Transforming growth factor-beta (TGF-β) is part of a family of molecules that is present in many body tissues and performs many different functions. Evidence has been obtained from mice and human cancer patients with bony metastases and non-metastatic disease, as well as pediatric burn patients, that inflammation leads to bone resorption and release of TGF-β from the bone matrix with paracrine effects on muscle protein balance, possibly mediated by the generation of reactive oxygen species. Whether immobilization, which confounds the etiology of bone resorption in burn injury, also leads to the release of TGF-β from bone contributing to muscle wasting in other conditions is unclear. The use of anti-resorptive therapy in both metastatic cancer patients and pediatric burn patients has been successful in the prevention of muscle wasting, thereby creating an additional therapeutic niche for this class of drugs. The liberation of TGF-β may be one way in which bone helps to control muscle mass, but further investigation will be necessary to assess whether the rate of bone resorption is the determining factor for the release of TGF-β. Moreover, whether different resorptive conditions, such as immobilization and hyperparathyroidism, also involve TGF-β release in the pathogenesis of muscle wasting needs to be investigated.  相似文献

Phase behaviour modelling of reservoir fluid is a fundamental step for reservoir simulation. Furthermore, as the complexity of the recovery process increases, the fluid model plays a more important role in the reliability of the simulation outputs. Although the in situ combustion enhanced oil recovery method (ISC) is one of the most complex recovery techniques available in the petroleum engineering literature, for most of the simulation jobs related to this elaborate process only simple and rudimentary fluid characterization layouts are considered. In this work, the principal fluid properties of Athabasca bitumen with regard to the ISC process are recognized, extracted from the literature, validated for consistency, and used for the development of an inclusive and accurate fluid model. Then the fluid model is fully developed while considering the ISC reaction kinetics so that the model has both accuracy, indispensable for phase behaviour modelling, and consistency, essential for the reactions definitions.  相似文献

The effect of various processing procedures on the composition and oxidative stability of coconut oil has been studied. The crude oil is relatively stable but major reductions in oxidative stability occur during the bleaching of oil degummed with phosphoric acid; during alkali refining; during the deodorization of oil degummed with citric acid and bleached; and during the deodorization of oil processed with a combined phosphoric acid degumming and bleaching operation. The reasons for the loss of oxidative stability during processing are discussed with reference to changes in the composition of the oil. Residual traces of citric acid or phosphoric acid play an important role in stabilizing processed oils. The tocopherol content is also important, although no additional stabilization of the oil occurs on adding levels of tocopherol above those present naturally in the crude oil. A combined phosphoric acid degumming and bleaching process leads to smaller losses of tocopherols than sequential treatments.  相似文献

A mutant of Lactobacillus casei dihydrofolate reductase hasbeen constructed in which Thr63, a residue which interacts withthe 2'-phosphate group of the bound coenzyme, is replaced byalanine. This substitution does not affect kcat, but producesan 800-fold increase in the Km for NADPH, which reflects dissociationof NADPH from the enzyme-NADPH-tetrahydrofolate complex, anda 625-fold increase (corresponding to 3.8 kcal/mol) in the dissociationconstant for the enzyme-NADPH complex. The difference in magnitudeof these effects indicates a small effect of the substitutionon the negative cooperativity between NADPH and tetrahydrofolate.Stopped-flow studies of the kinetics of NADPH binding show thatthe weaker binding arises predominantly from a decrease in theassociation rate constant. NMR spectroscopy was used to comparethe structures of the mutant and wild-type enzymes in solution,in their complexes with methotrexate and with methotrexate andNADPH. This showed that only minimal structural changes resultfrom the mutation; a total of 47 residues were monitored fromtheir resolved 1H resonances, and of these nine in the binarycomplex and six in the ternary differed in chemical shift betweenmutant and wild-type enzyme. These affected residues are confinedto the immediate vicinity of residue 63. There is a substantialdifference in the 31P chemical shift of the 2'-phosphate ofthe bound coenzyme, reflecting the loss of the interaction withthe side chain of Thr63. The only changes in nuclear Overhausereffects (NOEs) observed were decreases in the intensity of NOEsbetween protons of the adenine ring of the bound coenzyme andthe nearby residues Leu62 and Ile102, showing that the substitutionof Thr63 does cause a change in the position or orientationof the adenine ring in its binding site.  相似文献

