Investigating heavy metal resistance, bioaccumulation and metabolic profile of a metallophile microbial consortium native to an abandoned mine 总被引:1,自引:0,他引:1

Contaminated sites represent new ecological niches where historical pollution has originated an unusual microbial biodiversity. The knowledge of these microorganisms contributes to the discovery of new pathways and metabolic networks and may offer potential solutions for damaged areas. In the present work seven microbial consortia have been isolated from an abandoned mine of blend and galena (Ingurtosu, Italy) through a selection for resistance to zinc (tested up to 40 mM in solution). All the consortia were able to accumulate zinc and the best accumulator, named Ing5, has been studied for the following characteristics: resistance and accumulation of Zn, Cd, Hg, bioaccumulation mechanisms of Zn, and influence of Zn and Cd on the metabolic profile. The results indicate that the consortium Ing5 bears resistance systems for Cd and Hg as well as Zn and that, for some of the 5 isolates belonging to Ing5, the resistance thresholds are higher in consortium than in pure culture. The prevalent mechanism for zinc accumulation can be reasonably considered to be metabolism-dependent, inducible and regulated by metal concentrations. The study on the metabolic profile, carried out by the Biolog system, shows that Zn exerts a very low influence on the metabolic profile and that this influence can also be positive; Cd has a stronger negative influence but that, despite this, the consortium is able to maintain a wide metabolic potential in the presence of heavy metals. These features of Ing5 make it a good candidate for biotechnological applications and for further investigation of the degradation of organic pollutants in the presence of metals. 相似文献

The purpose of the article is to provide some evidence on the interconnection between capital structure, R&D investment and ownership concentration using a unique panel data-set of Italian firms. We study the effect of R&D intensity on leverage for two groups of firms which are different in terms of their degree of ownership concentration. Our results suggest for Public Limited Companies, a nonlinear relationship between R&D intensity and leverage, with the latter first increasing and then decreasing. Interestingly, the same result is not found to hold true for Private Limited Companies, which are characterized by a more concentrated ownership. 相似文献

Software testing is essential to guarantee high quality products. However, it is a very expensive activity, particularly when manually performed. One way to cut down costs is by reducing the input test suites, which are usually large in order to fully satisfy the test goals. Yet, since large test suites usually contain redundancies (i.e., two or more test cases (TC) covering the same requirement/piece of code), it is possible to reduce them in order to respect time/people constraints without severely compromising coverage. In this light, we formulated the TC selection problem as a constrained search based optimization task, using requirements coverage as the fitness function to be maximized (quality of the resultant suite), and the execution effort (time) of the selected TCs as a constraint in the search process. Our work is based on the Particle Swarm Optimization (PSO) algorithm, which is simple and efficient when compared to other widespread search techniques. Despite that, besides our previous works, we did not find any other proposals using PSO for TC selection, neither we found solutions treating this task as a constrained optimization problem. We implemented a Binary Constrained PSO (BCPSO) for functional TC selection, and two hybrid algorithms integrating BCPSO with local search mechanisms, in order to refine the solutions provided by BCPSO. These algorithms were evaluated using two different real-world test suites of functional TCs related to the mobile devices domain. In the performed experiments, the BCPSO obtained promising results for the optimization tasks considered. Also, the hybrid algorithms obtained statistically better results than the individual search techniques. 相似文献

A simple left atrial procedure for atrial fibrillation (AF) was performed in 16 elderly patients (age over 70 years old) with chronic AF associated with mitral and/or aortic valvular diseases. Chronic AF was eliminated upon discharge in 13 out of the 16 patients (81%). Twelve of the 13 patients (92%) whose AF had disappeared recovered the atrial kick of their right atrium, and 9 patients (70%) recovered the atrial kick of their left atrium. A simple surgical procedure to the left atrium was effective in the treatment of chronic AF associated with mitral valve disease in elderly patients. This simple procedure is preferable to other methods for the elimination of chronic AF with mitral valve disease in elderly patients. 相似文献

With the ratification of Multilateral Environmental Agreements (MEAs) the countries of the international community or of intentional communities—be they political, economic, financial, securitarian or strategic—endow these instruments of international cooperation with significant autonomy. From the 3550 dates of ratification of these MEAs recorded from 1979 to mid-September 2014, we produce a graph whose vertices are the 48 MEAs (ratified at least once) and whose links are induced by the succession of ratifications in time. On this basis we propose a diagnosis on the international acceptance of this type of legal instruments and their vulnerability in a global context that builds on the change in the balance of powers as a result of globalization, the break of the bipolar and then unipolar system, and the rise of new powers. Thus, it appears that a global environmental order has been promoted and implemented with some success in the 90s mainly by liberal Western countries who were then able to lead other countries less likely to bind to the fulfillment of environmental obligations. However, the expansion of this global environmental order now seems frozen, due to the current crisis of multilateralism. The rise of many countries, particularly in the South, whose environmental, political and economic weight grew, confronted with the “stable community” formed in the past 35 years suggests that there is a real power shift in the international arena and consequently, multilateralism needs to reflect this new reality. In other terms, the global environmental order is being slowly reformed. As a consequence, the treaties formed clusters in the past but they did not follow the same pattern since the twenty-first century began. 相似文献

Coronaviruses, including SARS-CoV-2 (the etiological agent of the current COVID-19 pandemic), rely on the surface spike glycoprotein to access the host cells, mainly through the interaction of their receptor-binding domain (RBD) with the human angiotensin-converting enzyme 2 (ACE2). Therefore, molecular entities able to interfere with the binding of the SARS-CoV-2 spike protein to ACE2 have great potential to inhibit viral entry. Starting from the available structural data on the interaction between SARS-CoV-2 spike protein and the host ACE2 receptor, we engineered a set of soluble and stable spike interactors, here denoted as S-plugs. Starting from the prototype S-plug, we adopted a computational approach by combining stability prediction, associated to single-point mutations, with molecular dynamics to enhance both S-plug thermostability and binding affinity to the spike protein. The best developed molecule, S-plug3, possesses a highly stable α-helical con-formation (with melting temperature Tm of 54 °C) and can interact with the spike RBD and S1 domains with similar low nanomolar affinities. Importantly, S-plug3 exposes the spike RBD to almost the same interface as the human ACE2 receptor, aimed at the recognition of all ACE2-accessing coronaviruses. Consistently, S-plug3 preserves a low nanomolar dissociation constant with the delta B.1.617.2 variant of SARS-CoV-2 spike protein (KD = 29.2 ± 0.6 nM). Taken together, we provide valid starting data for the development of therapeutical and diagnostic tools against coronaviruses accessing through ACE2. 相似文献
