Assessed the role of noradrenaline (NA) on the acquisition of an aversively motivated discrimination task and its reversal. A conditioned taste aversion procedure was used. NA depletions were achieved through 2 pharmacological means: systemic N-2 chloroethyl-N-ethyl-2-bromo-benzylamine (DSP4) and destruction of the dorsal noradrenergic bundle (DNAB) with 6-hydroxydopamine (6-OHDA). Both procedures caused marked reductions of NA in the frontal cortex and hippocampus. In neither of the studies (Exp 1, DSP4 and Exp 2, DNAB) were there any significant changes between controls and NA-depleted rats in either the rate of acquisition of the original discrimination (Phase 1) or the subsequent reversal (Phase 2). This occurred irrespective of which of the 2 stimuli (a taste cue or a tongue-tactile cue) initially was used as the conditioned stimulus/stimuli (CS) (the stimulus first followed by contingent administration of lithium chloride and later, by saline injections). Thus NA does not appear to be critically involved in the acquisition and reversal of a taste/tactile discrimination task. The significance of forebrain NA for other discrimination tasks is discussed. OBJECTIVES: This study sought to determine the spectrum of electrophysiologic abnormalities found in patients with cardiac involvement due to AL (primary) amyloidosis and to evaluate the prognostic implications, particularly in relation to subsequent sudden death. BACKGROUND: Only case reports, but no series of invasive electrophysiologic studies, exist in patients with cardiac AL. METHODS: Twenty-five patients with biopsy-proven AL and cardiac involvement underwent standard invasive electrophysiologic studies. RESULTS: The function of the sinus and the atrioventricular node was preserved in most patients, but the infra-His (HV) conduction times were usually abnormal. The mean (+/-SD) HV interval for the 25 patients was 79 +/- 18 ms (range 50 to 110), and 23 patients (92%) had an abnormally prolonged interval (> 55 ms). Marked HV prolongation (> or = 80 ms) occurred in 12 patients, 6 of whom had an interval > or = 100 ms. Among the 23 patients who died during follow-up, HV prolongation was the sole independent predictor of sudden death by multivariate analysis (p = 0.05). CONCLUSIONS: Patients with cardiac AL are prone to disease in the His-Purkinje system. Prolongation of the HV interval is common and may not be suspected from the surface electrocardiogram in the presence of a narrow QRS complex. These patients have a high prevalence of sudden death, of which the HV interval is an independent predictor. The association of HV prolongation and sudden death is probably multifactorial, representing either a marker of severe myocardial infiltration with an increased propensity to lethal ventricular arrhythmias or electromechanical dissociation, or indicating severe conduction system disease eventually leading to complete atrioventricular block and bradycardic death.  相似文献

Individuals can learn to control the amplitude of mu-rhythm activity in the EEG recorded over sensorimotor cortex and use it to move a cursor to a target on a video screen. The speed and accuracy of cursor movement depend on the consistency of the control signal and on the signal-to-noise ratio achieved by the spatial and temporal filtering methods that extract the activity prior to its translation into cursor movement. The present study compared alternative spatial filtering methods. Sixty-four channel EEG data collected while well-trained subjects were moving the cursor to targets at the top or bottom edge of a video screen were analyzed offline by four different spatial filters, namely a standard ear-reference, a common average reference (CAR), a small Laplacian (3 cm to set of surrounding electrodes) and a large Laplacian (6 cm to set of surrounding electrodes). The CAR and large Laplacian methods proved best able to distinguish between top and bottom targets. They were significantly superior to the ear-reference method. The difference in performance between the large Laplacian and small Laplacian methods presumably indicated that the former was better matched to the topographical extent of the EEG control signal. The results as a whole demonstrate the importance of proper spatial filter selection for maximizing the signal-to-noise ratio and thereby improving the speed and accuracy of EEG-based communication.  相似文献

The major interest in the development of the blood-brain barrier and its underlying induction mechanisms is given by the crucial role they play in the maturation of the central nervous system in general. Whilst it is believed that it is the microenvironment in the brain that destines the endothelial cells to become committed to barrier properties, the analysis of the multitude of factors probably responsible for this commitment is extremely difficult. Therefore, in a previous study, we inaugurated the pecten oculi of the avian eye as a relatively simple in vivo model of the blood-brain barrier [Gerhardt, S. et al, Cell Tissue Res., 285 (1996) 91-100]. In the present study, we demonstrate data on the development of the pecten which allow us to understand better the commitment of barrier properties in endothelial cells in an environment which is considerably less complex than that realized in the brain. The pecten is built up by mainly two cell types, the pigmented glial cells and the endothelial cells. The pigmented cells, which are believed to originate from the retinal pigment epithelium, lose their tight junctions in the microenvironment of the vitreous body, whereas the endothelial cells, which originate from the permeable choroidal vessels, gain tight junctions and other barrier properties in the microenvironment of the vitreous body. On embryonic day 7 (E7), tight and gap junctions between epithelial-like glial cells line the vitreal border of the developing pecten. By E16, these junctions disappear, and the endothelial cells gradually acquire barrier characteristics (continuous and P-face associated tight junctions, no extravasation of lanthanum nitrate, and the exclusive expression of the glucose transporter isoform 1 and the barrier specific antigen HT7 in their luminal and abluminal membranes). The results are discussed considering the switch from an epithelial (glial) to an endothelial barrier.  相似文献

