Adaptive indexing initializes and optimizes indexes incrementally, as a side effect of query processing. The goal is to achieve the benefits of indexes while hiding or minimizing the costs of index creation. However, index-optimizing side effects seem to turn read-only queries into update transactions that might, for example, create lock contention. This paper studies concurrency control and recovery in the context of adaptive indexing. We show that the design and implementation of adaptive indexing rigorously separates index structures from index contents; this relaxes constraints and requirements during adaptive indexing compared to those of traditional index updates. Our design adapts to the fact that an adaptive index is refined continuously and exploits any concurrency opportunities in a dynamic way. A detailed experimental analysis demonstrates that (a) adaptive indexing maintains its adaptive properties even when running concurrent queries, (b) adaptive indexing can exploit the opportunity for parallelism due to concurrent queries, (c) the number of concurrency conflicts and any concurrency administration overheads follow an adaptive behavior, decreasing as the workload evolves and adapting to the workload needs.  相似文献

Side‐chain engineering is increasingly being utilized as a technique to impact the structural order and enhance the electronic properties of semiconducting polymers. However, the correlations drawn between structural changes and the resulting charge transport properties are typically indirect and qualitative in nature. In the present work, a combination of grazing incidence X‐ray diffraction and crystallographic refinement calculations is used to determine the precise molecular packing structure of two thiophene‐based semiconducting polymers to study the impact of side‐chain modifications. The optimized structures provide high‐quality fits to the experimental data and demonstrate that in addition to a large difference in interchain spacing between the two materials, there exists a significant disparity in backbone orientation as well. The calculated structures are utilized in density functional theory calculations to determine the band structure of the two materials and are shown to exhibit a dramatic disparity in interchain dispersion which accounts for the large observed difference in charge carrier mobility. The techniques presented here are meant to be general and are therefore applicable to many other highly diffracting semicrystalline polymers.  相似文献

Multiobjective optimization (MOO) has been successfully used to improve the process design and operation, by finding trade-offs among conflicting objectives such as energy, capital cost, and profit. In this work, the cumene process design is modified to decrease the raw materials and product losses and to facilitate better energy integration. Here, two slightly different cumene process designs are presented and evaluated. One process design uses a column to vent off the undesired chemicals, whereas the other uses two flash tanks. Additionally, vapor recompression is applied in both the designs to recover energy. Then, MOO of both modified process designs is carried out to examine two trade-offs: total capital cost (TCC) versus material loss and TCC versus utility cost. For this, an Excel-based MOO program is used; it is based on the elitist nondominated sorting genetic algorithm. The cumene process design with column is found to be superior for the first trade-off, whereas the design with two flash tanks is better for the other trade-off. Further, both the designs are compared based on their cumene production capacities; column design is found to be overall superior. Finally, energy requirements of the developed cumene process designs are compared with those reported in recent studies.  相似文献

Large supramolecular protein complexes, such as the molecular machinery involved in gene regulation, cell signaling, or cell division, are key in all fundamental processes of life. Detailed elucidation of structure and dynamics of such complexes can be achieved by reverse‐engineering parts of the complexes in order to probe their interactions with distinctive binding partners in vitro. The exploitation of DNA nanostructures to mimic partially assembled supramolecular protein complexes in which the presence and state of two or more proteins are decisive for binding of additional building blocks is reported here. To this end, four‐way DNA Holliday junction motifs bearing a fluorescein and a biotin tag, for tracking and affinity capture, respectively, are site‐specifically functionalized with centromeric protein (CENP) C and CENP‐T. The latter serves as baits for binding of the so‐called KMN component, thereby mimicking early stages of the assembly of kinetochores, structures that mediate and control the attachment of microtubules to chromosomes in the spindle apparatus. Results from pull‐down experiments are consistent with the hypothesis that CENP‐C and CENP‐T may bind cooperatively to the KMN network.  相似文献

Sewage sludge ash (SSA) is a suitable raw material for fertilizers due to its high phosphorus (P) content. However, heavy metals must be removed before agricultural application and P should be transferred into a bioavailable form. The utilization of gaseous hydrochloric acid for thermochemical heavy metal removal from SSA at approximately 1000 °C was investigated and compared to the utilization of alkaline earth metal chlorides. The heavy metal removal efficiency increased as expected with higher gas concentration, longer retention time and higher temperature. Equivalent heavy metal removal efficiency were achieved with these different Cl-donors under comparable conditions (150 g Cl/kg SSA, 1000 °C). In contrast, the bioavailability of the P-bearing compounds present in the SSA after thermal treatment with gaseous HCl was not as good as the bioavailability of the P-bearing compounds formed by the utilization of magnesium chloride. This disadvantage was overcome by mixing MgCO(3) as an Mg-donor to the SSA before thermochemical treatment with the gaseous Cl-donor. A test series under systematic variation of the operational parameters showed that copper removal is more depending on the retention time than the removal of zinc. Zn-removal was declined by a decreasing ratio of the partial pressures of ZnCl(2) and water.  相似文献

