Background Continuous arterial spin labeling (CASL) is a non-invasive technique for the measurement of cerebral blood flow (CBF). The aim of the present study was to examine the reproducibility of CASL measurements and its suitability to consistently detect differences between groups, regions, and resting states. Materials and methods Thirty-eight healthy subjects (19 female) were examined at 1.5 T on two measurement occasions that were seven weeks apart. Resting CBF was measured with eyes open and eyes closed. Results In different regions of interest (ROIs) the repeatability estimates varied between 9 and 19 ml/100 g/min. There were no significant mean differences between occasions in all ROIs (P > 0.05). Greater CBF in the eyes-open than in the eyes-closed state was consistently present in the primary and secondary visual areas. Furthermore, CBF was consistently greater in the right than in the left hemisphere (P < 0.05) and differed between lobes and between arterial territories (P < 0.001). Finally, we consistently observed greater CBF in women than in men (P < 0.001). Conclusion This study demonstrates the suitability of CASL to consistently detect differences between groups, regions, and resting states even after seven weeks. This emphasizes its usefulness for longitudinal designs.  相似文献

An anaerobic enrichment culture reductively dehalogenated 1,2,4- and 1,2,3-trichlorodibenzo-p-dioxin (TrCDD) almost exclusively at peripheral positions forming the main products 1,3-dichloro-(DiCDD) and 2-monochlorodibenzo-p-dioxin (MCDD) from 1,2,4-TrCDD and 2,3-DiCDD from 1,2,3-TrCDD. Dehalococcoides was monitored in the mixed culture by quantitative real-time PCR. A yield of 2.5 x 10(8) to 2.75 x 10(8) copies of 16S rRNA genes per micromole of chloride released suggested growth by dehalorespiration with dibenzo-p-dioxins. For the analysis of carbon isotope fractionation, the dioxin congeners were isolated by solid-phase microextraction (SPME) from the headspace of the cultures. The delta13C composition of 1,2,4-TrCDD did not change remarkably during the course of reductive dehalogenation; however, the intermediate 1,3-DiCDD became enriched, and the final product 2-MCDD significantly depleted in 13C with a discrimination of 2.5-3.6 per thousand between 1,3-DiCDD and 2-MCDD. 1,2,3-TrCDD and its main product 2,3-DiCDD became slightly enriched in 13C, whereas the formed low concentrations of 2-MCDD were depleted in 13C by 5.5-4.8 per thousand. This study demonstrates carbon isotope fractionation during sequential reductive dehalogenation of chlorinated dibenzo-p-dioxins, whereby isotope fractionation upon dehalogenation of the intermediate was substantial. This can provide a basis for the development of a new method to monitor the fate of dioxins in the environment using compound specific stable isotope analyses.  相似文献

The gene encoding the 16S rRNA of Enterobacter (E.) sakazakii (ATCC 29544, plus four strains isolated from powdered infant formula) was studied, and the sequence compared with those of other Enterobacteriaceae in aspects of genetic variability. Sequence differences between E. sakazakii and other Enterobacteriaceae within the hypervariable regions V1, V2, and V3, respectively, were used to develop two PCR methods for E. sakazakii. PCR1 employed a primer pair located in V1/V2, while PCR2 utilized a primer pair located in V1/V3, respectively. The two PCR methods were tested against a set of 57 E. sakazakii and 148 non-E. sakazakii isolates. PCR1 gave an amplicon with a size of 406 bp and resulted in 100% positive results for E. sakazakii, but also detected Citrobacter koseri/amalonaticus and all nine tested Salmonella enterica serovars. In contrast, PCR2 (amplicon size of 952 bp) gave positive results only for E. sakazakii, thus allowing specific identification of this species.  相似文献

