The effect of silica and polymer coated silica particles as reinforcing agents on the structural and mechanical properties of polystyrene-poly(ethylene/butylene)-polystyrene (PS-PEB-PS) triblock gel has been investigated. Different types of chemically modified silica have been compared in order to evaluate the influence of the compatibility between gel and filler.Time-resolved SANS and small-angle X-ray scattering (SAXS) shows that the presence of silica particles affects the ordering of the polystyrene domains during gelsetting. The scattering pattern of silica-reinforced gels reveals strong scattering at very low q, but no structure and formfactor information. However, on heating above the viscoelastic to plastic transition, the ‘typical’ scattering pattern of the copolymer gel builds-up. All reinforced gels are strengthened by the addition of the reinforcing agent. The transitions from a viscoelastic rubber to a plastic fluid and from a plastic fluid to a viscoelastic liquid are shifted to more elevated temperatures when silica is added to the triblock copolymer gel.  相似文献

The purpose of this study was to estimate the genetic variation and the heritability of the ability to establish an immune response by producing antibodies to Mycobacterium avium subsp. paratuberculosis. Antibody levels were determined using an ELISA and measuring optical density (OD) values from milk samples of 11,535 cows from 99 herds. The pedigree of the 11,535 cows and information about days in milk, parity, milk yield, and others were obtained from the Danish Cattle database. The statistical analyses were made using a bivariate mixed animal model. The bivariate model with daily milk yield and OD as dependent variables showed a significant heritability of the ability to produce Mycobacterium avium subsp. paratuberculosis antibodies of 0.102 (genetic variance=0.054) and a nonsignificant genetic correlation of -0.037 between daily milk yield and OD. When a sire model was used, the estimated heritability was 0.091. To evaluate whether intrauterine infection could be reflected in the OD, variation among 2715 dam-daughter or maternal sister groups was also estimated. Variation in this data was nonsignificant, possibly because only very few clinical cases or end-stage cows, i.e., heavy bacterial shedders may have been represented among the cows sampled. It does not appear that intrauterine transmission is of major importance in the transmission of paratuberculosis.  相似文献

The objective of this experiment was to determine whether uterine or ovarian vascular dynamics could be used to identify cows at risk for pregnancy loss. Our hypothesis was that cows that subsequently lose their pregnancy will have decreased corpus luteal (CL) perfusion, or an increased resistance index (RI; reduced blood flow), or both, at d 34 of pregnancy. Day 34 was chosen because it is a common time for dairy cattle to be checked for pregnancy. This experiment was performed in 2 replicates from November 2011 to April 2012 (n = 69) and from November 2012 to April 2013 (n = 53). Cows were bred via timed artificial insemination using Ovsynch-56 and checked for pregnancy on d 32 after artificial insemination. At d 34, cows confirmed pregnant were examined via transrectal Doppler ultrasonography. Blood samples collected via coccygeal vein were used to measure circulating plasma progesterone concentrations. Diameter of the corpus luteum and crown-rump length were measured. Color power Doppler ultrasonography was used to determine vascular perfusion to the CL, and RI was measured for the uterine arteries just after branching from the umbilical artery. Records were later examined to identify pregnancy status of cows after reconfirmation. Abortion rate did not differ between replicates (11.6% in replicate 1, 9.4% in replicate 2). Mean crown-rump length of embryos that were carried to term was greater on d 34 than that in cows that aborted (14.23 ± 0.27 vs. 13.21 ± 0.53 mm). Circulating progesterone concentration at d 34 was greater for cows that carried pregnancies to term than for those that aborted (9.1 ± 0.7 vs. 7.5 ± 1.0 ng/mL). The final logistic regression model consisted of crown-rump length, progesterone concentration, and RI of the uterine artery contralateral to pregnancy. Decreased crown-rump length and progesterone concentration tended to be associated with increased odds ratio for pregnancy loss, whereas CL perfusion and uterine blood flow were not associated with increased odds ratio of pregnancy loss. In conclusion, examining CL perfusion and RI of the uterine arteries on d 34 of pregnancy does not offer a method to identify lactating Dairy cattle at risk for pregnancy loss after d 34.  相似文献

