During 2006, 82 samples of human mature milk were collected at Italian hospitals and checked for aflatoxin M1 (AFM1) and ochratoxin A (OTA) by immunoaffinity column extraction and HPLC. AFM1 was detected in four (5%) of milk samples (ranging from < 7 ng/L to 140 ng/L; mean level: 55.35 ng/L); OTA was detected in 61 (74%) of milk samples (ranging from < 5 ng/L to 405 ng/L; mean level: 30.43 ng/L. OTA levels were significantly higher (p less, not double equals 0.05) in milk of habitual consumers of bread, bakery products and cured pork meat. No other statistically significant differences were observed although habitual consumers of pasta (p = 0.059), cookies (p = 0.061) and juices (p = 0.063) had mean contamination values of OTA higher than the moderate consumer. The very few AFB1 positive samples did not allow statistical comparisons. The present study confirms that the occurrence of OTA in human milk is related to maternal dietary habits. The findings support the possibility of dietary recommendations to woman, during pregnancy and lactation, aimed to tentatively reduce the OTA contamination of human milk.  相似文献

The acceptance of many foods is related to traditional cooking practices, which create taste and texture and are important to digestibility, preservation, and the reduction of foodborne illnesses. A wide range of compounds are formed during the cooking of foods, a number of these have been shown to lead to adverse effects in classical toxicological models and are known as food processing contaminants (FPC). It is essential that the presence and effects of such compounds alone and in combination within the diet are understood such that proportionate risk management measures can be developed, while taking a holistic view across the whole value chain. Furan and alkylfurans (principally 2‐ and 3‐methylfuran) are highly volatile FPC, which are formed in a wide range of foods at low amounts. The focus of research to‐date has been on those foods, which have been identified to be most consequential in terms of being sources of exposure, namely jarred and canned foods for infants and young children (meals and drinks) and coffee (roast and ground, soluble). This report presents (i) new industry data on the occurrence of furan and methylfurans in selected food categories following previous coffee studies, (ii) the most salient parameters that impact furan formation, and (iii) aspects of importance for the risk assessment.  相似文献

The paper proposes a decision support system (DSS) for the supply chain of packaged fresh and highly perishable products. The DSS combines a unique tool for sales forecasting with order planning which includes an individual model selection system equipped with ARIMA, ARIMAX and transfer function forecasting model families, the latter two accounting for the impact of prices. Forecasting model parameters are chosen via two alternative tuning algorithms: a two-step statistical analysis, and a sequential parameter optimisation framework for automatic parameter tuning. The DSS selects the model to apply according to user-defined performance criteria. Then, it considers sales forecasting as a proxy of expected demand and uses it as input for a multi-objective optimisation algorithm that defines a set of non-dominated order proposals with respect to outdating, shortage, freshness of products and residual stock. A set of real data and a benchmark – based on the methods already in use – are employed to evaluate the performance of the proposed DSS. The analysis of different configurations shows that the DSS is suitable for the problem under investigation; in particular, the DSS ensures acceptable forecasting errors and proper computational effort, providing order plans with associated satisfactory performances.  相似文献

Thioethers, sulfoxides, and sulfonium ions, despite diverse physicochemical properties, all engage in noncovalent interactions with proteins. Thioether‐containing macrocycles are also attracting attention as protein–protein interaction (PPI) inhibitors. Here, we used a model PPI between α‐helical mixed lineage leukemia (MLL) protein and kinase‐inducible domain interacting (KIX) domain to evaluate oxidation effects on sulfurcontaining macrocycle structure, stability, and protein affinity. Desolvation effects from various polarity states were evaluated computationally and experimentally at the side chain, amino acid, and peptide level. Sulfur‐containing side chains spanned polarity ranges between all‐hydrocarbon and lactam bridges for modulating solubility, cellular uptake, and affinity. Helical propensity studies showed that, although oxidized sulfur‐containing side chains could be tolerated, conformational effects were sequence‐dependent. In some cases, proteolytic stability, binding capacity with KIX, and increased helicity were obtained as first steps toward developing PPI inhibitors.  相似文献

Although the relationship between polyamines and photosynthesis has been investigated at several levels, the main aim of this experiment was to test light-intensity-dependent influence of polyamine metabolism with or without exogenous polyamines. First, the effect of the duration of the daily illumination, then the effects of different light intensities (50, 250, and 500 μmol m–2 s–1) on the polyamine metabolism at metabolite and gene expression levels were investigated. In the second experiment, polyamine treatments, namely putrescine, spermidine and spermine, were also applied. The different light quantities induced different changes in the polyamine metabolism. In the leaves, light distinctly induced the putrescine level and reduced the 1,3-diaminopropane content. Leaves and roots responded differently to the polyamine treatments. Polyamines improved photosynthesis under lower light conditions. Exogenous polyamine treatments influenced the polyamine metabolism differently under individual light regimes. The fine-tuning of the synthesis, back-conversion and terminal catabolism could be responsible for the observed different polyamine metabolism-modulating strategies, leading to successful adaptation to different light conditions.  相似文献

The multiple uses of the Danube River for navigation, hydropower production and as a drinking water supply influence its ecological quality. Flow restrictions due to regulation as well as impacts on the sediment regime such as retention in the catchment area and interruption of the sediment continuum result in changes of the hydromorphological structures and can produce major problems in the context of sediment management. In light of these facts, cross-border investigations of sediment transport at the Danube River are called for. As there are no uniform research approaches shared by Austria and Hungary, the EU-funded project SEDDON (Sediment Research and Management at the Danube River) aims to compare different monitoring and modeling techniques used to determine sediment transport, and to develop mutual solutions. Further, the laboratory equipment currently used for hydraulic model experiments is evaluated and a research channel with a free flowing discharge of up to 10 m3/s without pumping will be constructed, allowing large-scale joint laboratory tests concerning sediment to be performed.  相似文献

Testing accounts for a relevant part of the production cost of complex or critical software systems. Nevertheless, time and resources budgeted to testing are often underestimated with respect to the target quality goals. Test managers need engineering methods to perform appropriate choices in spending testing resources, so as to maximize the outcome. We present a method to dynamically allocate testing resources to software components minimizing the estimated number of residual defects and/or the estimated residual defect density. We discuss the application to a real-world critical system in the homeland security domain. We describe a support tool aimed at easing industrial technology transfer by hiding to practitioners the mathematical details of the method application.  相似文献
