Although pollen structure and morphology evolved toward the optimization of stability and fertilization efficiency, its performance is affected by harsh environmental conditions, e.g., heat, cold, drought, pollutants, and other stressors. These phenomena are expected to increase in the coming years in relation to predicted environmental scenarios, contributing to a rapid increase in the interest of the scientific community in understanding the molecular and physiological responses implemented by male gametophyte to accomplish reproduction. Here, after a brief introduction summarizing the main events underlying pollen physiology with a focus on polyamine involvement in its development and germination, we review the main effects that environmental stresses can cause on pollen. We report the most relevant evidence in the literature underlying morphological, cytoskeletal, metabolic and signaling alterations involved in stress perception and response, focusing on the final stage of pollen life, i.e., from when it hydrates, to pollen tube growth and sperm cell transport, with these being the most sensitive to environmental changes. Finally, we hypothesize the molecular mechanisms through which polyamines, well-known molecules involved in plant development, stress response and adaptation, can exert a protective action against environmental stresses in pollen by decoding the essential steps and the intersection between polyamines and pollen tube growth mechanisms.  相似文献

This work was aimed to determine the effect of fermentation and drying on the content and profile of procyanidins (from monomers P1 to polymers P10) as well as on the antiradical and scavenging properties of cocoa beans. To this purpose, three experiments were carried out: a traditional fermentation process followed by air drying and two pilot-scale fermentation processes by either natural microbiota or starter followed by sun drying. Procyanidins were evaluated by HPLC analysis, while the total polyphenol index (TPI), the antiradical activity as well as the reducing power were determined by means of the reaction with the Folin–Ciocalteu reagent, the decolorization assays of the ABTS radical (TEAC) and the Ferric Reducing Antioxidant Power (FRAP) methods, respectively. Both the traditional and pilot-scale processes resulted to affect the profile and content of the procyanidins fractions as well as the antiradical and reducing power functionality. Drying caused a severe reduction of compounds and thus resulted to be the critical step for the loss of procyanidins and monomers in particular. The indices of functionality generally showed a decreasing trend as a consequence of processing, and their evolution was similar to that observed in procyanidins content. To study the relationship between the individual procyanidins and the antioxidant activity expressed as TEAC, FRAP and TPI, the data set were processed by modified partial least squares regression. The obtained models presented a good predictive ability. Normalised regression coefficients showed that the relative contribution of each single class of compounds to total antioxidant activity resulted as follows: P1?>?P2?>?P3?>?P4?>?P6?>?P8?>?P5?>?P7?>?P9?>>?P10.  相似文献

Abstract

The aim of this paper is to provide an analytical determination of temperature and thermal stress in a cylindrical nuclear fuel rod with cladding, including the actual neutron flux distribution in a unit cell, fur a thermal reactor in steady state. The temperature and stresses are evaluated by solving a set of differential and integral equations, derived from the heat conduction and Hooke's law. Some numerical applications are presented (with reference to a common pressurized water reactor) and discussed. The emphasis is placed on the influence of nuclear parameters on the thermal and mechanical behavior of the fuel element.  相似文献

The angular dependence of the thermoluminescent (TL) signal of thin alpha-Al2O3:C dosemeters was investigated for a series of beta-emitting radionuclides commonly employed in nuclear medicine and characterised by different mean energies (99Tc, 177Lu, 90Sr/90Y and 90Y). Irradiations were performed in a controlled geometry, using a properly designed irradiator intended to realistically reproduce the situation of exposure of hospital personnel to beta-emitting pharmaceuticals. Under the conditions of extended source and short source to detector distance, the TL signal of thin alpha-Al2O3:C layers per unit irradiation time was observed to be independent on the angle of incidence within acceptable limits, particularly for those radionuclides with maximum energy >500 keV. This property may be easily explained by using simple physical considerations, such as the limited thickness of the dosemeters. The results confirm that these detectors are suitable for beta-ray extremity dose measurements, when the photon contribution is negligible, as in the case considered.  相似文献

Although adaptive control has been used in numerous applications, the ability to obtain a predictable transient and steady‐state closed‐loop performance is still a challenging problem from the verification and validation standpoint. To that end, we considered a recently developed robust adaptive control methodology called low‐frequency learning adaptive control and utilized a set of theoretic analysis to show that the transitory performance of this approach can be expressed, analyzed, and optimized via a convex optimization problem based on linear matrix inequalities. This key feature of this design and analysis framework allows one to tune the adaptive control parameters rigorously so that the tracking error components of the closed‐loop nonlinear system evolve in a priori specified region of the state space whose size can be minimized by selecting a suitable cost function. Simulation examples are provided to demonstrate the efficacy of the proposed verification and validation architecture showing the possibility of performing parametric studies to analyze the interplay between the size of the tracking error residual set and important design parameters such as the adaptation rate and the low‐pass filters time constant of the weights adaptation algorithm. A collection of carbohydrate‐derived iminosugars belonging to three structurally diversified sub‐classes (polyhydroxylated pyrrolidines, piperidines, and pyrrolizidines) was evaluated for inhibition of human acid β‐glucosidase (glucocerebrosidase, GCase), the deficient enzyme in Gaucher disease. The synthesis of several new pyrrolidine analogues substituted at the nitrogen or α‐carbon atom with alkyl chains of different lengths suggested an interpretation of the inhibition data and led to the discovery of two new GCase inhibitors at sub‐micromolar concentration. In the piperidine iminosugar series, two N‐alkylated derivatives were found to rescue the residual GCase activity in N370S/RecNcil mutated human fibroblasts (among which one up to 1.5‐fold). This study provides the starting point for the identification of new compounds in the treatment of Gaucher disease.  相似文献

