The presented method for the determination of a number of phthalic acid esters into oily media is based on their extraction by acetonitrile with precipitation under freezing and subsequent solid-phase extraction using alumina. This technique was checked as the best procedure to separate lipophilic phthalates from fatty media achieving elimination of contamination by excess use of solvents. Extracted phthalates are determined in acetonitrile solutions with gas chromatography in conjunction to isotopic dilution mass spectrometry using D4 isotopic labeled derivatives. Full validation of the method was carried out and background effect was examined. Method calibration for extraction from oil resulted in limit of detection (LOD) of 7–10 ng g?1 and limit of quantification (LOQ) of 20–50 ng g?1 for DMP, DEP, DiBP, DBP, DEHP, and BBP and in LOD 500 ng g?1 and LOQ of 1,500 ng g?1 for DiNP and DiDP. Real samples of virgin olive oil from olive press establishments from three different areas were analyzed and the phthalates content was below detection limit for all phthalic esters under consideration for nonindustrial areas and in measurable concentrations for industrial areas specifically with regard to DiBP. 相似文献

Background
Although several epidemiological studies have examined the mortality among users of spit tobacco, none have compared mortality of former cigarette smokers who substitute spit tobacco for cigarette smoking (“switchers”) and smokers who quit using tobacco entirely.Methods
A cohort of 116 395 men were identified as switchers (n = 4443) or cigarette smokers who quit using tobacco entirely (n = 111 952) when enrolled in the ongoing US American Cancer Society Cancer Prevention Study II. From 1982 to 31 December 2002, 44 374 of these men died. The mortality hazard ratios (HR) of tobacco‐related diseases, including lung cancer, coronary heart disease, stroke and chronic obstructive pulmonary disease, were estimated using Cox proportional hazards regression modelling adjusted for age and other demographic variables, as well as variables associated with smoking history, including number of years smoked, number of cigarettes smoked and age at quitting.Results
After 20 years of follow‐up, switchers had a higher rate of death from any cause (HR 1.08, 95% confidence interval (CI) 1.01 to 1.15), lung cancer (HR 1.46, 95% CI 1.24 to 1.73), coronary heart disease (HR 1.13, 95% CI 1.00 to 1.29) and stroke (HR 1.24, 95% CI 1.01 to 1.53) than those who quit using tobacco entirely.Conclusion
The risks of dying from major tobacco‐related diseases were higher among former cigarette smokers who switched to spit tobacco after they stopped smoking than among those who quit using tobacco entirely.Several epidemiological studies have examined morbidity and mortality among users of spit tobacco (spit‐tobacco users),1,2 but none have compared the mortality of former cigarette smokers who substitute spit tobacco for cigarette smoking (“switchers”) to those of smokers who quit using tobacco entirely. Microbial, physical and structural changes in high pressured wheat dough were studied as a function of pressure level (50-250 MPa) and holding time (1-4 min). Thereafter, selected conditions of high hydrostatic processing (HPP) were applied to bread dough and the technological quality of the obtained breads was studied. The effect of HPP on wheat dough was investigated by determining microbial population (total aerobic mesophilic bacteria, moulds and yeasts), color and mechanical and texture surface related dough parameters (cohesiveness, adhesiveness, hardness and stickiness). HPP reduced the endogenous microbial population of wheat dough from 104 colony forming units/g (CFU) to levels of 102 CFU. HPP treatment significantly (P < 0.05) increased dough hardness and adhesiveness, whereas treatment time reduced its stickiness. Scanning electron micrographs suggested that proteins were affected when subjected to pressure levels higher than 50 MPa, but starch modification required higher pressure levels. HPP treated yeasted doughs led to wheat breads with different appearance and technological characteristics; crumb acquired brownish color and heterogeneous cell gas distribution with increased hardness due to new crumb structure. This study suggests that high hydrostatic processing in the range 50-200 MPa could be an alternative technique for obtaining novel textured cereal based products. 相似文献

Abstract: The natural mixed culture kefir was immobilized on boiled corn grains to produce an efficient biocatalyst for lactic acid fermentation with direct applications in food production, such as sourdough bread making. The immobilized biocatalyst was initially evaluated for its efficiency for lactic acid production by fermentation of cheese whey at various temperatures. The immobilized cells increased the fermentation rate and enhanced lactic acid production compared to free kefir cells. Maximum lactic acid yield (68.8 g/100 g) and lactic acid productivity (12.6 g/L per day) were obtained during fermentation by immobilized cells at 37 °C. The immobilized biocatalyst was then assessed as culture for sourdough bread making. The produced sourdough breads had satisfactory specific loaf volumes and good sensory characteristics. Specifically, bread made by addition of 60% w/w sourdough containing kefir immobilized on corn was more resistant regarding mould spoilage (appearance during the 11th day), probably due to higher lactic acid produced (2.86 g/Kg of bread) compared to the control samples. The sourdough breads made with the immobilized biocatalyst had aroma profiles similar to that of the control samples as shown by headspace SPME GC‐MS analysis. 相似文献

