Predicting comfort levels in cities is challenging due to the many metric assessment. To overcome these challenges, much research is being done in the computing community to develop methods capable of generating outdoor comfort data. Machine Learning (ML) provides many opportunities to discover patterns in large datasets such as urban data. This paper proposes a data-driven approach to build a predictive and data-generative model to assess outdoor thermal comfort. The model benefits from the results of a study, which analyses Computational Fluid Dynamics (CFD) urban simulation to determine the thermal and wind comfort in Tallinn, Estonia. The ML model was built based on classification, and it uses an opaque ML model. The results were evaluated by applying different metrics and show us that the approach allows the implementation of a data-generative ML model to generate reliable data on outdoor comfort that can be used by urban stakeholders, planners, and researchers.  相似文献

DNA-templated chemical reactions have found wide applications in drug discovery, programmed multistep synthesis, nucleic acid detection, and targeted drug delivery. The control of these reactions has, however, been limited to nucleic acid hybridization as a means to direct the proximity between reactants. In this work a system capable of translating protein–protein binding events into a DNA-templated reaction which leads to the covalent formation of a product is introduced. Protein-templated reactions by employing two DNA-antibody conjugates that are both able to recognize the same target protein and to colocalize a pair of reactant DNA strands able to undergo a click reaction are achieved. Two individual systems, each responsive to human serum albumin (HSA) and human IgG, are engineered and it is demonstrated that, while no reaction occurs in the absence of proteins, both protein-templated reactions can occur simultaneously in the same solution without any inter-system crosstalk.  相似文献

There are diverse diseases such as some infections, trauma, and tumor resections during cancer surgery that can cause bone damage or skeletal defects in persons. Most of the time, these defects cannot heal spontaneously due to several medical conditions that patients encounter, like diabetes, hormone-related problems, and autoimmune disorders. This issue is even worse for older people and some special treatments should be provided for them. Bone-tissue engineering has emerged to tackle these challenges. By investigating bone repair strategies, studying bone structures and biomechanics, and employing appropriate growth factors, suitable scaffolds, and biomaterial-centered regenerative approaches can be employed to treat bone defects more effectively. This study reviews some recent bone-tissue-engineering strategies relying on two-dimensional (2D) materials, including graphene and its derivatives, black phosphorus, and MXenes that are exhibiting a great potential in regenerative medicine.  相似文献

This paper aims to define the stability over the time of chemical elemental patterns in some citrus varieties of PGI (Protected Geographical Indication) and not PGI Interdonato and Lunario cultivars coming from Italy and Turkey and to set up a range of markers useful to trace their geographical origin. During the 2015–2017 growing seasons, all fruits were collected and subjected to Inductively Coupled Mass spectrometry (ICP-MS) analysis, in order to determine the multielemental chemical profiles. The chemical variability was calculated for each element by Multi-way analysis of variance. The results highlighted how the measured Cr, Ni, Al, K, Fe and Zn levels mainly depend on the soil composition and the fingerprint allow to trace the geographical origins. Moreover, the stepwise linear discriminant analysis (SLDA) has allowed to correctly classify the 100% of lemon pulps provenances, based on Ni, Al, K, Ca and Na contents. Finally, our study demonstrates the role played by harvest years, variety and soils composition whose interaction contributed to define the chemical fingerprints.  相似文献

Cigarette smoking has a negative impact on the skeletal system, as it reduces bone mass and increases fracture risk through its direct or indirect effects on bone remodeling. Recent evidence demonstrates that smoking causes an imbalance in bone turnover, making bone vulnerable to osteoporosis and fragility fractures. Moreover, cigarette smoking is known to have deleterious effects on fracture healing, as a positive correlation between the daily number of cigarettes smoked and years of exposure has been shown, even though the underlying mechanisms are not fully understood. It is also well known that smoking causes several medical/surgical complications responsible for longer hospital stays and a consequent increase in the consumption of resources. Smoking cessation is, therefore, highly advisable to prevent the onset of bone metabolic disease. However, even with cessation, some of the consequences appear to continue for decades afterwards. Based on this evidence, the aim of our review was to evaluate the impact of smoking on the skeletal system, especially on bone fractures, and to identify the pathophysiological mechanisms responsible for the impairment of fracture healing. Since smoking is a major public health concern, understanding the association between cigarette smoking and the occurrence of bone disease is necessary in order to identify potential new targets for intervention.  相似文献

