We describe quartz crystal electroacoustic admittance studies in thickness shear mode resonators loaded with self-assembled multilayers composed of alternate layers of glucose oxidase (GOx) and poly(allylamine) covalently attached to [Os(bpy)2ClPyCOH]- , (PAH-Os), deposited on a 3-mercaptopropanesulfonic acid (MPS)-modified gold on the quartz crystal. The complex acoustic impedance parameters, R(S) and X(LS) of a lumped-element Butterworth-Van Dyke (BVD) resonator have been determined for organized thin films of different thickness obtained by varying the number of enzyme layers, n, in (PAH-Os)n(GOx)n structures. The ellipsometric film thickness and mass for dry enzyme multilayer films and films in contact with water were evaluated, and the average film density was estimated. By combination of the estimated film thickness and density, the expression for the surface mechanical impedance of the lumped-element modified resonator (Granstaff and Martin model), and the liquid density and viscosity, we simulate the layer-by-layer film growth on the basis of the measured electroacoustic impedance. The complex impedance X(LS) and R(S) increase with film thickness and the enzyme films can be regarded as acoustically thin in the reduced state for films thinner than 600 nm. We have also measured electroacoustic parameters for PAH-Os/GOx self-assembled multilayers under electrochemical perturbation in a buffer electrolytic solution. The electrostatically self-assembled multilayers behaved as lossy viscoelastic films at 10 MHz with G'f and G'f on the order of 10(6) Pa. The films became viscoelastic upon oxidation to Os(III), resulting in an increase of R(S) and X(LS) in the oxidized state with the number of (PAH-Os)(GOx) bilayers due to film swelling and an increase in the shear moduli during oxidation.  相似文献

Cutting single-walled carbon nanotubes   总被引:3,自引:0,他引:3  
A two-step process is utilized for cutting single-walled carbon nanotubes (SWNTs). The first step requires the breakage of carbon-carbon bonds in the lattice while the second step is aimed at etching at these damage sites to create short, cut nanotubes. To achieve monodisperse lengths from any cutting strategy requires control of both steps. Room-temperature piranha and ammonium persulfate solutions have shown the ability to exploit the damage sites and etch SWNTs in a controlled manner. Despite the aggressive nature of these oxidizing solutions, the etch rate for SWNTs is relatively slow and almost no new sidewall damage is introduced. Carbon-carbon bond breakage can be introduced through fluorination to ~C(2)F, and subsequent etching using piranha solutions has been shown to be very effective in cutting nanotubes. The final average length of the nanotubes is approximately?100?nm with carbon yields as high as 70-80%.  相似文献

Assessed the validity of an initial intake interview with 12 couples seeking marital therapy (dissatisfied) and 16 couples seeking marital enhancement (satisfied). Ss (mean age 26.07 yrs) were interviewed with spouses together or separated. Interview items focused on marital satisfaction and perception, demographic variables, and comfort in making assertive responses. Couples were then administered several marital satisfaction and assertion questionnaires (e.g., Marital Adjustment Scale, Dyadic Adjustment Scale). Each couple also participated in a structured analog communication assessment in which they discussed a problem area in their marriage. The verbal interchanges during the communication exercises were coded by trained observers. Results suggest (a) a high degree of discriminant validity for the marital interview, (b) higher interspouse correlations for interview items during joint interviews, (c) higher indices of criterion-related validity for separate interviews, and (d) a significant relationship between the reactivity of joint interviews and the social sensitivity of the interview item. The organic electrochemical transistor (OECT) with a conjugated polymer as the active material is the elementary unit of organic bioelectronic devices. Improved functionalities, such as low power consumption, can be achieved by building complementary circuits featuring two or more OECTs. Complementary circuits commonly combine both p- and n-type transistors to reduce power draw. While p-type OECTs are readily available, n-type OECTs are less common mainly due to poor stability of the n-type active channel material in aqueous electrolyte. Here, a complementary circuit is made using a pair of OECTs having polyaniline (PANI) as the channel material in both transistors. PANI, with a finite electrochemical window accessible at voltages lower than 1 V, exhibits a peak in current versus gate voltage when used as an active channel in an OECT. The current peak has two slopes, one n-like and one p-like, which correspond to different electrochemical regimes of the same underlying conjugated polymer. The electrochemistry enables the design of a complementary circuit using only PANI as the channel material. The PANI-based circuit is shown to have excellent performance with gain of ≈7 and is transferred on a flexible biocompatible chitosan substrate with demonstrated operation in aqueous electrolyte.  相似文献

