A novel electrophoretic alpha-lactalbumin (alpha-la) variant was detected in the Italian water buffalo breed. The isoelectric point of the variant, labelled A, was lower than the most frequent variant B. It presented an allelic frequency of 0.5% compared with the 97.1% of the BB allele. From Liquid Chromatography-Electrospray Ionization/Mass spectrometry, the molecular mass of the two alpha-la A and B variants were measured as 14,235.1+/-0.8 and 14,236.1+/-0.9 Da, respectively. The two proteins were sequenced and differentiated from one another by a single amino acid substitution, Asn45(B)-->Asp45(A). As this amino acid substitution altered the N-glycosylation sequence consensus Asn45-X-Ser46 it may be deduced that the protein glycosylation level of the alpha-la A would decrease.  相似文献

A new method for decoding two-dimensional (2D) multicomponent separations based on the use of the 2D Autocovariance function (2D-ACVF) has been developed. Theoretical models of single component (SC) spot distributions in 2D separations, both random and structured, are developed as the basis for a nonlinear estimation of both sample and separation system parameters from experimental 2D separations. The number of SCs, the average spot size, the spot capacity, and the saturation factor can be evaluated in the case of random SC spot patterns. The procedure was validated by extensive numerical simulation under conditions close to those usually found in GC x GC or 2D-polyacrylamid gel electrophoresis of proteins. The worse precision degree was no greater than 10% in the case of maximum spot density. This imprecision was fully accounted for, and it seems acceptable owing to the intrinsic statistical character of the estimation method. Structured multicomponent 2D separations, where SCs are linked by linear relationships, give rise to specific structured patterns in 2D-ACVF plots from which the parameters (phase and frequency) of the structured SC sequences can be evaluated: the study of 2D-ACVF makes it possible to decode multicomponent 2D separation, that is, to determine the number, relative abundance, and structural similarities of the single components. Pertinent expressions of the theoretical 2D-ACVF were derived for simple cases, and a procedure for decoding cases of structured 2D separations was developed and applied. It was shown that 2D separations containing both random and structured patterns of SC spots give rise to 2D-EACVF, which is the superimposition of the two component parts. This feature allows one, in principle, to decode the two components. The relevance of these results for Giddings sample dimensionality and separation dimensionality and their effective experimental evaluation is discussed.  相似文献

In this paper we propose a combinatorial approach to the study of addressable machines. As a consequence of such an approach the set of instructions defining our machine is the union of the set of the generators of a free group and of the set of their inverses together with the identity. Time-delay estimation (TDE) is a common operation in ultrasound signal processing. In applications such as blood flow estimation, elastography, phase aberration correction, and many more, the quality of final results is heavily dependent upon the performance of the time-delay estimator implemented. In the past years, several algorithms have been developed and applied in medical ultrasound, sonar, radar, and other fields. In this paper we analyze the performances of the widely used normalized and non-normalized correlations, along with normalized covariance, sum absolute differences (SAD), sum squared differences (SSD), hybrid-sign correlation, polarity-coincidence correlation, and the Meyr-Spies method. These techniques have been applied to simulated ultrasound radio frequency (RF) data under a variety of conditions. We show how parameters, which include center frequency, fractional bandwidth, kernel window size, signal decorrelation, and signal-to-noise ratio (SNR) affect the quality of the delay estimate. Simulation results also are compared with a theoretical performance limit set by the Cramer-Rao lower bound (CRLB). Results show that, for high SNR, high signal correlation, and large kernel size, all of the algorithms closely match the theoretical bound, with relative performances that vary by as much as 20%. As conditions degrade, the performances of various algorithms differ more significantly. For signals with a correlation level of 0.98, SNR of 30 dB, center frequency of 5 MHz with a fractional bandwidth of 0.5, and kernel size of 2 /spl mu/s, the standard deviation of the jitter error is on the order of few nanoseconds. Normalized correlation, normalized covariance, and SSD have an approximately equal jitter error of 2.23 ns (the value predicted by the CRLB is 2.073 ns), whereas the polarity-coincidence correlation performs less well with a jitter error of 2.74 ns.  相似文献

