A monitoring program was undertaken to assess the microbial quality of greywater collected from 93 typical households in Melbourne, Australia. A total of 185 samples, comprising 75 washing machine wash, 74 washing machine rinse and 36 bathroom samples were analysed for the faecal indicator Escherichia coli. Of these, 104 were also analysed for genetic markers of pathogenic E coli and 111 for norovirus (genogroups GI and GII), enterovirus and rotavirus using RT-PCR. Enteric viruses were detected in 20 out of the 111 (18%) samples comprising 16 washing machine wash water and 4 bathroom samples. Eight (7%) samples were positive for enterovirus, twelve (11%) for norovirus genogroup GI, one (1%) for norovirus genogroup GII and another (1%) for rotavirus. Two washing machine samples contained more than one virus. Typical pathogenic E. coli were detected in 3 out of 104 (3%) samples and atypical enteropathogenic E. coli in 11 (11%) of samples. Levels of indicator E. coli were highly variable and the presence of E. coli was not associated with the presence of human enteric viruses in greywater. There was also little correlation between reported gastrointestinal illness in households and detection of pathogens in greywater. 相似文献

Lipids in almonds are present as oil bodies in the nut. These oil bodies are surrounded by a membrane of proteins and phospholipids and are a delivery vehicle of energy in the form of triglycerides, similarly to the more studied bovine milk fat globule membrane. Chemical, physical and microscopic analyses revealed major differences in the composition and structure of almond oil bodies and bovine milk fat globules. The lipids of both natural emulsions differed in degree of unsaturation, chain length, and class. The almond oil body membrane does not contain any cholesterol or sphingomyelin unlike the bovine milk fat globule membrane. Therefore, the phospholipid distribution at the surface of the oil bodies did not present any liquid-ordered domains. The membranes, a monolayer around almond oil bodies and a trilayer around bovine fat globules, may affect the stability of the lipid droplets in a food matrix and the way the lipids are digested. 相似文献

The most commonly used method for serotyping Salmonella spp. is based on the Kaufmann–White scheme, and is composed of serological reactions using antibodies to LPS agglutinins. The multiplex PCR used in this investigation was established by Kim et al. to serotype the 30 most common clinical Salmonella serotypes, as determined by CDC. The PCR assay consists of two five-plex reactions and a single two-plex PCR reaction, based on six genetic loci from Salmonella enterica serotype Typhimurium and four loci from S. enterica serotype Typhi. In this investigation, we further evaluated the method for serotyping Salmonella spp. using a reference collection, environmental samples collected from a Mid-Atlantic region tomato farm study, four food matrices spiked with different Salmonella serotypes and a proficiency test. The PCR assay was first evaluated using DNA isolated from pure cultures of isolates obtained from various clinical and environmental samples, and then DNA isolated from broth cultures of food matrices of “Red round” and Roma tomatoes, Romaine lettuce, green onions and Serrano peppers spiked with serotypes Newport, Typhimurium, Javiana and Saintpaul, respectively. The results showed that the PCR assay correctly serotyped Salmonella spp. from the clinical, environmental, spiked food matrices, and proficiency test samples. These findings are significant because the PCR assay was successful in the identification of Salmonella in the spiked samples in a broth culture containing other non-salmonella organism. The objectives of the present study were to compare the sulfur hexafluoride (SF6) and respiration chamber techniques for measuring methane (CH4) emissions from dairy cows and to determine the proportion of CH4 that is released through the rectum. Data used were derived from 20 early lactation dairy cows in a 2 × 2 factorial design study for 4 periods with 6 wk/period. The 4 treatment diets consisted of grass silage and 2 levels of concentrate (30 and 60% dry matter basis), with or without yeast supplement. At the end of each period, CH4 emissions were measured simultaneously using the SF6 and respiration chamber techniques when cows were housed in chambers. The SF6 technique was also used when cows were housed in digestibility units (barn location) before and after respiratory chamber measurements (chamber location). The simultaneous measurements in chamber location revealed that CH4 emission estimates by the SF6 technique were similar to those by the respiration chamber technique in the first 3 periods, although the SF6 estimates were significantly higher in period 4. The regression of all data from the 4 periods demonstrated a linear relationship between the SF6 and respiration chamber measurements for total CH4 emissions (g/d, R2 = 0.69) and for CH4 emissions per unit of milk yield (g/kg, R2 = 0.88), and a quadratic relationship for CH4 emissions per unit of dry matter intake (g/kg, R2 = 0.64). The CH4 emissions from the rectum were calculated as the difference between CH4 estimates from the SF6 technique when cows were housed in respiratory chambers and barn locations, which was 3% of the total CH4 emissions from the mouth, nostrils, and rectum. The SF6 estimates in the chamber location accounted for all sources of emissions, whereas those in the barn location, like that in grazing conditions, did not include CH4 emission from the rectum. Therefore, the SF6 measurements for grazing cattle should be adjusted for CH4 emissions from the rectum (3% of total). A complete microbial fuel cell (MFC) operating under continuous flow conditions and using Chlorella vulgaris at the cathode and Saccharomyces cerevisiae at the anode was investigated for the production of electricity. The MFC was loaded with different resistances to characterise its power capabilities and voltage dynamics. A cell recycle system was also introduced to the cathode to observe the effect of microalgae cell density on steady‐state power production and dynamic voltage profiles. At the maximum microalgae cell density of 2140 mg/L, a maximum power level of 0.6 mW/m2 of electrode surface area was achieved. The voltage difference between the cathode and anode decreased as the resistance decreased within the closed circuit, with a maximum open circuit voltage (infinite resistance) of 220 mV. The highest current flow of 1.0 mA/m2 of electrode surface area was achieved at an applied resistance of 250 Ω. 相似文献

