Negative-pressure wound therapy (NPWT) also known as V.A.C.?(Vacuum-assisted closure), is widely used to manage various type of wounds and accelerate healing. NPWT has so far been delivered mainly via open-cell polyurethane (PU) foam or medical gauze. In this study an experimental setup of sheep wound model was used to evaluate, under NPWT conditions, the performance of a silicone-coated non-woven polyester (N-WPE) compared with PU foam and cotton hydrophilic gauze, used as reference materials. Animals were anesthetized with spontaneous breathing to create three 3?×?3?cm skin defects bilaterally; each animal received three different samples on each side (n?=?6 in each experimental group) and was subjected to negative and continuous 125?mmHg pressure up to 16?days. Wound conditions after 1, 8 and 16?days of treatment with the wound dressings were evaluated based on gross and histological appearances. Skin defects treated with the silicone-coated N-WPE showed a significant decrease in wound size, an increase of re-epithelialization, collagen deposition and wound neovascularisation, and a minimal stickiness to the wound tissue, in comparison with gauze and PU foam. Taken all together these findings indicate that the silicone-coated N-WPE dressing enhances wound healing since stimulates higher granulation tissue formation and causes minor tissue trauma during dressing changes.  相似文献

The adhesive properties of the blends of isotactic polypropylene with ethylene-acrylic acid copolymers were investigated. It was found that the surface free energy, polar component of surface free energy, polar fraction and mechanical work of adhesion increased with concentration of EAA copolymers in the iPP blends to a degree exceeding additive function. The proportionality of mechanical work of adhesion of the polymer blends to polar fraction that reflects the strength of adhesive joint was also revealed. This observation permits us to claim that the increase in polarity or hydrophilicity of iPP due to modification in bulk by the use of polar EAA copolymer raises the strength of adhesive joints between iPP and more polar polymers. The relationship between mechanical properties of the studied polymer blends and their composition showed the functional additivity of the mechanical parameters.  相似文献

Traumatic brain injury (TBI) triggers both central and peripheral inflammatory responses. Existing pharmacological drugs are unable to effectively and quickly target the brain inflamed regions, setting up a major roadblock towards effective brain trauma treatments. Nanoparticles (NPs) have been used in multiple diseases as drug delivery tools with remarkable success due to their rapid diffusion and specificity in the target organ. Here, leukocyte-based biomimetic NPs are fabricated as a theranostic tool to directly access inflamed regions in a TBI mouse model. This NP systemic delivery is visualized using advanced in vivo imaging techniques, including intravital microscopy and in vivo imaging system. The results demonstrate selective targeting of NPs to the injured brain and increased NPs accumulation among the peripheral organs 24 h after TBI. Interestingly, increased microglial proliferation, decreased macrophage infiltration, and reduced brain lesion following the NPs treatments compared to sham vehicle-treated mice are also found. In summary, the results suggest that NPs represent a promising future theranostic tool for TBI treatment.  相似文献

A technique based on liquid chromatography has been developed to facilitate studies of enantioselectivity in penicillin G acylase (PGA)-catalyzed hydrolysis of some 2-aryloxyalkanoic acid methyl esters and isosteric analogues. PGA was covalently immobilized on an aminopropyl monolithic silica support to create an immobilized HPLC-enzyme reactor. Two sets of experimental data were drawn to calculate the enantioselectivity (E) of the kinetically controlled enantiomer-differentiating reaction, the degree of substrate conversion and the enantiomeric excess of the product. The developed enzymatic reactor was coupled through a switching valve to an achiral analytical column for separation and quantitation of the hydrolysis products. The enantiomeric excess was determined off-line on a PGA-chiral stationary phase. In this way, highly precise E values were determined. A computational study related to the hydrolysis of the considered racemic esters was also carried out in order to unambiguously clarify both the substrate specificity and the enantioselectivity displayed by PGA.  相似文献

