A two-dimensional pressure-temperature phase diagram was constructed for neon adsorbed on exfoliated graphite using the heat capacity data obtained in our laboratory for this system. The two-dimensional pressures at the triple and critical points were found to be t=64×10–6 N/m and c=128×10–6 N/m, respectively. From Clapeyron's equation and assuming an ideal behavior for the two-dimensional gas phase, the latent heat of sublimation was calculated as a function of temperature. The latent heat of vaporization was also calculated at the triple point and consequently the latent heat of fusion was found. The following values were obtained at the triple point:l s/k=86 K,l v/k=24 K, andl f/k=62 K.  相似文献

This article aims to tackle a practical three-dimensional packing problem, where a number of cartons of diverse sizes are to be packed into a bin with fixed length and width but open height. Each carton is allowed to be packed in any one of the six orientations, and the carton edges are parallel to the bin edges. The allowance of variable carton orientations exponentially increases the solution space and makes the problem very challenging to solve. This study first elaborately devises the packing procedure, which converts an arbitrary sequence of cartons into a compact packing solution and subsequently develops an improved genetic algorithm (IGA) to evolve a set of solutions. Moreover, a novel global search framework (GSF), utilizing the concept of evolutionary gradient, is proposed to further improve the solution quality. Numerical experiments indicate that IGA provides faster and better results and GSF demonstrates its superior performance, especially in solving relative large-size and heterogeneous instances. Applying the proposed algorithms to some benchmarking cases of the three-dimensional strip packing problem also indicates that the algorithms are robust and effective compared to existing methods in the literature.  相似文献

Lipase B from Candida antarctica was immobilized on heterofunctional support octyl agarose activated with vinyl sulfone to prevent enzyme release under drastic conditions. Covalent attachment was established, but the blocking step using hexylamine, ethylenediamine or the amino acids glycine (Gly) and aspartic acid (Asp) altered the results. The activities were lower than those observed using the octyl biocatalyst, except when using ethylenediamine as blocking reagent and p-nitrophenol butyrate (pNPB) as substrate. The enzyme stability increased using these new biocatalysts at pH 7 and 9 using all blocking agents (much more significantly at pH 9), while it decreased at pH 5 except when using Gly as blocking agent. The stress inactivation of the biocatalysts decreased the enzyme activity versus three different substrates (pNPB, S-methyl mandelate and triacetin) in a relatively similar fashion. The tryptophane (Trp) fluorescence spectra were different for the biocatalysts, suggesting different enzyme conformations. However, the fluorescence spectra changes during the inactivation were not too different except for the biocatalyst blocked with Asp, suggesting that, except for this biocatalyst, the inactivation pathways may not be so different.  相似文献

Ecto-nucleoside triphosphate diphosphohydrolases (NTPDases) are enzymes located on the surface of the T. cruzi plasma membrane, which hydrolyze a wide range of tri-/-diphosphate nucleosides. In this work, we used previously developed genetically modified strains of Trypanosoma cruzi (T. cruzi), hemi-knockout (KO +/−) and overexpressing (OE) the TcNTPDase-1 gene to evaluate the parasite infectivity profile in a mouse model of acute infection (n = 6 mice per group). Our results showed significantly higher parasitemia and mortality, and lower weight in animals infected with parasites OE TcNTPDase-1, as compared to the infection with the wild type (WT) parasites. On the other hand, animals infected with (KO +/−) parasites showed no mortality during the 30-day trial and mouse weight was more similar to the non-infected (NI) animals. In addition, they had low parasitemia (45.7 times lower) when compared with parasites overexpressing TcNTPDase-1 from the hemi-knockout (OE KO +/−) group. The hearts of animals infected with the OE KO +/− and OE parasites showed significantly larger regions of cardiac inflammation than those infected with the WT parasites (p < 0.001). Only animals infected with KO +/− did not show individual electrocardiographic changes during the period of experimentation. Together, our results expand the knowledge on the role of NTPDases in T. cruzi infectivity, reenforcing the potential of this enzyme as a chemotherapy target to treat Chagas disease (CD).  相似文献

