Titanium dioxide (TiO2) bulk with pure anatase phases was fabricated by an explosive compaction technique using an underwater shockwave. Dynamic shock pressure of 6 GPa was used to consolidate anatase TiO2 powders. Its microstructural, crystalline structural and photocatalytic characteristics were observed and measured by various techniques, such as X-ray diffraction (XRD), scanning electron microscopy (SEM) and photocatalytic activity measurement system. It was confirmed that the relative density of anatase TiO2 compact is about 96% (3.73 g/cm3) of the theoretical density (3.89 g/cm3) and a strong surface bonding between particles is formed by a shock energy. In X-ray diffraction analysis, high purity anatase phases, broadened peaks due to lattice defects and decreased crystallite size were found. For the photocatalytic activities, the anatase TiO2 compact was quite excellent compared to the commercial sintered TiO2 bulk.  相似文献

In this study, complexes composed of poly-l-tyrosine (pLT) and single-walled carbon nanotubes (SWCNTs) were produced and the dispersibility of the pLT/SWCNT complexes in water by measuring the ζ potential of the complexes and the turbidity of the solution were investigated. It is found that the absolute value of the ζ potential of the pLT/SWCNT complexes is as high as that of SWCNTs modified with double-stranded DNA (dsDNA) and that the complexes remain stably dispersed in the water at least for two weeks. Thermogravimetry analysis (TGA) and visualization of the surface structures of pLT/SWCNT complexes using an atomic force microscope (AFM) were also carried out.  相似文献

There is a great deal of interest in proteins that can bind covalently to target molecules, as they allow unambiguous experiments by tight binding to molecules of interest. Here, we report the generation of proteins that undergo covalent labeling with small molecules through in vitro selection by using ribosome display. Selection was performed from a mutant library of the WW domain with a biotinylated peptide as its binding target, in which the biotin and the peptide are connected by a disulfide bond. After five rounds of selection, we identified mutants carrying a particular cysteine mutation. The binding target reacted specifically with the selected mutant, even in the presence of other proteins, and resulted in the generation of biotin- or peptide-labeled WW domains by thiol-disulfide exchange. When the mutant was fused to a protein of interest, the fusion protein was also labeled with biotin. Thus, the characteristics of the selected mutant should be suitable as a tag sequence that can be covalently labeled with small synthetic molecules. These results indicate that the rapid and efficient generation of such proteins is possible by ribosome display.  相似文献

One of the most effective precipitation methods for submicrometer particles is the charged droplet scrubber. The dust collection in the charged droplet scrubber is considered to consist of some processes with respect to the interaction between charged dust particles and a charged water droplet. That is, the charged dust particles are attracted to, collide with, adhere to, penetrate into, rebound from, accumulate on, and reentrain from a charged water droplet. The direct observation of the collection process for dust particles by a charged water droplet was conducted by using the experimental setup designed by the authors. It was found from the experiments that particles with good wettability were captured inside the water droplet, particles with poor wettability floated at regular intervals on the droplet surface, and particles with medium wettability were trapped on or inside the water droplet. When particles were trapped on the droplet surface the growth of dendritic depositions was found, and their behavior was very distinct from those obtained on the fibrous filter due to the fluidity of the droplet surface. The deposition mechanisms of charged particles on the charged water droplet are discussed qualitatively.  相似文献

In cancer therapy, radioresistance or chemoresistance cells are major problems. We established clinically relevant radioresistant (CRR) cells that can survive over 30 days after 2 Gy/day X-ray exposures. These cells also show resistance to anticancer agents and hydrogen peroxide (H2O2). We have previously demonstrated that all the CRR cells examined had up-regulated miR-7-5p and after miR-7-5p knockdown, they lost radioresistance. However, the mechanism of losing radioresistance remains to be elucidated. Therefore, we investigated the role of miR-7-5p in radioresistance by knockdown of miR-7-5p using CRR cells. As a result, knockdown of miR-7-5p increased reactive oxygen species (ROS), mitochondrial membrane potential, and intracellular Fe2+ amount. Furthermore, miR-7-5p knockdown results in the down-regulation of the iron storage gene expression such as ferritin, up-regulation of the ferroptosis marker ALOX12 gene expression, and increases of Liperfluo amount. H2O2 treatment after ALOX12 overexpression led to the enhancement of intracellular H2O2 amount and lipid peroxidation. By contrast, miR-7-5p knockdown seemed not to be involved in COX-2 and glycolysis signaling but affected the morphology of CRR cells. These results indicate that miR-7-5p control radioresistance via ROS generation that leads to ferroptosis.  相似文献