This work is a contribution towards the understanding of certain features of mathematical models of single neurons. Emphasis is set on neuronal firing, for which the first passage time (FPT) problem bears a fundamental relevance. We focus the attention on modeling the change of the neuron membrane potential between two consecutive spikes by Gaussian stochastic processes, both of Markov and of non-Markov types. Methods to solve the FPT problems, both of a theoretical and of a computational nature, are sketched, including the case of random initial values. Significant similarities or diversities between computational and theoretical results are pointed out, disclosing the role played by the correlation time that has been used to characterize the neuronal activity. It is highlighted that any conclusion on this matter is strongly model-dependent. In conclusion, an outline of the asymptotic behavior of FPT densities is provided, which is particularly useful to discuss neuronal firing under certain slow activity conditions.  相似文献

The generation of 3D networks of primary neurons is a big challenge in neuroscience. Here, a novel method is presented for a 3D neuronal culture on superhydrophobic (SH) substrates. How nano‐patterned SH devices stimulate neurons to build 3D networks is investigated. Scanning electron microscopy and confocal imaging show that soon after plating neurites adhere to the nanopatterned pillar sidewalls and they are subsequently pulled between pillars in a suspended position. These neurons display an enhanced survival rate compared to standard cultures and develop mature networks with physiological excitability. These findings underline the importance of using nanostructured SH surfaces for directing 3D neuronal growth, as well as for the design of biomaterials for neuronal regeneration.  相似文献

Colloidal organic matter (COM) collected in small and large watercourse tributaries of the Negro River (Brazil) were fractionated and characterized by an asymmetrical flow field-flow fractionation (AFlFFF) coupled to UV and seven angle laser light scattering (LS) detectors, and by size exclusion chromatography (SEC) coupled with a UV detection. Number and weight average molar masses, weight average gyration radiuses RGw and polydispersity indexes were obtained for each sample in two separate runs under conditions optimized for lower and larger size fractions. The results demonstrate the existence of a decrease of size of the colloidal matter when passing from first order streams to higher order rivers. No significant changes were found in size distributions of samples collected during the low and high flow stages at the same site. The influence of selected pre-treatments such as filtration and reverse osmosis pre-concentration on the size and molar mass distributions was also studied.  相似文献

In recent years carbon monoxide (CO) has been employed by the fishery industry to preserve the fresh appearance of fish products during frozen storage, particularly in vacuum packaged products. CO reacts with oxy-myoglobin to form a stable cherry coloured carboxy-myoglobin complex. We propose a method that allows the quantitative determination of CO in the meat drip of tuna fish by UV–Vis spectroscopy. It consists of: (i) preparations of CO secondary standard solutions titrated against horse heart Mb primary standard solutions; (ii) determination of calibration curves to measure CO concentration in treated and untreated samples. The accuracy of the spectrophotometric method has been evaluated in terms of trueness and precision by using fortified tuna fish samples. The spectroscopic results have been compared with those obtained using a head space gas chromatographic technique (HS-GC–MS). The CO levels measured in tuna fish samples by UV–Vis are substantially lower than those revealed by HS-GC–MS. The origin of this discrepancy is discussed.  相似文献

The aim of this study was to assess the performance of the Diatheva STEC FLUO and BAX System real-time PCR assays for detection of Shiga toxin-producing Escherichia coli (STEC) (stx1/stx2 and eae target genes) and O-group identification in ground beef and bean sprout samples. Ground beef (325 g or 25 g) and mung bean sprout (25 g) samples were inoculated with ~?10 CFU of the “top five” STEC (O157:H7, O26, O103, O111, and O145 as specified in EU regulation ISO13136:2012), enriched using different broths and incubation temperatures, and tested using the Diatheva and BAX real-time PCR assays. In ground beef, both molecular methods were able to detect the “top five” STEC, and lower Ct values were observed for the Diatheva kits compared to BAX System. The O111-contaminated samples gave negative results with both methods using mTSB?+?novobiocin for enrichment. In bean sprouts, both methods provided positive results, although detection was not possible using mTSB?+?acriflavin/cefsulodin/vancomycin for enrichment. In conclusion, the Diatheva and BAX methods detected the “top five” STEC in ground beef and bean sprouts when inoculated at low levels. Both assays provided equivalent results in terms of performance and reliability. Thus, the Diatheva kits are comparable to reference STEC-detection methods and could be used by the food industry to reliably detect the “top five” STEC.  相似文献