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has been responsible for the severe pandemic of acute respiratory disease, coronavirus disease 2019 (COVID-19), experienced in the 21st century. The clinical manifestations range from mild symptoms to abnormal blood coagulation and severe respiratory failure. In severe cases, COVID-19 manifests as a thromboinflammatory disease. Damage to the vascular compartment caused by SARS-CoV-2 has been linked to thrombosis, triggered by an enhanced immune response. The molecular mechanisms underlying endothelial activation have not been fully elucidated. We aimed to identify the proteins correlated to the molecular response of human umbilical vein endothelial cells (HUVECs) after exposure to SARS-CoV-2, which might help to unravel the molecular mechanisms of endothelium activation in COVID-19. In this direction, we exposed HUVECs to SARS-CoV-2 and analyzed the expression of specific cellular receptors, and changes in the proteome of HUVECs at different time points. We identified that HUVECs exhibit non-productive infection without cytopathic effects, in addition to the lack of expression of specific cell receptors known to be essential for SARS-CoV-2 entry into cells. We highlighted the enrichment of the protein SUMOylation pathway and the increase in SUMO2, which was confirmed by orthogonal assays. In conclusion, proteomic analysis revealed that the exposure to SARS-CoV-2 induced oxidative stress and changes in protein abundance and pathways enrichment that resembled endothelial dysfunction.  相似文献

The circadian clock (CC) is a daily system that regulates the oscillations of physiological processes and can respond to the external environment in order to maintain internal homeostasis. For the functioning of the CC, the clock genes (CG) act in different metabolic pathways through the clock-controlled genes (CCG), providing cellular regulation. The CC’s interruption can result in the development of different diseases, such as neurodegenerative and metabolic disorders, as well as cancer. Leukemias correspond to a group of malignancies of the blood and bone marrow that occur when alterations in normal cellular regulatory processes cause the uncontrolled proliferation of hematopoietic stem cells. This review aimed to associate a deregulated CC with the manifestation of leukemia, looking for possible pathways involving CG and their possible role as leukemic biomarkers.  相似文献

Minimal Learning Machine (MLM) is a recently proposed supervised learning algorithm with performance comparable to most state-of-the-art machine learning methods. In this work, we propose ensemble methods for classification and regression using MLMs. The goal of ensemble strategies is to produce more robust and accurate models when compared to a single classifier or regression model. Despite its successful application, MLM employs a computationally intensive optimization problem as part of its test procedure (out-of-sample data estimation). This becomes even more noticeable in the context of ensemble learning, where multiple models are used. Aiming to provide fast alternatives to the standard MLM, we also propose the Nearest Neighbor Minimal Learning Machine and the Cubic Equation Minimal Learning Machine to cope with classification and single-output regression problems, respectively. The experimental assessment conducted on real-world datasets reports that ensemble of fast MLMs perform comparably or superiorly to reference machine learning algorithms.  相似文献

The CYFIP2 protein (cytoplasmic FMR1-interacting protein 2) is part of the WAVE regulatory complex (WRC). CYFIP2 was recently correlated to neurological disorders by the association of the R87C variant with early infantile epileptic encephalopathy (EIEE) patients. In this set of syndromes, the epileptic spasms and seizures since early childhood lead to impaired neurological development in children. Inside the WRC, the variant residue is at the CYFIP2 and WAVE1 protein interface. Thus, the hypothesis is that the R87C modification weakens this interaction, allowing the WRC complex’s constant activation. This work aimed to investigate the impacts of the mutation on the structure of the WRC complex through molecular dynamics simulation. For that, we constructed WRC models containing WAVE1-NCKAP1 proteins complexed with WT or R87C CYFIP2. Our simulations showed a flexibilization of the loop comprising residues 80–110 due to the loss of contacts between internal residues in the R87C CYFIP2 as well as the key role of residues R/C87, E624, and E689 in structural modification. These data could explain the mechanism by which the mutation impairs the stability and proper regulation of the WRC.  相似文献