Supracricoid laryngectomy is a new development in the treatment of laryngeal cancer in Australia. It allows the removal of both vocal cords, vestibular folds, and the entire thyroid cartilage including the paraglottic space without the loss of laryngeal function. This technique may offer a significant advance in the treatment of squamous cell carcinoma of the larynx. This paper reviews the surgical management of laryngeal cancer and discusses this new technique with modifications, indications for its use and the advantages of supracricoid laryngectomy. Case reports of seven patients operated on in our institution are included.  相似文献

Anti-neutrophil cytoplasmic antibodies (ANCA), including anti-myeloperoxidase (MPO) antibodies, are associated with pauci-immune necrotizing small vessel vasculitis or glomerulonephritis. In order to substantiate a pathogenic role for ANCA, an animal model of pauci-immune ANCA-induced glomerulonephritis or vasculitis is required. Brouwer et al. reported pauci-immune glomerulonephritis in rats immunized with human MPO followed by perfusion of kidneys with lysosomal enzyme extract combined with H2O2, and suggested that this could serve as a model of ANCA-induced disease. We repeated these studies in spontaneously hypertensive rats (SHR) and Brown Norway rats (BNR). We immunized rats with human MPO. When circulating anti-MPO antibodies were detectable by indirect immunofluorescence microscopy and ELISA, blood pressure was measured, then perfusion of the left kidney of each rat was done via the renal artery in a closed, blood-free circuit with either MPO + H2O2, MPO, H2O2 alone or MPO + H2O2 + neutral protease. Rats were killed on day 4 or day 10 after perfusion, and specimens were examined by light and immunofluorescence microscopy. Pathological lesions and deposits of IgG, C3, and MPO were found in immunized rats perfused with MPO + H2O2 with or without neutral protease, or MPO alone, in both rat strains and on both day 4 and day 10. The degree of histologic injury was proportional in intensity to the amount of IgG immune deposits. Spontaneously hypertensive rats sustained more damage and higher blood pressure than Brown Norway rats. No lesion was observed in immunized rats perfused with H2O2 or in the non-perfused right kidneys. Some of the non-immunized rats perfused with MPO + H2O2 developed pathological lesions. In conclusion, these rat models are examples of immune complex-mediated glomerulonephritis, and therefore are not similar to human ANCA-associated disease.  相似文献

Four commercial spruce lignosulfonates representing the most common acidic, neutral, and alkaline sulfite pulping processes and varying significantly in molecular weight characteristics were tested as partial (40 wt %) phenol substitute materials for the manufacture of lignosulfonate‐phenol‐formaldehyde (LPF) resole resins. Similar as recently reported for technical lignins from nonsulfite pulping processes (kraft, soda, organosolv), all lignosulfonates of this study effectuated a faster viscosity gain during resole cooking compared to the lignin‐free reference resin (1000 mPa s after 120 min vs. 250 min to reach 1000 mPa s). Sodium lignosulfonate featuring the lowest weight average molecular weight (Mw 5780 g mol?1) and dispersity (Ð 6.1) turned out to be superior to the other lignosulfonates with regard to curing rate (B‐time; 3:37 min vs. 6:41–9:08 min) and tensile shear strength development under hot pressing (120 °C; TS,max = 5.64 N mm?2 after 8 min) for beech veneer strips glued together with the respective LPF resins. Calcium and magnesium lignosulfonates are less suited with regard to phenol replacement due to the poor performance of the respective LPF adhesives in terms of tensile shear strength (TS,max = 3.29–3.49 N mm?2 after 12 min) most likely caused by considerable amounts of side products formed in the course of formose‐type reactions. Phenolation of the two promising lignosulfonates, that is, sodium and ammonium lignosulfonate, did neither considerably increase the rate of PF network formation during resin cooking and curing nor improve tensile strength development during hot pressing. © 2017 Wiley Periodicals, Inc.
