Four different spices produced in Mexico, namely black pepper, two kinds of chilli and oregano, were studied by electron paramagnetic resonance (EPR) technique for detection purposes. In all the non‐irradiated spices we tested, the same single line signal at g = 2.0041 (native signal) was registered. Irradiation induced at least two EPR signals overlapping the native signal: an intense singlet and, with the exclusion of oregano, a weak triplet with hyperfine splitting of 3 mT due to cellulose free radicals. The recombination time of the free radicals produced by irradiation was short compared with the shelf‐life of the spices, thus limiting the application of the EPR technique for detection. To extend the time when these products may be identified as irradiated, the method proposed by Yordanov and Gancheva (Appl Radiat Isot 2000; 52 :195) was applied to the spices, but with no success. Thermoluminescence tests provided correct results for all the spices, even 2 years after irradiation.  相似文献

Pulsed electron‐beam irradiation of a semi‐dilute poly(N‐vinyl pyrrolidone) (PVP) aqueous solution in the presence of acrylic acid has led to a carboxyl functionalized nanogel system. Nanoparticles hydrodynamic size and surface charge density, in water and as a function of pH, were investigated by dynamic light scattering and laser doppler velocimetry, respectively. Nanogels (NGs) were proved not to be cytotoxic at the cellular level. Indeed, they rapidly bypass the cellular membrane to accumulate in specific cell portions of the cytoplasm, in the perinuclear area. The availability of pendant carboxyl groups on the crosslinked PVP NGs core prompted us to attempt their decoration with a single strand oligonucleotide, which holds a terminal amino group. Abstract

Multi-quantum well Si/Ge nanowires (NWs) were realized by combining molecular beam epitaxy deposition and metal-assisted wet etching, which is a low-cost technique for the synthesis of extremely dense (about 1011 cm−2) arrays of NWs with a high and controllable aspect ratio. In particular, we prepared ultrathin Si/Ge NWs having a mean diameter of about 8 nm and lengths spanning from 1.0 to 2.7 μm. NW diameter is compatible with the occurrence of quantum confinement effects and, accordingly, we observed light emission assignable to the presence of Si and Ge nanostructures. We performed a detailed study of the photoluminescence properties of the NWs, with particular attention to the excitation and de-excitation properties as a function of the temperature and of the excitation photon flux, evaluating the excitation cross section and investigating the presence of non-radiative phenomena.

Dairy products are characterized by reduced shelf life because they are an excellent growth medium for a wide range of microorganisms. For this reason, it is important to monitor the microbiological quality of dairy products and, in particular, the total viable count and concentration of Escherichia coli, as they are indicators of the hygienic state of these products. In addition, in dairy products such as Mozzarella cheese, it is important to monitor the concentration of lactic acid bacteria (LAB), as they are the major components of starter cultures used in cheese production, contributing to the taste and texture of fermented products and inhibiting food spoilage bacteria by producing growth-inhibiting substances. For these reasons, to ensure the quality and safety of their products, cheese makers should monitor frequently, during fresh cheese production, the concentration of LAB and spoilage bacteria. However, usually, small- to medium-size dairy factories do not have an internal microbiological laboratory and external laboratories of analysis are often too expensive and require several days for the results. Compared with traditional methods, the microbiological survey (MBS) method developed by Roma Tre University (Rome, Italy) allows faster and less-expensive microbiological analyses to be conducted wherever they are necessary, without the need for a microbiological laboratory or any instrumentation other than MBS vials and a thermostat. In this paper, we report the primary validation of the MBS method to monitor LAB concentration in Mozzarella cheese and the analysis, using the MBS method, of total viable count, E. coli, and LAB concentrations in the production line of Mozzarella cheese as well as during the shelf life of the product stored at 20°C. The results obtained indicate that the MBS method may be successfully used by small- to medium-size dairy factories that do not have an internal microbiological laboratory. Using the MBS method, these dairy factories can monitor autonomously the microbiological safety and quality of their products, saving both time and money.  相似文献

Hypercholesterolemia is one of the major causes of cardiovascular disease, the risk of which is further increased if other forms of dyslipidemia occur. Current therapeutic strategies include changes in lifestyle coupled with drug administration. Statins represent the most common therapeutic approach, but they may be insufficient due to the onset of resistance mechanisms and side effects. Consequently, patients with mild hypercholesterolemia prefer the use of food supplements since these are perceived to be safer. Here, we investigate the phytochemical profile and cholesterol-lowering potential of Protium heptaphyllum gum resin extract (PHE). Chemical characterization via HPLC-APCI-HRMS2 and GC-FID/MS identified 13 compounds mainly belonging to ursane, oleanane, and tirucallane groups. Studies on human hepatocytes have revealed how PHE is able to reduce cholesterol production and regulate the expression of proteins involved in its metabolism. (HMGCR, PCSK9, LDLR, FXR, IDOL, and PPAR). Moreover, measuring the inhibitory activity of PHE against HMGR, moderate inhibition was recorded. Finally, molecular docking studies identified acidic tetra- and pentacyclic triterpenoids as the main compounds responsible for this action. In conclusion, our study demonstrates how PHE may be a useful alternative to contrast hypercholesterolemia, highlighting its potential as a sustainable multitarget natural extract for the nutraceutical industry that is rapidly gaining acceptance as a source of health-promoting compounds.  相似文献

