An efficient and simple method for determination of methyl anthranilate (MA) in Citrus spp. honeys by headspace-solid-phase microextraction-gas chromatography (HS-SPME-GC) was developed and validated. Experimental design was used to investigate the effects of the principal extraction parameters. The central composite design (CCD) and the desirability function were used to find the experimental conditions providing the optimal HS-SPME result. Validation was carried out in terms of specificity, linearity, limit of detection (LOD) and quantitation (LOQ) (0.149 and 0.324 μg/g, respectively), method precision (RSD 7%), LOQ precision (RSD 6.5%), and was resulted accurate and robust. Finally, the applicability of the method to the determination of MA in a number of commercial Citrus spp. honey samples was demonstrated, the content ranged from 0.63 to 3.26 μg/g.  相似文献

BACKGROUND: Seven ionic liquids (ILs) based on 1-alkyl-3-methylimidazolium cation in combination with hexafluorophosphate and bis{(trifluoromethyl)sulfonyl}imide anions were tested as reaction media for lipase-catalyzed transesterification in low water conditions. With the aim of improving the activity and/or selectivity of the lipase, various treatments were applied to ionic liquid media such as equilibration with aqueous solutions of salts, NaHCO3 or Na2CO3, or the addition of a catalytic amount of a non-reactive organic base to the reaction mixture, triethylamine. RESULTS: The treated ionic liquids were shown to be excellent media for lipase-catalyzed ester synthesis by transesterification compared with conventional organic solvents, such as n-hexane. All treatments were found to enhance the synthetic activity of the enzyme, the best results being achieved with the addition of triethylamine. The addition of a catalytic amount of this base to the ILs resulted in a significant increase in both the synthetic activity and selectivity values. For instance, the synthetic activity in [emim+][TfN2] was enhanced more than 12 times and the selectivity increased from 86% to 95% when triethylamine was used. CONCLUSION: These treatments could be easy-to-use approaches to improve the efficiency of enzymatic reactions in ionic liquids when the reaction does not proceed smoothly. Cameroon is blessed with abundant water resources. Rapid population increase, unplanned urbanisation, intensive industrial and socio-economic development have led to poor and unsustainable management of these resources. Integrated Water Resources Management (IWRM) is a promising approach in ensuring sustainable management of Cameroon’s water resources. It entails management of water for various purposes and not for a single purpose which therefore involves different stake holders aiming at achieving sustainable water resources management. This paper seeks to evaluate recent efforts to implement in IWRM in Cameroon by examining the institutional framework for IWRM in Cameroon, conditions for the implementation of IWRM and proposes reforms for improving IWRM in Cameroon. The paper concludes that reforms such as public participation at local council levels, recognition of water as both an economic and a social good, putting IWRM within the larger context of Integrated Natural Resource Management (INRM) and the exploitation of mathematical models within hydrological basins will improve IWRM in Cameroon.  相似文献

The effect of treatment with α‐galactosidase, tannase or a cell‐wall‐degrading enzyme complex under optimal conditions of pH, temperature and length of incubation time on the chemical composition and nutritive utilisation of protein and carbohydrates from pea (Pisum sativum L.) flour was studied. Soaking of pea flours in combination with enzyme treatment led to reductions of 77–90% in the levels of α‐galactosides, and of 60–80% in the levels of trypsin inhibitor activity, increasing the content of total available sugars, which was highest in the pea flour treated with the cell‐wall‐degrading enzyme complex. All the treatments assayed caused a significant improvement in daily food intake, whereas the nutritive utilisation of protein was not increased in any of the pea products tested when compared to the raw pea flour. However, all the soaking and enzymatic treatments led to a significant improvement in daily weight gain associated with a higher dietary intake of food and total available sugars. 27 Black college students viewed 3 excerpts showing racist situations involving Blacks; anger-provoking, nonracist situations; and neutral situations. After each scene, blood pressure (BP) was taken; a mood checklist, the Framingham Anger Scale (S. Haynes et al; see PA, Vol 68:10702), and the Anger Expression Scale of C. Spielberger et al (1985) were administered. Analyses revealed that BP significantly increased during the presentation of racist stimuli but not of anger-provoking or neutral stimuli. Self-reports of state anger, as measured by the mood checklist, were significant for both the anger-provoking and racist stimuli. BP scores were significantly correlated to the 2 trait anger measures. Exposure to racist stimuli was associated with BP increases among Blacks. Lipid stability of standard infant formula was evaluated at ambient temperatures, namely 25, 30 and 37 °C, during 3 months. Lipids were thoroughly analysed to evaluate changes in fatty acid composition and trans fatty acid isomers, non‐volatile oxidation compounds including oxidised, dimeric and polymeric triacylglycerols, and tocopherol. No significant changes in either of the parameters examined were found in total lipids extracted from infant formula along the storage period. However, the minor free oil fractions (about 7.5% of total lipids) showed a significant increase in oxidation compounds and marked decrease in tocopherol levels during storage at all temperatures. Samples stored at 37 °C for 3 months were rancid and, accordingly, contained the highest oxidation level in the free oil fraction, whereas total lipids extracted were apparently not oxidised. Results showed the necessity of analysing separately the free oil fraction in infant formulae to obtain a clear picture of the oxidation status.  相似文献