Mussel‐inspired polydopamine (PD) coating represents a promising route for constructing functional materials and finely tuning or completely changing their surface properties. In this work, a mild and effective method to realize the deposition of PD on keratin‐based films is reported. Reactive oxygen species (ROS), generated by keratin films doped with a photosensitizer, e.g., Azure A (AzA), upon UV–vis and vis‐near IR irradiation, are exploited to obtain the PD coating. Interestingly, the use of vis‐near IR irradiation leads to an increased production of ROS by AzA resulting in a greater PD deposition. Compared to uncoated keratin films, the PD‐keratin coated materials show a granular but homogeneous surface and an increased hydrophilicity, maintaining the ROS generation ability of AzA. This work demonstrates a useful method to effectively and rapidly functionalize with PD materials that are sensitive to temperature, pH and UV light, such as keratin. The proposed strategy allows obtaining new multifunctional biomaterials of potential interest in tissue engineering and drug delivery; in particular, the use of PD‐keratin films for near IR laser bonding of ophthalmic tissue, which is a representative case in wound healing purpose, is shown.  相似文献

A complex interaction between genetic and external factors determines the development of amyotrophic lateral sclerosis (ALS). Epidemiological studies on large patient cohorts have suggested that ALS is a multi-step disease, as symptom onset occurs only after exposure to a sequence of risk factors. Although the exact nature of these determinants remains to be clarified, it seems clear that: (i) genetic mutations may be responsible for one or more of these steps; (ii) other risk factors are probably linked to environment and/or to lifestyle, and (iii) compensatory plastic changes taking place during the ALS etiopathogenesis probably affect the timing of onset and progression of disease. Current knowledge on ALS mechanisms and therapeutic targets, derives mainly from studies involving superoxide dismutase 1 (SOD1) transgenic mice; therefore, it would be fundamental to verify whether a multi-step disease concept can also be applied to these animal models. With this aim, a meta-analysis study has been performed using a collection of primary studies (n = 137), selected according to the following criteria: (1) the studies should employ SOD1 transgenic mice; (2) the studies should entail the presence of a disease-modifying experimental manipulation; (3) the studies should make use of Kaplan–Meier plots showing the distribution of symptom onset and lifespan. Then, using a subset of this study collection (n = 94), the effects of treatments on key molecular mechanisms, as well as on the onset and progression of disease have been analysed in a large population of mice. The results are consistent with a multi-step etiopathogenesis of disease in ALS mice (including two to six steps, depending on the particular SOD1 mutation), closely resembling that observed in patient cohorts, and revealed an interesting relationship between molecular mechanisms and disease manifestation. Thus, SOD1 mouse models may be considered of high predictive value to understand the determinants of disease onset and progression, as well as to identify targets for therapeutic interventions.  相似文献

Infertility is a worldwide health issue defined by the World Health Organization (WHO) as the inability to establish a pregnancy after 12 months or more of regular and unprotected sexual intercourse. Male infertility etiology can be related to either congenital or acquired factors. The therapeutical approach to male infertility depends on the underlying causes and includes medical and surgical treatments. In recent studies, the potential role of nerve growth factor (NGF) in male reproductive physiology has been proposed. It has been hypothesized that neurotrophins might be involved in testis morphogenesis and regulation of several aspects of spermatogenesis. Moreover, it has been shown that NGF exerts its role on gonadotropin-releasing hormone (GnRH) neurons through the activation of the PKC/p–ERK1/2/p–CREB cascade, which leads to the activation of hypothalamic cells and the consequent activation of hypothalamus–pituitary–gonadal axis (HPG) with the secretion of GnRH. Lastly, it has been shown that the physiology of mature sperm is affected by both exogenous and endogenous NGF. The NGF impact on the HPG axis and its effect on GnRH neurons might be exploited in the therapy of male hypogonadism or used as a protective strategy against gonadal dysfunction related to chemotherapeutic agents. Moreover, the improving effect of NGF on sperm motility and vitality could be useful to enhance assisted reproduction outcomes. NGF could be supplemented to cryopreserved sperm samples to counteract the oxidative stress induced by the frozen and thawing processes. Indeed, the potential clinical applications of NGF in male infertility treatment have been discussed.  相似文献
