are modifications of the surface of dead cells, i.e., sugar residues and phosphatydilserine exposition. Vitronectin receptor is not involved in such a recognition. The adhesions between modified cell surfaces of apoptotic cells and phagocytes generate cytoplasmatic signaling pathways that drive apoptotic cells to their final fate within the phagocytes (i.e., lysosomal digestion). 相似文献

Recently, several test case prioritization (TCP) techniques have been proposed to order test cases for achieving a goal during test execution, particularly, revealing faults sooner. In the model-based testing (MBT) context, such techniques are usually based on heuristics related to structural elements of the model and derived test cases. In this sense, techniques’ performance may vary due to a number of factors. While empirical studies comparing the performance of TCP techniques have already been presented in literature, there is still little knowledge, particularly in the MBT context, about which factors may influence the outcomes suggested by a TCP technique. In a previous family of empirical studies focusing on labeled transition systems, we identified that the model layout, i.e., amount of branches, joins, and loops in the model, alone may have little influence on the effectiveness of TCP techniques investigated, whereas characteristics of test cases that actually fail definitely influences this aspect. However, we considered only synthetic artifacts in the study, which reduced the ability of representing properly the reality. In this paper, we present a replication of one of these studies, now with a larger and more representative selection of techniques and considering test suites from industrial systems as experimental objects. Our objective is to find out whether the results remain while increasing the validity in comparison to the original study. Results reinforce that there is no best performer among the investigated techniques and characteristics of test cases that fail represent an important factor, although adaptive random-based techniques are less affected by it. 相似文献

This paper analyses the role of the employers' education on the spread of temporary contracts. Taking advantage of a unique firm‐level dataset, we test whether the share of fixed term workforce in a firm is affected by the employer's level of education. Furthermore, we test whether knowledge spillovers arising from the agglomeration of university graduate employers affect the incidence of temporary employment in the firms located in the area. In both cases we find a negative effect. Interestingly, only small firms are influenced by the spillovers. The possible problems of endogeneity of the agglomeration variable are coped with an IV approach. 相似文献

Milk filtration procedures are gaining relevance in the dairy industry because milk ultra- and nanofiltrates are used to increase milk processing efficiency, and as additives for products with improved nutraceutical properties. This study aimed to develop Fourier-transformed mid-infrared spectroscopy calibrations for ultra- and nanopermeate and retentate fractions of defatted and delactosated milk. A total of 154 samples from different milk fractions were collected and analyzed using reference methods to determine protein, solids-not-fat, glucose, and galactose content. The obtained values were matched with their respective Fourier-transformed mid-infrared spectroscopy spectra to develop new prediction models. Calibrations for each trait were built following 3 different approaches to get the best prediction models: (1) using the entire data set, (2) using 3 subsets based on component concentrations (level approach), and (3) using hierarchical clusters calculated with pairwise Mahalanobis distance among spectra (cluster approach). Calibrations were developed using partial least squares regression, after removing low signal-to-noise ratio wavelengths, and validated through a leave-one-out cross-validation procedure. In addition, the accuracy of the predicted values within each fraction was checked for each approach. Dividing the data set into subsets improved prediction models for each trait and for the samples in each milk fraction. Without considering milk fraction, the best improvement was observed for glucose and galactose. Glucose ratio performance deviation in cross-validation (RPD) increased from 7.42 to 11.31 and 11.06, for cluster and level approaches, respectively, whereas galactose RPD increased from 8.86 to 11.69 and 11.27 for cluster and level approaches, respectively. Considering milk fractions, the best improvement was observed for protein content, where RPD ranged from 0.08 to 6.06 for the whole data set calibration, whereas it ranged from 0.43 to 40.34 for the subset calibration approaches. Cluster and level approaches to build calibration models were comparable for samples from different fractions, suggesting that the 2 subsetting protocols should be both investigated to get the best prediction performances. 相似文献

Photofrin/photodynamic therapy (PDT) at sub-lethal doses induced a transient stall in proteasome activity in surviving A549 (p53+/+) and H1299 (p53−/−) cells as indicated by the time-dependent decline/recovery of chymotrypsin-like activity. Indeed, within 3 h of incubation, Photofrin invaded the cytoplasm and localized preferentially within the mitochondria. Its light activation determined a decrease in mitochondrial membrane potential and a reversible arrest in proteasomal activity. A similar result is obtained by treating cells with Antimycin and Rotenone, indicating, as a common denominator of this effect, the ATP decrease. Both inhibitors, however, were more toxic to cells as the recovery of proteasomal activity was incomplete. We evaluated whether combining PDT (which is a treatment for killing tumor cells, per se, and inducing proteasome arrest in the surviving ones) with Bortezomib doses capable of sustaining the stall would protract the arrest with sufficient time to induce apoptosis in remaining cells. The evaluation of the mitochondrial membrane depolarization, residual proteasome and mitochondrial enzymatic activities, colony-forming capabilities, and changes in protein expression profiles in A549 and H1299 cells under a combined therapeutic regimen gave results consistent with our hypothesis. 相似文献