Articular cartilage defects do not heal. Biodegradable scaffolds have been studied for cartilage engineering in order to implant autologous chondrocytes and help cartilage repair. We tested some new collagen matrices differing in collagen type, origin, structure and methods of extraction and purification, and compared the behavior of human chondrocytes cultured on them. Human chondrocytes were grown for three weeks on four different equine type I collagen matrices, one type I, III porcine collagen matrix and one porcine type II collagen matrix. After 21 days, samples were subjected to histochemical, immunohistochemical and histomorphometric analysis to study phenotype expression and cell adhesion. At 7, 14 and 21 days cell proliferation was studied by incorporation of [3H]-thymidine. Our data evidence that the collagen type influences cell morphology, adhesion and growth; indeed, cellularity and rate of proliferation were significantly higher and cells were rounder on the collagen II matrix than on either of the collagen I matrices. Among the collagen I matrices, we observed a great variability in terms of cell adhesion and proliferation. The present study allowed us to identify one type I collagen matrix and one type II collagen matrix that could be usefully employed as a scaffold for chondrocyte transplantation.  相似文献

Selective hydrolysis of borage (Borago officinalis L.) oil was catalyzed by two lipase preparations of Nigella sativa L. seeds at 40°C in a mixture of borage oil, water, and hexane. Ammonium sulfate-precipitated lipase (Nigella PL) and lipase partially purified by DEAE-ion exchange chromatography (Nigella CPL) exhibited a negative specificity toward γ-linolenic acid (GLA). Best results were obtained in the experiments conducted with 330 U/g oil of Nigella PL and 200 U/g oil of nigella CPL. When 330 U/g oil of Nigella PL was used, after 8 h the GLA level rose from 21.9% in the starting oil to 29.6 and 41.8% in TAG and DAG fractions of the product mixtures, respectively (1.5-fold enrichment of GLA in the total unhydrolyzed acylglycerol fraction). At 200 U/g oil enzyme concentration of Nigella CPL, after 77 h maximum GLA enrichment was observed in the DAG fraction. The GLA content of the DAG increased to 34.6%, corresponding to almost 1.6-fold enrichment. The relative inability of Nigella sativa lipase(s) to hydrolyze γ-linolenoyl moieties of TAG can be used for the enrichment of this acid in the unhydrolyzed acylglycerol fractions of GLA-containing oils.  相似文献

In this paper we propose an application-independent model for the definition of artificial institutions that can be used to define open multi-agent systems. Such a model of institutional reality makes us able also to define an objective and external semantics of a commitment-based Agent Communication Language (ACL). In particular we propose to regard an ACL as a set of conventions to act on a fragment of institutional reality, defined in the context of an artificial institution. Another contribution of the work presented in this paper is an operational definition of norms, a crucial component of artificial institutions. In fact in open systems interacting agents might not conform to the specifications. We regard norms as event-driven rules that when are fired by events happening in the system create or cancel a set of commitments. An interesting aspect of our proposal is that both the definition of the ACL and the definition of norms are based on the same notion of commitment. Therefore an agent capable of reasoning on commitments can reason on the semantics of communicative acts and on the system of norms.  相似文献

Multiagent cooperative negotiation is a promising technique for modeling and controlling complex systems. Effective and flexible cooperative negotiations are especially useful for open complex systems characterized by high decentralization (which implies a low amount of exchanged information) and by dynamic connection and disconnection of agents. Applications include ad hoc network management, vehicle formation, and physiological model combination. To obtain an effective control action, the stability of the negotiation, namely the guarantee that an agreement will be eventually reached, is of paramount importance. However, the techniques usually employed for assessing the stability of a negotiation can be hardly applied in open scenarios. In this paper, whose nature is mainly theoretical, we make a first attempt towards engineering stable cooperative negotiations proposing a framework for their analysis and design. Specifically, we present a formal protocol for cooperative negotiations between a number of agents and we propose a criterion for negotiation stability based on the concept of connective stability. This is a form of stability that accounts for the effects of structural changes on the composition of a system and that appears very suitable for multiagent cooperative negotiations. To show its possible uses, we apply our framework for connective stability to some negotiations taken from literature.  相似文献

In recent years, several publications have appeared on virtual reality (VR) therapy in the treatment of anxiety disorders. However, a limited number of these studies targeted stress management and relaxation. To evaluate the efficacy of VR as a support tool in the relaxation process, this study compared its efficacy with two other media (DVD and audiotape). The goals of the experiment were to test the efficacy of each condition in increasing relaxation and reducing anxiety, and whether the sense of presence-the feeling of being inside the media content-correlated with the efficacy of the treatment. Results suggested the importance of the sense of presence as a mediating variable between the media experience and the efficacy of the protocol.