The phase behaviors of crystalline solids embedded within nanoporous matrices have been studied for decades. Classic nucleation theory conjectures that phase stability is determined by the balance between an unfavorable surface free energy and a stabilizing volume free energy. The size constraint imposed by nanometer-scale pores during crystallization results in large ratios of surface area to volume, which are reflected in crystal properties. For example, melting points and enthalpies of fusion of nanoscale crystals can differ drastically from their bulk scale counterparts. Moreover, confinement within nanoscale pores can dramatically influence crystallization pathways and crystal polymorphism, particularly when the pore dimensions are comparable to the critical size of an emerging nucleus. At this tipping point, the surface and volume free energies are in delicate balance and polymorph stability rankings may differ from bulk. Recent investigations have demonstrated that confined crystallization can be used to screen for and control polymorphism. In the food, pharmaceutical, explosive, and dye technological sectors, this understanding and control over polymorphism is critical both for function and for regulatory compliance. This Account reviews recent studies of the polymorphic and thermotropic properties of crystalline materials embedded in the nanometer-scale pores of porous glass powders and porous block-polymer-derived plastic monoliths. The embedded nanocrystals exhibit an array of phase behaviors, including the selective formation of metastable amorphous and crystalline phases, thermodynamic stabilization of normally metastable phases, size-dependent polymorphism, formation of new polymorphs, and shifts of thermotropic relationships between polymorphs. Size confinement also permits the measurement of thermotropic properties that cannot be measured in bulk materials using conventional methods. Well-aligned cylindrical pores of the polymer monoliths also allow determination and manipulation of nanocrystal orientation. In these systems, the constraints imposed by the pore walls result in a competition between crystal nuclei that favors those with the fastest growth direction aligned with the pore axis. Collectively, the examples described in this Account provide substantial insight into crystallization at a size scale that is difficult to realize by other means. Moreover, the behaviors resulting from nanoscopic confinement are remarkably consistent for a wide range of compounds, suggesting a reliable approach to studying the phase behaviors of compounds at the nanoscale. Newly emerging classes of porous materials promise expanded explorations of crystal growth under confinement and new routes to controlling crystallization outcomes. 相似文献

Metamaterials or artificial negative index materials (NIMs) have generated great attention because of their unique electromagnetic properties. The main challenge in current NIM development is creating a homogenous NIM without the need of complex geometric architectures consisting of capacitors and inductors or aggregated fillers, but possessing a tunable plasma frequency. A natural material that can exhibit negative values for permittivity and permeability simultaneously has not been found, or discovered. If one can design a negative dielectric constant material with a tunable plasma frequency of interest, implementing negative permeability into the material or system would be much more readily achievable to create a metamaterial. In this regard, a novel negative dielectric constant material, which is an essential key to creating the NIMs, was developed by doping ions into a polymer, a protonated poly(benzimidazole) (PBI). The doped PBI showed a negative dielectric constant at frequencies of kHz to MHz because of its reduced plasma frequency and an induction effect. As temperature increased, the dielectric spectrum changed from a relaxation to a resonance behavior and exhibited a larger magnitude of negative dielectric constant at a lower frequency. The conductivity of the doped PBI measured as a function of both temperature and frequency followed the same trend as the dielectric constant. With respect to the dielectric constant and the conductivity data, it can be assumed that the origin of the negative dielectric constant is attributed to the resonance behavior of the highly mobile ions at elevated temperatures and high frequencies. © 2012 Wiley Periodicals, Inc. Estrogen receptor 1 and 2 (ESR1 and 2) mediate estrogen (E) action on gonadotrope function. While much is known about the effects of ESR1 on the gonadotrope, there is still some controversy regarding the effects of ESR2. To investigate the role of ESR2 in the gonadotrope, 45-day-old female mice of two different genotypes were used: wild type (WT) and pituitary (gonadotropes and thyrotropes)-specific Esr1 knockout (KO). All mice were ovariectomized (OVX) and 15 days later injected over 3 days with 2.5 μg 17β-estradiol (E(2)), 0.2 mg of the selective ESR1 or 2 agonists, propylpyrazole triol and diarylpropionitrile, respectively, or 0.1 ml oil. The day after treatment, anterior pituitary glands were dissected out for evaluation of gonadotrope ultrastructural morphology and pituitary immunohistochemical expression of progesterone receptor (Pgr (Pr)). Blood was collected and serum LH levels were assessed. Activation of ESR1 in WT mice resulted in the following: i) uterine ballooning and vaginal cornification, ii) negative feedback on LH secretion, iii) increased number of homogeneous (functional) gonadotropes, and iv) pituitary Pgr expression (35.9±2.0% of pituitary cells). Activation of ESR1 in KO mice induced normal uterine, vaginal, and LH secretion responses, but failed to increase the number of functional gonadotropes, and induced significantly lower Pgr expression (21.0±3.0% of pituitary cells) than in WT mice. Whilst activation of ESR2 had no significant effects in WT mice, it doubled the number of functional gonadotropes exhibited by KO mice injected with oil.
