Gluten films obtained in acid conditions display some protein dispersion difficulties. Ultrasound treatment (UT) could represent an interesting strategy for improving gluten film appearance. Different UT exposure times were applied to film-forming dispersion. The aim of this work was to investigate the effect of ultrasound treatment on gluten-based films at molecular and mesoscopic levels. Distribution in gliadin and glutenins was determined with SE-HPLC. The UT improved protein dispersion and final film appearance. Sonication did not lead to large changes in various gluten fractions, which suggests absence of important protein breakdown at the molecular level. Gluten showed high tolerance to UT. Surface properties of untreated and treated films were investigated by contact angle measurement: sonication promoted hydrophilic surface properties.Industrial relevanceBio-based packaging has been receiving increasing attention in view of its beneficial impact on the environment. Among proteins, gluten resulted as a very interesting film-forming material. Gluten films prepared in acid conditions showed problems in protein dispersion. The behavior of fluoroquinolone antibacterial agents (FQs) during mechanical-biological wastewater treatment was studied by mass flow analysis. In addition, the fate of FQs in agricultural soils after sludge application was investigated. Concentrations of FQs in filtered wastewater (raw sewage, primary, secondary, and tertiary effluents) were determined using solid-phase extraction with mixed phase cation exchange disk cartridges and reversed-phase liquid chromatography with fluorescence detection. FQs in suspended solids, sewage sludge (raw, excess, and anaerobically digested sludge), and sludge-treated soils were determined as described for the aqueous samples but preceded by accelerated solvent extraction. Wastewater treatment resulted in a reduction of the FQ mass flow of 88-92%, mainly due to sorption on sewage sludge. A sludge-wastewater partition coefficient (log Kd approximately 4) was calculated in the activated sludge reactors with a hydraulic residence time of about 8 h. No significant removal of FQs occurred under methanogenic conditions of the sludge digesters. These results suggest sewage sludge as the main reservoir of FQ residues and outline the importance of sludge management strategies to determine whether most of the human-excreted FQs enter the environment. Field experiments of sludge-application to agricultural land confirmed the long-term persistence of trace amounts of FQs in sludge-treated soils and indicated a limited mobility of FQs into the subsoil. 相似文献

Post rigor bovine M. semimembranosus was analysed by differential scanning calorimetry (d.s.c.). After extractive removal of sarcoplasmic proteins, subsequent pH adjustment and manual connective tissue removal, d.s.c. yielded reproducible thermograms which permitted investigation of the individual major myofibrillar proteins in various pH and salt environments without prior isolation. The positions of two major peaks, interpreted as myosin transitions, proved to be strongly pH dependent. At pH 5.4, the peak maxima occurred at 58 and 65°C, respectively, at a heating rate of 10°C min-1. Above pH 6.5 their order of denaturation was reversed. In the pH range 5.4–6.5 the peak ascribed to actin had its maximum near 80°C in intact muscle. Above this pH range it was displaced to lower temperature. The thermal stability of actin was studied after treatment of the muscle tissue with different salt solutions. At equal ionic strengths (μ = 0.15) at pH 5.5, calcium chloride and sodium chloride caused 6.5°C and 4°C displacement to lower temperature, respectively. The thermograms of bovine semimembranosus muscle were compared to those of two red and two white muscle types (bovine cardiac and rabbit soleus muscles, chicken breast and rabbit semimembranosus muscles, respectively) at two pH levels. Greater myosin differences were found between red and white muscles than between muscles from different animal species. All muscles gave similar actin transitions, with exception of the heart muscle where the actin peak appeared at 3 °C lower temperature. Staphylococcus aureus is a bacterial pathogen considered a principal etiological agent of food poisoning. The aim of this study was to develop
and evaluate a rapid and sensitive method for the detection of S. aureus in food by using selective enrichment and a new species-specific real-time polymerase chain reaction (PCR). Specific primers
and a TaqMan probe targeted to specific S. aureus gene encoding for acriflavine resistance protein were designed. The real-time PCR was highly specific for S. aureus with 100% inclusivity and 100% exclusivity determined using 83 S. aureus strains and 64 non-S.-aureus strains. PCR detection limit of 6.8 × 101 and 3.4 × 101 CFU ml−1 were obtained with 100% and 70% detection probability, respectively. The single selective enrichment based on the study of
different enrichment conditions was selected and a lysis by boiling was used to obtain bacterial DNA. Out of 112 food samples
analyzed, 61 were positive by the PCR-based method and 53 by the standard method. Out of ten food matrices artificially contaminated
at a level of 10° CFU g−1, ten and six were positive by the respective methods. Moreover, 10° CFU 10 g−1 was detected in all ten artificially contaminated samples after a large-scale enrichment using PCR-based detection, in contrast