Food allergies represent a risk for many people in industrialized countries. Unrecognizable allergenic proteins of foodstuffs may be present as ingredients that are not labeled or as unknown cross-contamination. Such hidden allergens can cause severe reactions in allergics, even at minute quantities, sometimes with fatal outcome. For the verification of the presence of allergenic food constituents, analytical methods such as ELISA and PCR have been developed. However, these tests cannot measure allergenic potential. For this reason, a test system that measures the biological activity of allergens has been developed. It is based on the cellular mechanisms of the type I allergy. Rat basophilic leukemia cells (RBL-2H3) were transfected with the genes of the human high affinity receptor for IgE. The resulting cell line expressed the human receptor α-chain and could bind allergenspecific IgE from allergic subjects, in contrast to the parent cell line. After cross-linking of receptor-bound, allergen-specific human IgE by allergens, the cells released measurable inflammatory mediators. These cells were used for the analysis of a variety of allergen extracts, including extracts prepared from foods containing allergenic hazelnut and peanut. The comparative validation with existing ELISA and PCR for hazelnut and peanut demonstrated similar sensitivity and specificity. The established cell line will be a novel tool in the detection of allergens in complex mixtures, especially to address the issue of their allergenic potential, which cannot be accomplished by classical analytical methods. This will add valuable information about the allergenic potential of food constituents to the risk assessment of foods.
Zusammenfassung. Allergene Lebensmittelbestandteile stellen für viele Nahrungsmittelallergiker in industrialisierten L?ndern ein reales Gesundheitsrisiko dar, wenn sie als nicht deklarierte Zutaten oder unerkannte Kontaminationen in Lebensmitteln enthalten sind. Diese so genannten versteckten Allergene k?nnen, teilweise in geringsten Mengen, schwere Reaktion bei sensibilisierten Personen auszul?sen, manchmal auch mit t?dlichem Ausgang. Zum Nachweis allergener Lebensmittelbestandteile wurden bereits verschiedene analytische Methoden, wie z. B. ELISA- und PCR-Verfahren, entwickelt. Allerdings sind diese Methoden nicht in der Lage, das allergene Potenzial solcher Bestandteile zu erfassen. Um jedoch die biologische Aktivit?t von Allergenen messen zu k?nnen wurde ein Testsystem entwickelt, das auf dem Mechanismus der Typ-I Allergie basiert. Dazu wurden basophile Leuk?miezellen der Ratte mit den Genen des humanen hochaffinen Rezeptors für IgE transfiziert. Die daraus resultierende Zelllinie exprimiert einen chim?ren Rezeptor (mit der IgE-bindenden humanen α-Kette) stabil auf ihrer Oberfl?che und ist somit in der Lage, auch allergenspezifisches IgE aus Allergikerseren zu binden, was mit der Ausgangszelllinie nicht m?glich ist. Die Vernetzung des rezeptorgebundenen IgEs durch das Allergen führt in der Folge zur Freisetzung entzündungsausl?sender Mediatoren, die im Zellkulturüberstand gemessen werden k?nnen. Diese transfizierte Zelllinie wurde zur Analyse einer Vielzahl von Allergenextrakten eingesetzt, darunter auch Extrakte von Lebensmitteln, die Haselnuss- oder Erdnussallergene enthielten. Der Vergleich des biologischen Testsystems mit den ELISA- und PCR-Verfahren für Hasel- und Erdnuss ergab eine ?hnliche Sensitivit?t und Spezifit?t. Die etablierte Zelllinie ist ein neues, wertvolles Hilfsmittel für den Nachweis von Allergenen in komplexen, zusammengesetzten Lebensmitteln und im Besonderen zur Erfassung des allergenen Potenzials solcher Nahrungsmittelbestandteile. Somit kann dieses neue Nachweisverfahren helfen, zus?tzliche wichtige Informationen für die Risikobewertung von Lebensmitteln zu gewinnen.

Reducing sugars, free amino acids, and the potential for acrylamide formation were determined in more than 50 potato samples from the 2003 harvest in Switzerland. The reducing sugar content strongly correlated with acrylamide, whereas no correlation was found between acrylamide and free asparagine or the pool of free amino acids. The reducing sugar contents and the acrylamide potentials were higher in most of the cultivars tested than in the samples from 2002. This was probably due to the hot and dry summer of 2003. Monitoring sugars and amino acids during heating at 120 °C and 180 °C showed that glucose and fructose reacted much faster than sucrose and the amino acids. Glutamine was consumed to a larger extent than any of the other amino acids. During prolonged storage, the reducing sugars decreased considerably while only moderate changes in the free amino acids were observed. Altogether, glucose and fructose remain the critical factors for acrylamide formation in potatoes and represent the most feasible way of reducing the formation of acrylamide in potato products.  相似文献
