Hfq is a pleiotropic regulator that mediates several aspects of bacterial RNA metabolism. The protein notably regulates translation efficiency and RNA decay in Gram-negative bacteria, usually via its interaction with small regulatory RNAs. Previously, we showed that the Hfq C-terminal region forms an amyloid-like structure and that these fibrils interact with membranes. The immediate consequence of this interaction is a disruption of the membrane, but the effect on Hfq structure was unknown. To investigate details of the mechanism of interaction, the present work uses different in vitro biophysical approaches. We show that the Hfq C-terminal region influences membrane integrity and, conversely, that the membrane specifically affects the amyloid assembly. The reported effect of this bacterial master regulator on membrane integrity is discussed in light of the possible consequence on small regulatory RNA-based regulation.  相似文献

Many modern program verifiers translate the program to be verified and its specification into a simple intermediate representation and then compute verification conditions on this representation. Using an intermediate language improves the interoperability of tools and facilitates the computation of small verification conditions. Even though the translation into an intermediate representation is critical for the soundness of a verifier, this step has not been formally verified. In this paper, we formalize the translation of a small subset of Java bytecode into an imperative intermediate language similar to BoogiePL. We prove soundness of the translation by showing that each bytecode method whose BoogiePL translation can be verified, can also be verified in a logic that operates directly on bytecode.  相似文献

Introduction: Three-dimensional bioprinting can be considered as an advancement of the classical tissue engineering concept. For bioprinting, cells have to be dispersed in hydrogels. Recently, a novel semi-synthetic thiolene hydrogel system based on norbornene-functionalized gelatin (GelNB) and thiolated gelatin (GelS) was described that resulted in the photoclick hydrogel GelNB/GelS. In this study, we evaluated the printability and biocompatibility of this hydrogel system towards adipose-tissue-derived mesenchymal stem cells (ASCs). Methods: GelNB/GelS was synthesized with three different crosslinking densities (low, medium and high), resulting in different mechanical properties with moduli of elasticity between 206 Pa and 1383 Pa. These hydrogels were tested for their biocompatibility towards ASCs in terms of their viability, proliferation and differentiation. The extrusion-based bioprinting of ASCs in GelNB/GelS-high was performed to manufacture three-dimensional cubic constructs. Results: All three hydrogels supported the viability, proliferation and chondrogenic differentiation of ASCs to a similar extent. The adipogenic differentiation of ASCs was better supported by the softer hydrogel (GelNB/GelS-low), whereas the osteogenic differentiation was more pronounced in the harder hydrogel (GelNB/GelS-high), indicating that the differentiation fate of ASCs can be influenced via the adaption of the mechanical properties of the GelNB/GelS system. After the ex vivo chondrogenic differentiation and subcutaneous implantation of the bioprinted construct into immunocompromised mice, the production of negatively charged sulfated proteoglycans could be observed with only minimal inflammatory signs in the implanted material. Conclusions: Our results indicate that the GelNB/GelS hydrogels are very well suited for the bioprinting of ASCs and may represent attractive hydrogels for subsequent in vivo tissue engineering applications.  相似文献

An across track interferometric synthetic aperture radar (InSAR) is used to image ocean waves. Across track InSAR data were acquired during the SAR INnterferometry Experiment for validation of ocean Wave imaging models (SINEWAVE) in the North Sea using an airborne X-band radar with horizontal polarization. A wind sea system was imaged at different flight levels and with different flight directions with respect to the ocean wave propagation direction. Simultaneously, ocean wave spectra were measured by a directional wave rider buoy. Thus, the experiment data comprises synthetic aperture radar (SAR) intensity, coherence, and phase images together with in situ measurements. As shown in a recent theoretical study by Schulz-Stellenfleth and Lehner (2001), across track InSAR provides distorted (bunched) digital elevation models (DEMs) of the sea surface. Using SINEWAVE data the DEM bunching mechanism is verified with in situ ocean wave measurements available for the first time. It is shown that significant waveheight as well as one-dimensional (1D) wavenumber spectra derived from bunched DEMs and buoy data are in good agreement for small nonlinearities. Peak wave directions and peak wavelength detected in bunched DEMs and SAR intensity images are compared with the buoy spectrum. Peak rotations of up to 30° with respect to the buoy spectrum are found depending on flight direction and flight level. Two-dimensional (2D) spectra of bunched DEMs, corresponding coherency maps, and SAR intensity images are intercompared. The signal-to-noise ratio (SNR) of bunched DEM spectra is shown to be about 5 to 10 dB higher than the SNR of SAR intensity image spectra  相似文献

Trace elements, functionalized nanoparticles and labeled entities can be localized with sub-mm spatial resolution by X-ray fluorescence imaging (XFI). Here, small animals are raster scanned with a pencil-like synchrotron beam of high energy and low divergence and the X-ray fluorescence is recorded with an energy-dispersive detector. The ability to first perform coarse scans to identify regions of interest, followed by a close-up with a sub-mm X-ray beam is desirable, because overall measurement time and X-ray dose absorbed by the (biological) specimen can thus be minimized. However, the size of X-ray beams at synchrotron beamlines is usually strongly dependent on the actual beamline setup and can only be adapted within specific pre-defined limits. Especially, large synchrotron beams are non-trivial to generate. Here, we present the concept of graphite-based, convex reflection optics for the one-dimensional enlargement of a 1 mm wide synchrotron beam by a factor of 5 to 10 within a 1 m distance. Four different optics are tested and characterized and their reflection properties compared to ray tracing simulations. The general shape and size of the measured reflection profiles agree with expectations. Enhancements with respect to homogeneity and efficiency can be expected with improved optics manufacturing. A mouse phantom is used for a proof-of-principle XFI experiment demonstrating the applicability of coarse and fine scans with the suggested optics design.  相似文献

Safety Certificate is a musical performance based on sensor data from high-speed trains. The original purpose of this data is to provide a basis for the assessments of the mechanical aspects of train safety. In this performance, the data, which represents dynamical processes below the audible range, are converted into sound through audification. The sound that is generated live during the performance is manipulated through the Manta control interface, which allows for the convenient layering of 48 different timbres. Safety Certificate was premiered at Seconde Nature in Aix-en-Provence in March 2010 during the Sonification symposium–What, Where, How, Why, organized by Locus Sonus. The following short article gives details about the data, the audification technique, use of the control interface, and the musical structure of the performance.  相似文献

A variety of schemes have been proposed in the literature to speed up query processing and analytics by incrementally maintaining a bounded-size uniform sample from a dataset in the presence of a sequence of insertion, deletion, and update transactions. These algorithms vary according to whether the dataset is an ordinary set or a multiset and whether the transaction sequence consists only of insertions or can include deletions and updates. We report on subtle non-uniformity issues that we found in a number of these prior bounded-size sampling schemes, including some of our own. We provide workarounds that can avoid the non-uniformity problem; these workarounds are easy to implement and incur negligible additional cost. We also consider the impact of non-uniformity in practice and describe simple statistical tests that can help detect non-uniformity in new algorithms.  相似文献
