136 1-yr-old infants were tested on tasks of visual–tactual cross-modal transfer and tactual intramodal processing. In Exp I, Ss successfully differentiated novel from familiar objects by tactual exploration after 60 sec of either visual familiarization (V–T) or 60 sec of tactual familiarization (T–T); all testing that involved tactual exploration was carried out in total darkness, using infrared videotaping. In both tasks, Ss spent significantly more time manipulating novel than familiar shapes. In the T–T task they also engaged in more manipulatory episodes and exhibited more hand-to-hand transfers with the novel shapes. Neither type of transfer was shown with shorter (30-sec) familiarization periods. The finding of successful T–T transfer with 60-sec familiarization was confirmed in Exp II using different stimuli and a modified testing procedure. Protein isolates prepared from yeasts by isoelectric precipitation and ultrafiltration, respectively, show differences in their solubility and gelchromatographic behaviour. The ultrafiltrated preparation represents a less aggregated protein. The SDS-discelectrophoresis patterns are identical, which is a proof for an identical protein polypeptide distribution in the isoelectric precipitated and ultrafiltrated samples Significant differences were found between ultrafiltrated proteins from Saccharomyces cerevisiae and Candida guilliermondi yeasts using SDS-disc-electrophoresis, gel chromatography, as well as determination of solubility profiles, amino acid composition, and thiol/disulphide content All the main fractions of the isolated protein posses a molecular weight distribution mainly in the range of 50000-70000. Investigated in 4 studies with 101 infants 25.5–32 wks of age the ability of Ss to transfer information about shape across modalities. Ss were familiarized either orally or tactually and then tested for visual recognition memory. In Exp I, Ss failed to show evidence of cross-modal transfer on any of the tasks (1 oral–visual, 2 tactual–visual). When familiarization times were increased from 30 to 60 sec in Exp II, Ss showed evidence of transfer on both tactual–visual tasks. Exp III eliminated the 5–7 sec delay that generally intervenes between the familiarization and test phase. Ss were permitted to retain the stimulus in their hand (or mouth) during the test phase while simultaneously viewing a novel stimulus and a duplicate of the familiar stimulus. This modification resulted in successful transfer on 1 of the 2 tactual–visual tasks. Ss did not show evidence of transfer on the oral–visual problem in any of these studies, despite evidence from Exp IV that they could visually discriminate the paired stimuli used in these tasks and that they showed recognition memory when familiarization and testing were both visual. Results suggest that, although cross-modal transfer of information about shape is present among 6-mo-olds, it is a less robust phenomenon than that seen in older infants. An improved procedure for the isolation and purification of the 11 S globulin from sunflower seeds (helianthinin) is described, including a combined purification by gel chromatography and ionexchange chromatography. The protein has a sedimentation constant of s20, w = 12.8 x 10?13 s, a Stokes radius of 57 Å and a diffusion constant of 3.76 x 10?7 cm2 s?1 (the last two derived from gel chromatographic analysis). Hence it follows a molecular weight of Ms, D = 305000. The isoelectric point determined by isoelectric focusing lies at pH 4.7. High contents of glutamic (26%) and aspartic (14%) acid and arginine (9.7%) as well as a low content of sulphur containing amino acids are characteristic for the amino acid composition. 59% of the acidic amino acids are present in an amidated form. Seven rapeseed varieties were characterized by investigation of the patterns of buffer soluble proteins using polyacrylamide gel electrophoresis and high-performance liquid chromatography (size-exclusion and cation-exchange chromatography). The 12 S globulin of rapeseed represents an oligomeric protein with a molecular weight of 300,000. It is composed of 6 subunits, which are arranged in a trigonal antiprism with the point group symmetry 32 (D3). Each subunit contains smaller units (polypeptide chains) with molecular weights in the range of 18,500 to 31,000. The protein contains the following 4 polypeptide chains differing by their molecular weights in the SDS-electrophoresis; 18,500 ± 800, 21,100 ± 500, 26,800 ± 900, 31,200 ± 1,600. According to its quaternary structure the protein dissociates under milieu conditions. The secondary structure of the protein is characterized by a low (11 %) content of α-helix and a relatively high (31%) content of β-conformation. Owing to its structure and physico-chemical properties the rapeseed protein is closely related to other 11/12 S globulins in the seeds of different plant species and botanical families. 相似文献

Information on chemical speciation is much needed in mechanistic and kinetic studies on catalyst formation processes in pharmaceutical research. Speciation analysis was applied to the identification and quantification of various rhodium species involved in a ligand exchange process leading to formation of catalyst dirhodium(II) tetrakis[methyl 2-oxopyrrolidin-5(S)-carboxylate]. Inductively coupled plasma mass spectrometry (ICPMS) was used as an element-specific detector following species separation by reversed-phase high-performance liquid chromatography (RP-HPLC), and electrospray ionization mass spectrometry (ESI-MS) was used for species identification and confirmation. A novel interface between the HPLC and ICPMS, which consisted of an eluent splitter, a desolvation unit, and the ICPMS built-in peristaltic pump, enabled the use of RP-HPLC with gradient elution and up to 100% organic components in the LC eluent without organic loading in the plasma. A variety of reaction intermediates were identified and quantified along the pathway to formation of the desired product, including isomeric di-, tri-, and tetrasubstituted species previously believed to be absent. This has provided new insights into the mechanism and kinetics of the reaction. The combination of HPLC-ICPMS and HPLC-ESI-MS has proven to be a valuable tool for the investigation of species evolution in catalyst formation process. 相似文献
