Few data are available on the effect of biomaterials on surface antigens of mammalian bone marrow-derived, adult mesenchymal stromal cells (MSCs). Since poly(l-lactic acid) or PLLA is largely used in tissue engineering of human bones, and we are developing a reverse engineering program to prototype with biomaterials the vascular architecture of bones for their bioartificial reconstruction, both in humans and animal models, we have studied the effect of porous, flat and smooth PLLA scaffolds on the immunophenotype of in vitro grown, rat MSCs in the absence of any coating, co-polymeric enrichment, and differentiation stimuli. Similar to controls on plastic, we show that our PLLA scaffold does not modify the distribution of some surface markers in rat MSCs. In particular, the maintained expression of CD73 and CD90 on two different subpopulations (small and large cells) is consistent with their adhesion to the PLLA scaffold through specialized appendages, and to their prominent content in actin. In addition, our PLLA scaffold favours retention of the intermediate filament desmin, believed a putative marker of undifferentiated state. Finally, it preserves all rat MSCs morphotypes, and allows for their survival, adhesion to the substrate, and replication. Remarkably, a subpopulation of rat MSCs grown on our PLLA scaffold exhibited formation of membrane protrusions of uncertain significance, although in a size range and morphology compatible with either motility blebs or shedding vesicles. In summary, our PLLA scaffold has no detrimental effect on a number of features of rat MSCs, primarily the expression of CD73 and CD90. 相似文献

Recent investigators have proposed that amodal completion is a sequential process requiring a preliminary mosaic stage. Results of 6 studies of the time course of completion processes show support for this mosaic-first view with pictorial displays, but not with displays involving occlusion specified by binocular parallax or when pictorial displays were observed monocularly. A lower bound on the Shannon capacity of the uplink channel of a narrowband cellular system is evaluated as a function of the reuse distance and the level of shadowing affecting the interfering signals assuming perfect channel state information (CSI) at the receiver. The effect of the first two tiers of interferers is determined, and compared with a simplified evaluation that takes into account only the first tier.Spectral efficiency is evaluated for different traffic loads, to assess the usefulness of traffic control. A number of matrix metalloproteinases (MMPs), proteins important in the balance of bone remodeling, play a critical role both in cancer metastasis and in bone matrix turnover associated with the presence of cancer cells in bone. Here, we report the synthesis and biological evaluation of a new class of MMP inhibitors characterized by a bisphosphonate function as the zinc binding group. Since the bisphosphonate group is also implicated in osteoclast inhibition and provides a preferential affinity to biological apatite, the new molecules can be regarded as bone-seeking medicinal agents. Docking experiments were performed to clarify the mode of binding of bisphosphonate inhibitors in the active site of MMP-2. The most promising of the studied bisphosphonates showed nanomolar inhibition against MMP-2 and resulted in potent inhibition of osteoclastic bone resorption in vitro. 相似文献

Axonal transport and neuronal survival depend critically on active transport and axon integrity both for supplying materials and communication to different domains of the cell body. All these actions are executed through cytoskeleton, transport and regulatory elements that appear to be disrupted in neurodegenerative diseases. Motor-driven transport both supplies and clears distal cellular portions with proteins and organelles. This transport is especially relevant in projection and motor neurons, which have long axons to reach the farthest nerve endings. Thus, any disturbance of axonal transport may have severe consequences for neuronal function and survival. A growing body of literature indicates the presence of alterations to the motor molecules machinery, not only in expression levels and phosphorylation, but also in their subcellular distribution within populations of neurons, which are selectively affected in the course of neurodegenerative diseases. The implications of this altered subcellular localization and how this affects axon survival and neuronal death still remain poorly understood, although several hypotheses have been suggested. Furthermore, cytoskeleton and transport element localization can be selectively disrupted in some disorders suggesting that specific loss of the axonal functionality could be a primary hallmark of the disorder. This can lead to axon degeneration and neuronal death either directly, through the functional absence of essential axonal proteins, or indirectly, through failures in communication among different cellular domains. This review compares the localization of cytoskeleton and transport elements in some neurodegenerative disorders to ask what aspects may be essential for axon survival and neuronal death. 相似文献

The effect of amount of clay content on the thermomechanical and electrical properties of epoxy/organoclay nanocomposites is investigated in the present research. An organoclay, cloisite 30B (C30B), was dispersed in the epoxy resin and was cured with an amine curing agent. The morphology of the nanocomposite examined by X‐ray diffraction shows exfoliation for nanocomposites with lesser clay content and intercalation for nanocomposites with higher clay content. The storage modulus (E′) of the nanocomposites increases monotonously with the increase in the amount of clay. The short time alternating current breakdown strength of the nanocomposites increases by the addition of C30B up to a certain clay content and then show a decrease. The space charge measured by pulsed electroacoustic method shows that the nanocomposite accumulate a very less amount of space charge and the charge decay in the nanocomposites are quicker than in the pure polymer. The thermoelectric properties of defected graphene nanoribbons (GNRs) and multi-junction (MJ) GNRs coupling periodic armchair
sections of different width are analyzed by means of Green’s function techniques to simulate electron and phonon transport.
Among the different strategies likely to enhance the thermoelectric performance, the effects of edge disorder and random vacancies
are shown to be small since they lead to the concomitant degradation of the phonon thermal conductance and of the electronic
conductance, which finally reduces the thermoelectric factor ZT. However, the periodic distribution of vacancies and the structuring of GNRs in MJ-GNRs both lead to the enhancement of the
figure of merit ZT. In the latter case, in addition to the strong reduction of the phonon thermal conductance, an effect of resonant tunneling
of electrons allows retaining high electronic conductance and enhancing significantly the thermopower. Finally, by introducing
a periodic distribution of vacancies in the MJ-GNR, the maximum value ZT=0.4 is reached at room temperature. 相似文献