Repetitive transcranial magnetic stimulation (rTMS) is a non-invasive brain stimulation technique that has the potential to treat a variety of neurologic and psychiatric disorders. The extent of rTMS-induced neuroplasticity may be dependent on a subject’s brain state at the time of stimulation. Chronic low intensity rTMS (LI-rTMS) has previously been shown to induce beneficial structural and functional reorganisation within the abnormal visual circuits of ephrin-A2A5-/- mice in ambient lighting. Here, we administered chronic LI-rTMS in adult ephrin-A2A5-/- mice either in a dark environment or concurrently with voluntary locomotion. One day after the last stimulation session, optokinetic responses were assessed and fluorescent tracers were injected to map corticotectal and geniculocortical projections. We found that LI-rTMS in either treatment condition refined the geniculocortical map. Corticotectal projections were improved in locomotion+LI-rTMS subjects, but not in dark + LI-rTMS and sham groups. Visuomotor behaviour was not improved in any condition. Our results suggest that the beneficial reorganisation of abnormal visual circuits by rTMS can be significantly influenced by simultaneous, ambient visual input and is enhanced by concomitant physical exercise. Furthermore, the observed pathway-specific effects suggest that regional molecular changes and/or the relative proximity of terminals to the induced electric fields influence the outcomes of LI-rTMS on abnormal circuitry. 相似文献

Non-alcoholic fatty liver disease (NAFLD) is the most prevalent form of chronic liver disease worldwide, but a reliable non-invasive method to quantify liver steatosis in primary healthcare is not available. Circulating microRNAs have been proposed as biomarkers of severe/advanced NAFLD (steatohepatitis and fibrosis). However, the use of circulating miRNAs to quantitatively assess the % of liver fat in suspected NAFLD patients has not been investigated. We performed global miRNA sequencing in two sets of samples: human livers from organ donors (n = 20), and human sera from biopsy-proven NAFLD patients (n = 23), both with a wide range of steatosis quantified in their liver biopsies. Partial least squares (PLS) regression combined with recursive feature elimination (RFE) was used to select miRNAs associated with steatosis. Moreover, regression models with only 2 or 3 miRNAs, with high biological relevance, were built. Comprehensive microRNA sequencing of liver and serum samples resulted in two sets of abundantly expressed miRNAs (418 in liver and 351 in serum). Pearson correlation analyses indicated that 18% of miRNAs in liver and 14.5% in serum were significantly associated with the amount of liver fat. PLS-RFE models demonstrated that 50 was the number of miRNAs providing the lowest error in both liver and serum models predicting steatosis. Comparison of the two miRNA subsets showed 19 coincident miRNAs that were ranked according to biological significance (guide/passenger strand, relative abundance in liver and serum, number of predicted lipid metabolism target genes, correlation significance, etc.). Among them, miR-10a-5p, miR-98-5p, miR-19a-3p, miR-30e-5p, miR-32-5p and miR-145-5p showed the highest biological relevance. The stability and functionality of GCC-bOBP, a monomeric triple mutant of bovine odorant binding protein, was investigated, in the presence of denaturant and in acidic pH conditions, by both protein and 1-aminoanthracene ligand fluorescence measurements, and compared to that of both bovine and porcine wild type homologues. Complete reversibility of unfolding was observed, though refolding was characterized by hysteresis. Costa Rica is not an exception in the prevalence of ochratoxin A in human plasma, in this research the presence of the micotoxin was found in 95% of the 149 samples studied. The presence of ocratoxina A also was studied in 110 samples of toasted and grounded coffee from the most important 12 coffee factories of the country and from 7 supermarkets. With the exception of one negative sample the rest of them have concentrations of micotoxin below 4000 ng/kg. An association between the coffee consumption and the presence of ochratoxin A in plasma was attempted to be found as well as in the consumption of beer, but there were any statistically significant difference in the average level of mycotoxin between the coffee consumers and non coffee consumers neither between beer consumers and no beer consumers. 相似文献