A mixed culture of dye-decolorising Pseudomonas spp. was immobilised on polyurethane foam, and used in a bench-scale continuous culture bioreactor to treat textile effluent for colour removal. The provision of soluble wheat starch (0.2% w/v) to the culture medium promoted growth of biomass and enhanced decolorisation of effluent in the bioreactor. The bioprocess was further combined with membrane filtration technology to improve the quality of treated effluent. The treated effluent showed 90, 82 and 60% reduction in COD, BOD5 and toxicity levels, respectively, and had potential for re-use in dyeing processes. The colour difference of the cotton fabrics dyed using the treated effluent samples compared to those dyed using normal supply water was found to be industrially acceptable. The described process could improve treatment efficiency and allow water recycling within the textile factory.  相似文献

This work evaluates chronic safety in middle‐aged ovariectomized rats supplemented with different dosages of green tea polyphenols (GTP) in drinking water. The experiment used 6‐mo‐old sham (n = 39) and ovariectomized (OVX, n = 143) female rats. All sham (n = 39) and 39 of the OVX animals received no GTP treatment and their samples were collected for outcome measures at baseline, 3 mo, and 6 mo (n = 13 per group for each). The remaining OVX animals were randomized into 4 groups receiving 0.15%, 0.5%, 1%, and 1.5% (n = 26 for each) of GTP (wt/vol), respectively, in drinking water for 3 and 6 mo. No mortality or abnormal treatment‐related findings in clinical observations or ophthalmologic examinations were noted. No treatment‐related macroscopic or microscopic findings were noted for animals administered 1.5% GTP supplementation. Throughout the study, there was no difference in the body weight among all OVX groups. In all OVX groups, feed intake and water consumption significantly decreased with GTP dose throughout the study period. At 6 mo, GTP intake did not affect hematology, clinical chemistry, and urinalysis, except for phosphorus and blood urea nitrogen (increased), total cholesterol, lactate dehydrogenase, and urine pH (decreased). This study reveals that the no‐observed‐adverse‐effect level (NOAEL) of GTP is 1.5% (wt/vol) in drinking water, the highest dose used in this study.  相似文献

Riverine ecosystems have been altered in many large catchments by dam development to provide water, power, flood control and navigational benefits to humans. Conservation actions in these river ecosystems are commonly focused on minimum releases of water to downstream ecosystems. Increasingly minimum release approaches are being replaced with ‘experimental’ flows that mimic natural conditions in order to benefit riverine ecosystems. While these new policies are intuitive in their design, there is limited data of how riverine ecosystems actually respond to more natural flows. A test of more natural steady‐flow water release was compared with typical fluctuating hydropower flows in the adaptive management programme at Glen Canyon Dam, Arizona, during 2008–2011 to assess growth improvements of endangered juvenile humpback chub Gila cypha. Our results are counterintuitive and show that more natural steady flows reduced growth rates of juvenile humpback chub compared with fluctuating flows when both treatments occurred within the same year. Daily growth rates during steady flows of 2009 and 2010 were 0.05 and 0.07 mm day?1 slower, respectively, than fluctuating flows those same years, despite similar water temperatures. Juvenile humpback chub also grew more slowly during steady flows that occurred in the same season. During the summer, juvenile humpback chub grew 0.12 and 0.16 mm day?1 in fluctuating flow regimes in 2009 and 2010, respectively, and only 0.07 mm day?1 in the experimental steady flow regime in 2011, despite higher water temperatures. Our results suggest that optimal conservation management policies for endangered species in regulated rivers may not always be achieved with more natural flows.