Carbonated autoclaved aerated concretes (AACs) show no shrinkage at a degree of carbonation approximately less than 20%. The 29Si MAS NMR spectrum showed that at a degree of carbonation less than 25%, the typical double-chain silicate anion structure of tobermorite-11Å was well maintained and interlayer Ca ions were exchanged with protons. This corresponded to the absence of carbonation shrinkage at a degree of carbonation less than 20%. When the degree of carbonation increased from 25% to 50% up to 60%, the double-chain silicate anion structure of tobermorite-11Å was decomposed and Ca ions in the Ca-O layers were dissolved, showing a possible mechanism of carbonation shrinkage.  相似文献

Methanosarcina species pyrrolysyl‐tRNA synthetase (PylRS) attaches Pyl to its cognate amber suppressor tRNA. The introduction of two mutations (Y384F and Y306A) into PylRS was previously shown to generate a mutant, designated LysZ‐RS, that was able to attach N‐benzyloxycarbonyl‐L ‐lysine (LysZ) to its cognate tRNA. Despite the potential of LysZ derivatives, further LysZ‐RS engineering has not been performed; consequently, we aimed to generate LysZ‐RS mutants with improved LysZ incorporation activity through in vitro directed evolution. Using a liposome‐based in vitro compartmentalization (IVC) approach, we screened a randomly mutagenized gene library of LysZ‐RS and obtained a mutant that showed increased LysZ incorporation activity both in vitro and in vivo. The ease and high flexibility of liposome‐based IVC should enable the evolution of not only LysZ‐RS that can attach various LysZ derivatives but also of other enzymes involved in protein translation.  相似文献

Sodium niobate NaNbO3 hydrate (NN‐hydrate) particles with a plate‐like morphology were prepared at 140°C for 2 h in 12 mol/L of NaOH by the hydrothermal method. Bar‐like Na8Nb6O19·13H2O particles were synthesized at as low as 100°C for 2 h. This work demonstrates that by carefully optimizing the reaction condition, we can selectively fabricate niobate structures, including the bar‐like, plate‐like, fibers and cube particles through a direct reaction between NaOH solution and Nb2O5. It was found that Nb6O198? formed was an important premise for formation of the NN‐hydrate, and lower [OH] was not favorable in preparing the NN‐hydrate as there was an optimum [OH?]. Through researching effects of the reaction temperature, time, concentration of NaOH, and content of Nb2O5 on the NN‐hydrate structure and evolution, the formation mechanism from solid reactants to the intermediate were investigated. After calcining at 800°C, the synthesized NN‐hydrate particles made a phase almost transform to the perovskite NaNbO3, and the morphology of these calcined particles was still plate‐like.  相似文献

Sodium niobate (NaNbO3) particles with plate‐like morphology and hexagonal unit cells were prepared by the hydrothermal method. The result of SEM showed that the hexagonal NaNbO3 were characterized by plate‐like morphology with a diameter of 5–15 μm and a thickness of 1–2 μm. The crucial influences on the morphology and crystal phase of the NaNbO3, such as concentration of [OH?], surfactant, and K+:Na+ ratio, were established. By further calcination treatment, the plate‐like hexagonal NaNbO3 particles could be completely transformed into perovskite structure without morphology change. The XRD and EBSD results indicate that the major face of the calcined particles is parallel to the crystallographic (001)pc (pseudo cubic index) plane. Compared with the traditional high‐temperature molten salt method, this work provides a simpler way to prepare the template for fabricating textured ceramics.  相似文献