Pompe disease is an autosomal recessive disorder caused by a deficiency in the enzyme acid alpha-glucosidase. The late-onset form of Pompe disease (LOPD) is characterized by a slowly progressing proximal muscle weakness, often involving respiratory muscles. In LOPD, the levels of GAA enzyme activity and the severity of the clinical pictures may be highly variable among individuals, even in those who harbour the same combination of GAA mutations. The result is an unpredictable genotype–phenotype correlation. The purpose of this study was to identify the genetic factors responsible for the progression, severity and drug response in LOPD. We report here on a detailed clinical, morphological and genetic study, including a whole exome sequencing (WES) analysis of 11 adult LOPD siblings belonging to two Italian families carrying compound heterozygous GAA mutations. We disclosed a heterogeneous pattern of myopathic impairment, associated, among others, with cardiac defects, intracranial vessels abnormality, osteoporosis, vitamin D deficiency, obesity and adverse response to enzyme replacement therapy (ERT). We identified deleterious variants in the genes involved in autophagy, immunity and bone metabolism, which contributed to the severity of the clinical symptoms observed in the LOPD patients. This study emphasizes the multisystem nature of LOPD and highlights the polygenic nature of the complex phenotype disclosed in these patients.  相似文献

Traumatic peripheral nerve lesions affect hundreds of thousands of patients every year; their consequences are life-altering and often devastating and cause alterations in movement and sensitivity. Spontaneous peripheral nerve recovery is often inadequate. In this context, nowadays, cell therapy represents one of the most innovative approaches in the field of nerve repair therapies. The purpose of this systematic review is to discuss the features of different types of mesenchymal stem cells (MSCs) relevant for peripheral nerve regeneration after nerve injury. The published literature was reviewed following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines. A combination of the keywords “nerve regeneration”, “stem cells”, “peripheral nerve injury”, “rat”, and “human” were used. Additionally, a “MeSH” research was performed in PubMed using the terms “stem cells” and “nerve regeneration”. The characteristics of the most widely used MSCs, their paracrine potential, targeted stimulation, and differentiation potentials into Schwann-like and neuronal-like cells are described in this paper. Considering their ability to support and stimulate axonal growth, their remarkable paracrine activity, their presumed differentiation potential, their extremely low immunogenicity, and their high survival rate after transplantation, ADSCs appear to be the most suitable and promising MSCs for the recovery of peripheral nerve lesion. Traditional milk coagulation properties are used to predict the suitability of milk for cheese-making. In bovine and ovine species, the introduction of the concept of curd firming over time, continuously recorded by a lactodynamograph during prolonged tests, provides additional information about milk coagulation, curd-firming, and syneresis processes. The aims of present study were (1) to test the adaptability of a 4-parameter curd-firming model in the assessment of goat milk (also comparing published data of other species); (2) to describe variability of coagulation, curd firming, and syneresis processes among individual goat milk samples; (3) to quantify the effects of farm and animal factors (breed, parity, and stage of lactation); and (4) to compare 6 goat breeds for their model parameters. Milk samples from 1,272 goats reared in 35 farms were collected. Goats were of 6 breeds: Saanen and Camosciata delle Alpi for the Alpine type; and Murciano-Granadina, Maltese, Sarda, and Sarda Primitiva for the Mediterranean type. During a lactodynamographic analysis (60 min), 240 measures of curd firmness (mm) were recorded for each milk sample. The modeling of curd firming allowed us to achieve the rennet coagulation time estimated on the basis of all the data points (min); the curd firming and the curd syneresis instant rate constants; the asymptotical potential value of curd firming; the actual maximum curd firmness; and the time at which the curd firming maximum level is attained. Modeling parameter data were analyzed using a linear mixed model. Comparison with other dairy species showed several differences: goat milk coagulated later than sheep but earlier than bovine, and curd firming and curd syneresis instant rate constants were greater in small ruminants. Modeling parameters of goat milk were mostly affected by the farm effect (37% of the total variance, on average) compared with the results found for bovine and ovine samples, and this was probably attributable to the marked differences among goat farming systems. Small differences were demonstrated between Alpine and Mediterranean breeds, but the time of maximum curd firmness was lower in Murciano-Granadina compared with Maltese, Sarda, and Sarda Primitiva. Sarda and Sarda Primitiva were very similar and exhibited the most favorable coagulation properties of milk. For almost all the model parameters, the direct effect of breed was increased after correction for milk yield and composition. In conclusion, this approach allowed us to fully depict the effects of the different factors on coagulation of goat milk, and clarified the different renneting pattern among goat breeds, and with other species. Results could be used for the valorization of goat dairy products, also when these are linked to particular local breeds, and to stimulate further studies about relationships between coagulation and cheese-making traits.  相似文献