A monomeric version of triosephosphate isomerase from Trypanosomabrucei, MonoTIM, has very low activity, and the same is truefor all of the additional monomeric variants so far constructed.Here, we subjected MonoTIM to directed evolution schemes toachieve an activity improvement. The construction of a suitablestrain for genetic selection provided an effective way to obtainactive catalysts from a diverse population of protein variants.We used this tool to identify active mutants from two differentstrategies of mutagenesis: random mutagenesis of the whole geneand randomization of loop 2. Both strategies converged in theisolation of mutations Ala43 to Pro and Thr44 to either Alaor Ser, when randomizing the entire gene or to Arg in the caseof randomization of loop 2. The kinetic characterization ofthe two more active mutants showed an increase of 11-fold inkcat and a reduction of 4-fold in Km for both of them, demonstratingthe sensitivity of the selection method. A small differencein growth rate is observed when both mutant genes are compared,which seems to be attributable to a difference in solubilityof the expressed proteins.  相似文献

Herein we describe a class of unconventional nucleosides (methyloxynucleosides) that combine unconventional nucleobases such as substituted aminopyrimidines, aminopurines, or aminotriazines with unusual sugars in their structures. The allitollyl or altritollyl derivatives were pursued as ribonucleoside mimics, whereas the tetrahydrofuran analogues were pursued as their dideoxynucleoside analogues. The compounds showed poor, if any, activity against a broad range of RNA and DNA viruses, including human immunodeficiency virus (HIV). This inactivity may be due to lack of an efficient metabolic conversion into their corresponding 5′‐triphosphates and poor affinity for their target enzymes (DNA/RNA polymerases). Several compounds showed cytostatic activity against proliferating human CD4+ T‐lymphocyte CEM cells and against several other tumor cell lines, including murine leukemia L1210 and human prostate PC3, kidney CAKI‐1, and cervical carcinoma HeLa cells. A few compounds were inhibitory to Moloney murine sarcoma virus (MSV) in C3H/3T3 cell cultures, with the 2,6‐diaminotri‐O‐benzyl‐D ‐allitolyl‐ and ‐D ‐altritolyl pyrimidine analogues being the most potent among them. This series of unconventional nucleosides may represent a novel family of potential antiproliferative agents.  相似文献

Apparent rate constants for the reactions of four selected pharmaceutical compounds (metoprolol, naproxen, amoxicillin, and phenacetin) with chlorine in ultra-pure (UP) water were determined as a function of the pH. It was found that amoxicillin (in the whole pH range 3-12), and naproxen (in the low pH range 2-4) presented high reaction rates, while naproxen (in the pH range 5-9), and phenacetin and metoprolol (in the pH range 2.5-12 for phenacetin, and 3-10 for metoprolol) followed intermediate and slow reaction rates. A mechanism is proposed for the chlorination reaction, which allowed the evaluation of the intrinsic rate constants for the elementary reactions of the ionized and un-ionized species of each selected pharmaceutical with chlorine. An excellent agreement is obtained between experimental and calculated rate constants by this mechanism.The elimination of these substances in several waters (a groundwater, a surface water from a public reservoir, and two effluents from municipal wastewater treatment plants) was also investigated at neutral pH. The efficiency of the chlorination process with respect to the pharmaceuticals elimination and the formation THMs was also established. It is generally observed that the increasing presence of organic and inorganic matter in the water matrices demand more oxidant agent (chlorine), and therefore, less chlorine is available for the oxidation of these compounds. Finally, half-life times and oxidant exposures (CT) required for the removal of 99% of the four pharmaceuticals are also evaluated. These parameters are useful for the establishment of safety chlorine doses in oxidation or disinfection stages of pharmaceuticals in treatment plants.  相似文献

The objective of this study was to determine associated factors to the consumption of fruit and vegetables (FV) in the adult population of each socioeconomic status (SES) in Bucaramanga, Colombia. We conducted a cross-sectional in a sample of 317 people and we measured socio-demographic, economic and anthropometric variables and dietary factors related to the purchase and consumption of FV. We made a survey, a 24-Hour dietary recall and we did anthropometric measurements. Portion sizes were estimated with a standardized set of utensils and food-photography atlas. Frequencies, measures of central tendency and dispersion were established and Logistic Regression models were used for the bi and multivariate analysis. Average consumption of FV was 162.6 g/day (188.5 g in high SES and 145.7 g in low SES, p = 0.056). On average 2 servings per day of FV (2.3 servings in high SES and 1.5 in low SES, p = 0.055). The prevalence of adequate intake of FV (more than 5 servings / day) was 8.6% (8.9% in high SES and 8.4% in low SES, p = 0.896). The associated factor to consumption of FV was BMI > or =25 (OR: 0.31, CI (95%): 0.11-0.89, p = 0.030), adjusted for occupation, income, SES, and number of family members. FV consumption is low; excess weight is the only variable associated to no-consumption of five or more servings; causes: cost to the consumption of fruit and flavor in the case of vegetables.  相似文献