Amyloid-β aggregation is one of the principal causes of amyloidogenic diseases that lead to the loss of neuronal cells and to cognitive impairments. The use of gold nanoparticles treating amyloidogenic diseases is a promising approach, because the chemistry of the gold surface can be tuned in order to have a specific binding, obtaining effective tools to control the aggregation. In this paper, we show, by means of Replica Exchange Solute Tempering Molecular Simulations, how electrostatic interactions drive the absorption of Amyloid-β monomers onto citrates-capped gold nanoparticles. Importantly, upon binding, amyloid monomers show a reduced propensity in forming β-sheets secondary structures that are characteristics of mature amyloid fibrils.  相似文献

Functional alterations in irritable bowel syndrome have been associated with defects in bioenergetics and the mitochondrial network. Effects of high fat, adequate-protein, low carbohydrate ketogenic diet (KD) involve oxidative stress, inflammation, mitochondrial function, and biogenesis. The aim was to evaluate the KD efficacy in reducing the effects of stress on gut mitochondria. Newborn Wistar rats were exposed to maternal deprivation to induce IBS in adulthood. Intestinal inflammation (COX-2 and TRL-4); cellular redox status (SOD 1, SOD 2, PrxIII, mtDNA oxidatively modified purines); mitochondrial biogenesis (PPAR-γ, PGC-1α, COX-4, mtDNA content); and autophagy (Beclin-1, LC3 II) were evaluated in the colon of exposed rats fed with KD (IBD-KD) or standard diet (IBS-Std), and in unexposed controls (Ctrl). IBS-Std rats showed dysfunctional mitochondrial biogenesis (PPAR-γ, PGC-1α, COX-4, and mtDNA contents lower than in Ctrl) associated with inflammation and increased oxidative stress (higher levels of COX-2 and TLR-4, SOD 1, SOD 2, PrxIII, and oxidatively modified purines than in Ctrl). Loss of autophagy efficacy appeared from reduced levels of Beclin-1 and LC3 II. Feeding of animals with KD elicited compensatory mechanisms able to reduce inflammation, oxidative stress, restore mitochondrial function, and baseline autophagy, possibly via the upregulation of the PPAR-γ/PGC-1α axis.  相似文献

Acadesine (ACA), a pharmacological activator of AMP-activated protein kinase (AMPK), showed a promising beneficial effect in a mouse model of colitis, indicating this drug as an alternative tool to manage IBDs. However, ACA displays some pharmacodynamic limitations precluding its therapeutical applications. Our study was aimed at evaluating the in vitro and in vivo effects of FA-5 (a novel direct AMPK activator synthesized in our laboratories) in an experimental model of colitis in rats. A set of experiments evaluated the ability of FA5 to activate AMPK and to compare the efficacy of FA5 with ACA in an experimental model of colitis. The effects of FA-5, ACA, or dexamethasone were tested in rats with 2,4-dinitrobenzenesulfonic acid (DNBS)-induced colitis to assess systemic and tissue inflammatory parameters. In in vitro experiments, FA5 induced phosphorylation, and thus the activation, of AMPK, contextually to the activation of SIRT-1. In vivo, FA5 counteracted the increase in spleen weight, improved the colon length, ameliorated macroscopic damage score, and reduced TNF and MDA tissue levels in DNBS-treated rats. Of note, FA-5 displayed an increased anti-inflammatory efficacy as compared with ACA. The novel AMPK activator FA-5 displays an improved anti-inflammatory efficacy representing a promising pharmacological tool against bowel inflammation.  相似文献