Bacteria often reside in communities where the cells have secreted sticky, polymeric compounds that allow them to attach to surfaces. This sessile lifestyle, referred to as a biofilm, affords the cells within these communities a tolerance of antibiotics and antimicrobial treatments. Biofilms of the bacterium Pseudomonas aeruginosa have been implicated in cystic fibrosis and are capable of colonizing medical implant devices, such as heart valves and catheters, where treatment of the infection often requires the removal of the infected device. This mode of growth is in stark contrast to planktonic, free floating cells, which are more easily eradicated with antibiotics. The mechanisms contributing to a biofilm's tenacity and a planktonic cell's susceptibility are just beginning to be explored. In this study, we have used a metabolomic approach employing nuclear magnetic resonance (NMR) techniques to study the metabolic distinctions between these two modes of growth in P. aeruginosa. One-dimensional 1H NMR spectra of fresh growth medium were compared with spent medium supernatants from batch and chemostat planktonic and biofilms generated in continual flow system culture. In addition, 1H high-resolution magic angle spinning NMR techniques were employed to collect 1H NMR spectra of the corresponding cells. Principal component analysis and spectral comparisons revealed that the overall metabolism of planktonic and biofilm modes of growth appeared similar for the spent media, while the planktonic and biofilm cells displayed marked differences. To determine the robustness of this technique, we prepared cell samples under slightly different preparation methods. Both techniques showed similar results. These feasibility studies show that there exist chemical differences between planktonic and biofilm cells; however, in order to identify these metabolomic differences, more extensive studies would have to be performed, including 1H-1H total correlated spectroscopy.  相似文献

A method is described for identifying bioactive compounds in complex mixtures based on the use of capillary-scale monolithic enzyme-reactor columns for rapid screening of enzyme activity. A two-channel nanoLC system was used to continuously infuse substrate coupled with automated injections of substrate/small molecule mixtures, optionally containing the chromogenic Ellman reagent, through sol-gel derived acetylcholinesterase (AChE) doped monolithic columns. This is the first report of AChE encapsulated in monolithic silica for use as an immobilized enzyme reactor (IMER), and the first use of such IMERs for mixture screening. AChE IMER columns were optimized to allow rapid functional screening of compound mixtures based on changes in the product absorbance or the ratio of mass spectrometric peaks for product and substrate ions in the eluent. The assay had robust performance and produced a Z' factor of 0.77 in the presence of 2% (v/v) DMSO. A series of 52 mixtures consisting of 1040 compounds from the Canadian Compound Collection of bioactives was screened and two known inhibitors, physostigmine and 9-aminoacridine, were identified from active mixtures by manual deconvolution. The activity of the compounds was confirmed using the enzyme reactor format, which allowed determination of both IC(50) and K(I) values. Screening results were found to correlate well with a recently published fluorescence-based microarray screening assay for AChE inhibitors.  相似文献

Previous evidence has suggested that analogical reasoning (recognizing similarities among object relations when the objects themselves are dissimilar) is limited to humans and apes. This study investigated whether capuchin monkeys (Cebus apella) can use analogical reasoning to solve a 3-dimensional search task. The task involved hiding a food item under 1 of 2 or 3 plastic cups of different sizes and then allowing subjects to search for food hidden under the cup of analogous size in their own set of cups. Four monkeys were exposed to a series of relational matching tasks. If subjects reached criterion on these tasks, they were exposed to relational transfer tasks involving novel stimuli. Three of the monkeys failed to reach criterion on the basic relational matching tasks and therefore were not tested further. One monkey, however, revealed above-chance performance on a series of transfer tasks with 3 novel stimuli. This evidence suggests that contrary to previous arguments, a member of a New World monkey species can solve an analogical problem.