While recent studies have shown that rotating a single radio-frequency (RF) coil during the acquisition of magnetic resonance (MR) images provides a number of hardware advantages (i.e., requires only one RF channel, avoids coil-coil coupling and facilitates large-scale multinuclear imaging), they did not describe in detail how to build a rotating RF coil system. This paper presents detailed engineering information on the electromechanical design and construction of a MR-compatible RRFC system for human head imaging at 2 T. A custom-made (bladeless) pneumatic Tesla turbine was used to rotate the RF coil at a constant velocity, while an infrared optical encoder measured the selected frequency of rotation. Once the rotating structure was mechanically balanced and the compressed air supply suitably regulated, the maximum frequency of rotation measured ~14.5 Hz with a 2.4% frequency variation over time. MR images of a water phantom and human head were obtained using the rotating RF head coil system.  相似文献

Objectives  To quantify the gain in time-series SNR that can be achieved in the amygdala by reducing EPI voxel size, and to assess the extent to which this advantage is carried through to statistical significance in a group fMRI study, using a cognitive task to trigger task-independent deactivation of anterior medial temporal structures. Materials and methods  Two groups of seven subjects were posed number-series tasks to induce deactivation of the Default Mode network. This is known from PET work to include the amygdala, which lies in a region of high magnetic field gradient. In 3 T imaging, one group was studied with high resolution EPI with 6 μl voxels, the other with lower resolution EPI with 17 μl voxels. Field maps were acquired to allow field gradients in relevant ROIs to be assessed. Results  Time-series SNR was 45% higher in the amygdala in the high resolution EPI data than in the low resolution data. In activation results, whilst there was good agreement between other areas, the involvement of the amygdala could only be demonstrated in the high resolution data. Conclusion  We find that reduction in signal dephasing afforded by high resolution EPI is realized as a substantial increase in SNR and BOLD sensitivity in group fMRI data. This has allowed the first demonstration of the involvement of the amygdala in the Default Mode in fMRI.  相似文献

Controlling structure and function to switch ionic transport through synthetic membranes is a major challenge in the fabrication of functional nanodevices. Here we describe the combination of mesoporous silica thin films as structural unit, functionalized with two different redox‐responsive ferrocene‐containing polymers, polyvinylferrocene (PVFc) and poly(2‐(methacryloyloxy)ethyl ferrocenecarboxylate) (PFcMA), by using either a grafting to, or a grafting from approach. Both mesoporous film functionalization strategies are investigated in terms of polymer effect on ionic permselectivity. A significantly different ionic permselective behavior can be observed. This is attributed to different polymer location within the mesoporous film, depending on the functionalization strategies used. Additionally, the influence of chemical oxidation on the ionic permselective behavior is studied by cyclic voltammetry showing a redox‐controlled membrane gating as function of polymer location and the pH value. This study is a first step of combining redox‐responsive ferrocene‐containing polymers and mesoporous membranes, and thus towards redox‐controlled ionic transport through nanopores.  相似文献

Sustainable bioenergy systems are, by definition, embedded in social, economic, and environmental contexts and depend on support of many stakeholders with different perspectives. The resulting complexity constitutes a major barrier to the implementation of bioenergy projects. The goal of this paper is to evaluate the potential of Multi Criteria Analysis (MCA) to facilitate the design and implementation of sustainable bioenergy projects. Four MCA tools (Super Decisions, DecideIT, Decision Lab, NAIADE) are reviewed for their suitability to assess sustainability of bioenergy systems with a special focus on multi-stakeholder inclusion. The MCA tools are applied using data from a multi-stakeholder bioenergy case study in Uganda. Although contributing to only a part of a comprehensive decision process, MCA can assist in overcoming implementation barriers by (i) structuring the problem, (ii) assisting in the identification of the least robust and/or most uncertain components in bioenergy systems and (iii) integrating stakeholders into the decision process. Applying the four MCA tools to a Ugandan case study resulted in a large variability in outcomes. However, social criteria were consistently identified by all tools as being decisive in making a bioelectricity project viable.  相似文献
