The seeds of Araucaria angustifolia, named pinhão, are consumed in the south and southeast of Brazil. They are big in size and have high nutritious value. The literature about technological aspects of pinhão is very scarce and there are no reports about moisture sorption models. In this work, moisture desorption isotherms of raw pinhão were determined at 15, 25, 30 and 40 °C. Results show that temperature has little effect on the sorption behaviour and the Chirife model was found to best represent the experimental data. The isosteric heat of sorption (differential enthalpy) was calculated through direct use of moisture desorption isotherm by applying the Clausius‐Clapeyron equation. The differential enthalpy of desorption decreased with increasing moisture content. The enthalpy–entropy compensation theory was applied to desorption isotherms and plots of differential enthalpy vs. differential entropy for pinhão provided the isokinetic temperature, indicating an enthalpy‐controlled desorption process. 相似文献

Mutant LGM-128 of Hansenula polymorpha harbors the recessive mutation glr2-1 which confers a complex pleiotropic phenotype, the major feature of which is the metabolically unnecessary induction of methanol utilization metabolism (C1 metabolism) during growth on glucose, whether or not methanol is in the medium. Therefore, in this mutant, peroxisomes are formed and proliferate upon cultivation in glucose-containing media. In these media, LGM-128 shows induction levels of C1 metabolism that are similar to those observed in methanol-containing media. This indicates that GLR2 controls the repression-derepression process stimulated by glucose and that the induction process triggered by methanol plays only a minor role in activating C1 metabolism. Cultivating LGM-128 in methanol and then transferring it to glucose media revealed that active degradative processes occur, leading to the disappearance of C1 metabolism. This observation suggests that, although stimulated by glucose, the two processes are controlled by elements which are, at least in part, distinct. Finally, glr2-1 does not affect ethanol repression, suggesting that in H. polymorpha the two repressing circuits are separated. 相似文献

The catalytic subunit (L-microCANP) of human calpain I (muCANP, the high Ca2+ affinity form) and two of its mutants were expressed in Escherichia coli or using the baculovirus Sf9 system. The mutants lacked domain III (L-mu CANPDelta3) and the calmodulin-like domain IV (L-mu CANPDelta4), respectively. The bacterially expressed proteins were solubilized from the inclusion bodies and refolded with polyethylene glycol. In Sf9 cells, co-expression of the inhibitor calpastatin was necessary to prevent autolysis of L-muCANP, whereas co-expression of the regulatory subunit enhanced it. Only very low levels of mRNA of the truncated form L-mu CANPDelta4 were found in bacmid-transfected Sf9 cells, and it proved impossible to isolate this mutant using the baculovirus expression system. While the apparent Km(Ca2+) of freshly isolated human erythrocyte muCANP was about 60 microM, the recombinant monomeric forms L-mu CANP and L-mu CANPDelta3 required 65-215 and 400-530 microM Ca2+, respectively. Bacterially expressed L-mu CANPDelta4 was Ca2+-independent; the presence of inhibitors during its renaturation was necessary to prevent its autolysis. A chimeric form (L-mu mCANP) composed by domains I-III of muCANP and domain IV of calpain II (mCANP, the low Ca2+ affinity form) was also expressed in Sf9 cells. This mutant required less Ca2+ (about 50 microM) than native erythrocyte calpain for half-maximal activity and had the highest specific activity of all calpains tested. Domain III proved unnecessary for the activity of the recombinant catalytic subunit, but its absence raised the Km(Ca2+) and removed its inactivation at high Ca2+ concentrations. All recombinant proteins were active as monomers in polyethylene glycol-containing buffers; the in vitro association with the regulatory subunit enhanced only slightly the Vmax and the Ca2+ dependence of the expressed proteins. The design of bioactive scaffold materials able to guide cellular processes involved in new-tissue genesis is key determinant in bone tissue engineering. The aim of this study was the design and characterization of novel multi-phase biomaterials to be processed for the fabrication of 3D porous scaffolds able to provide a temporary biocompatible substrate for mesenchymal stem cells (MSCs) adhesion, proliferation and osteogenic differentiation. The biomaterials were prepared by blending poly(ε-caprolactone) (PCL) with thermoplastic zein (TZ), a thermoplastic material obtained by de novo thermoplasticization of zein. Furthermore, to bioactivate the scaffolds, microparticles of osteoconductive hydroxyapatite (HA) were dispersed within the organic phases. Results demonstrated that materials and formulations strongly affected the micro-structural properties and hydrophilicity of the scaffolds and, therefore, had a pivotal role in guiding cell/scaffold interaction. In particular, if compared to neat PCL, PCL–HA composite and PCL/TZ blend, the three-phase PCL/TZ–HA showed improved MSCs adhesion, proliferation and osteogenic differentiation capability, thus demonstrating potential for bone regeneration. 相似文献

Natural materials are becoming a valid alternative to traditional synthetic ones for sound absorption applications. In this paper, the acoustic properties of broom fibers are reported and discussed. Broom plants grow up on hills and mountains and are common in many Mediterranean countries. The materials were cut and shredded to prepare several samples. Test specimens made of broom pieces with a diameter from 1.5 mm to 4 mm were prepared. The normal sound absorption for samples with a thickness of 60 mm, 80 mm and 120 mm was then measured. Results are particularly promising especially above 500 Hz, as the samples consistently reported a normal sound absorption above 0.5. The increase in the sample thickness allowed to obtain higher sound absorption at lower frequencies, as generally expected for porous materials. The main limit of the investigated samples was the appearance of sound absorption resonances. These were more evident for the samples composed of pieces with larger diameter, and may be attributed to the loose nature of the tested samples. 相似文献

Passive vibration absorbers are really effective only at the tuned frequency. This limit is overcome by adaptive absorbers which offer the possibility to adjust their behaviour according to needs. The paper focuses on a smart actuator for vibration control made up by a dielectric electro-active polymer (DEAP). The DEAP is obtained constraining a silicone corrugated sheet between two silver layers and it is manufactured by Danfoss Polypower A/S. The actuator is fabricated by rolling the DEAP sheet in a cylindrical and core-free shape. The paper describes the static and dynamic characterizations of the actuator. A theoretical model is developed by considering the actuator as a multi-elements model. Moreover, a modal test of the actuator is performed driving it with an electro-dynamic shaker in a frequency range up to 1 kHz. A good agreement between theoretical and experimental data is obtained at lower frequencies. Stiffness and damping laws of the actuator are determined by fitting theoretical expectations and test results. The actuator is proved useful for controlling vibrations at low frequencies because it shows internal resonances at frequencies above 75 Hz. Finally, first results about the use of the DEAP actuator for vibration control of harmonic excitations in a band-limited frequency range below 10 Hz are reported. 相似文献

Abstract Half-fresh apples were immersed in sucrose solution (50% w/w, 27°C) during different times of exposition (2, 4, and 8 h). Then each fruit was sliced from the transversal exposed surface. Density, water, and sugar content were determined for each slice. A mathematical model was fitted to experimental data of water and sucrose content considering the global flux and the tissue shrinkage. By numerical analysis, the binary effective diffusion coefficients as a function of concentration were calculated, using material coordinates and integrating simultaneously two differential equations (for water and sucrose). The coefficients obtained are one or even two orders of magnitude lower than the ones for pure solutions and present an unusual concentration dependence. This comparison shows the influence of the tissue resistance to the diffusion. 相似文献