to seven false negative by standard detection. The developed method facilitated the detection of S. aureus on the next day after the sample reception. A rapid ultra-performance liquid chromatographic (UPLC) method for the determination of biogenic amines (putrescine, cadaverine, spermidine, spermine, phenylethylamine, histamine, tyramine and tryptamine) in selected food samples is described. The eight biogenic amines, which are the most important to be determined in food samples, were derivatized with dansyl chloride prior to UPLC separation. The dansylated amines were separated on an Agilent Zorbax Eclipse XDB – C18 column (50 × 4.6 mm ID, 1.8 μm) using gradient elution with a binary system of acetonitrile–water, a flow rate of 1.0 ml/min and UV detection at 225 nm. The analysis is very fast, all amines are well resolved and are eluted from the column in less than 6 min. The average repeatability of the method ranged between 1.02% and 2.14%. Limits of detection (LODs) for considered amines ranged between 0.032 and 0.098 μg/l; calibration curves showed very good linearity (r = 0.9994–1.0000). Enterocin AS-48 was tested in apple juice against the cider-spoilage, exopolysaccharide-producing strain Lactobacillus diolivorans 29 in combination with high-intensity pulsed-electric field (HIPEF) treatment (35 kV/cm, 150 Hz, 4 μs and bipolar mode). A response surface methodology was applied to study the bactericidal effects of the combined treatment, with AS-48 concentration and HIPEF treatment time as process variables. At subinhibitory bacteriocin concentrations, microbial inactivation by the combined treatment increased as the bacteriocin concentration and the HIPEF treatment time increased (from 0.5 to 2.0 μg/ml and from 100 to 1000 μs, respectively). Highest inactivation (4.87 logs) was achieved by 1000 μs HIPEF treatment in combination with 2.0 μg/ml AS-48. While application of treatments separately did not protect juice from survivors during storage, survivors to the combined treatment were inactivated within the following 24 h of storage, and the treated samples remained free from detectable lactobacilli for at least 15 days at temperatures of 4 °C as well as 22 °C. The combined treatment could be useful for inactivation of exopolysaccharide-producing L. diolivorans in apple juice. 相似文献

Describes the testing of an active-alert induction procedure which sufficiently controlled conditions to permit a comparison between the alterations produced by that procedure and those produced by the traditional relaxation induction technique. Ss were 50 university students. In the active-alert induction the S rode a bicycle ergometer under load, keeping eyes open while exercising and receiving suggestions of alertness. The alternate form, used in random alternation with the same Ss, consisted of the standard eye-fixation and relaxation induction of the Stanford Hypnotic Susceptibility Scale, Form B (SHSS-B). Within each session on 2 days during which each S served, the induction procedure was followed by 8 tests of responses to suggestion, from the SHSS-A and SHSS-B. The mean measured hypnotic responsiveness was independent of the type of induction. The active-alert condition was characterized by an acceleration of the rate of pedaling for the more responsive Ss. BACKGROUND: Cynara cardunculus is a species native to the Mediterranean basin. It includes globe artichoke and cultivated cardoon as well as their progenitor wild cardoon. The species is a source of biophenols, and its leaf extracts have been widely used in herbal medicine as hepatoprotectors and choleretics since ancient times. The aim of this study was to determine the effect of increasing the level of salinity in the nutrient solution (1 or 30 mmol L?1 NaCl) on biomass production, mineral composition, radical‐scavenging activity, caffeoylquinic acids and flavonoids in three artichoke (‘Romolo’, ‘Violetto di Provenza’ and ‘Violetto di Romagna’) and three cultivated cardoon (‘Bianco Avorio’, ‘Bianco Gigante Inerme’ and ‘Gigante di Romagna’) cultivars grown in a floating system. RESULTS: Increased salinity in the nutrient solution decreased the leaf dry biomass and leaf number of artichoke and cultivated cardoon cultivars. Salinity reduced macro‐ and microelement accumulation in leaves (e.g. N, K, Ca, Mg, Fe, Mn and B) but improved their antioxidant activity, total polyphenols, chlorogenic acid, cynarin and luteolin. The cultivated cardoons, especially ‘Bianco Avorio’ and ‘Gigante di Romagna’, showed higher biomass and leaf number than those observed in artichoke genotypes. ‘Violetto di Provenza’ exhibited the highest content of chlorogenic acid, closely followed by ‘Violetto di Romagna’, whereas for cynarin content the highest values were recorded in ‘Violetto di Provenza’, ‘Bianco Avorio’ and ‘Gigante di Romagna’. The highest content of luteolin was recorded in ‘Gigante di Romagna’ and ‘Bianco Avorio’, while the highest content of apigenin was observed in ‘Gigante di Romagna’. CONCLUSION: The results showed that the floating system could be considered an effective tool to improve quality aspects through proper management of the salt concentration in the nutrient solution. Yacon roots are a promising source of inulintype fructans (35 g/100 g dry matter) with a total amount of dietary fiber of about 45 g/100 g dry matter. The polydispersity of the particle size distribution of yacon aqueous suspension was reduced with increasing the degree of homogenization due to two phenomena: aggregation of particles, and disruption of large particles. Nonhomogenized yacon suspensions exhibited large cell clusters that were disrupted into small cell clusters, single cells, and aggregates. The most concentrated suspension exhibited mainly aggregates.The volume fraction of the suspensions decreased significantly with increasing the degree of homogenization. This was attributed to a denser packing of small particles and aggregates in between large particles thus allowing the water within the structure to be released.Yacon suspensions exhibited high elastic modulus (750 Pa) at low water insoluble solids content (0.9% WIS). Additional results suggested that inulintype fructans contributed to the elastic properties of yacon suspensions.The yacon suspensions studied in this work can be considered as semiconcentrated suspensions, i.e. the plot elastic modulus of the suspensions versus concentration exhibited a region immediately after the transition region. Particle interactions seem to be of great importance in this region in the particular case of yacon suspension since the elastic modulus (G′) reached 750 Pa at low water insoluble solids content (< 1%). 相似文献