Many medical and biotechnological applications rely on protein labeling, but a key challenge is the production of homogeneous and site‐specific conjugates. This can rarely be achieved by simple residue‐specific random labeling, but generally requires genetic engineering. Using site‐selective DNA‐templated reductive amination, we created DNA–protein conjugates with control over labeling stoichiometry and without genetic engineering. A guiding DNA strand with a metal‐binding functionality facilitates site‐selectivity by directing the coupling of a second reactive DNA strand in the vicinity of a protein metal‐binding site. We demonstrate DNA‐templated reductive amination for His6‐tagged proteins and metal‐binding proteins, including IgG1 antibodies. We also used a cleavable linker between the DNA and the protein to remove the DNA and introduce a single aldehyde on the protein. This functions as a handle for further modifications with desired labels. In addition to directing the aldehyde positioning, the DNA provides a straightforward route for purification between reaction steps.  相似文献

In the margarine and butter industry, sustainable and more efficient refrigerants, such as CO2, are introduced to industrial scraped surface heat exchangers, allowing an increased capacity compared to conventional use of NH3. The effect of such changes in capacity and a varied rotational speed was studied in relation to the structural behavior of puff pastry butter during 4 weeks of isothermal storage at 20 °C. The physical properties of the fat crystal network were studied in detail at several length scales by combining X-ray diffraction with differential scanning calorimetry, confocal laser scanning microscopy, LR-NMR and rheology. Our data shows that a high capacity combined with high rotational speed decreases the brittleness of puff pastry butter after 7 days of storage. This effect is, however, diminished after 28 days of storage. Likewise, changes in capacity and rotational speed are shown to induce no microstructural and polymorphic differences after 28 days of storage. However, the degree of work softening is related to the manufacturing conditions: a high capacity and a high rotational speed increase the ability of the puff pastry butter to resist structural breakdown during working. With this being the only observed difference; a wide operational window exists on an industrial level to produce puff pastry butter with similar structural behavior.  相似文献

The immature electrophysiology of human-induced pluripotent stem cell-derived cardiomyocytes (hiCMs) complicates their use for therapeutic and pharmacological purposes. An insufficient inward rectifying current (IK1) and the presence of a funny current (if) cause spontaneous electrical activity. This study tests the hypothesis that the co-culturing of hiCMs with a human embryonic kidney (HEK) cell-line expressing the Kir2.1 channel (HEK-IK1) can generate an electrical syncytium with an adult-like cardiac electrophysiology. The mechanical activity of co-cultures using different HEK-IK1:hiCM ratios was compared with co-cultures using wildtype (HEK–WT:hiCM) or hiCM alone on days 3–8 after plating. Only ratios of 1:3 and 1:1 showed a significant reduction in spontaneous rate at days 4 and 6, suggesting that IK1 was influencing the electrophysiology. Detailed analysis at day 4 revealed an increased incidence of quiescent wells or sub-areas. Electrical activity showed a decreased action potential duration (APD) at 20% and 50%, but not at 90%, alongside a reduced amplitude of the aggregate AP signal. A computational model of the 1:1 co-culture replicates the electrophysiological effects of HEK–WT. The addition of the IK1 conductance reduced the spontaneous rate and APD20, 50 and 90, and minor variation in the intercellular conductance caused quiescence. In conclusion, a 1:1 co-culture HEK-IK1:hiCM caused changes in electrophysiology and spontaneous activity consistent with the integration of IK1 into the electrical syncytium. However, the additional electrical effects of the HEK cell at 1:1 increased the possibility of electrical quiescence before sufficient IK1 was integrated into the syncytium.  相似文献

The capability to perform fast load changes has been an important issue in the power market, and will become increasingly more so due to the increasing commercialization of the European power market. An optimizing control system for improving the loadfollowing capability of power-plant units has therefore been developed. The system is implemented as a complement, producing control signals to be added to those of the existing boiler control system, a concept which has various practical advantages in terms of implementation and commissioning. The optimizing control system takes account of the multivariable and load-dependent nonlinear characteristics of the boiler process, as a scheduled LQG controller with feedforward action is utilized. The LQG controller improves the control of critical process variables, making it possible to increase the load-following capability of a specific plant. Field tests on a 265 MW coal-fired power-plant unit reveals that the maximum allowable load gradient that can be imposed on the plant, can be increased from 4 to 8 MW/min.  相似文献

The high-temperature wettability of alumina particulate preforms by copper is investigated by means of infiltration experiments conducted at 1473 K under low oxygen partial pressure. Wetting is quantified in terms of drainage curves, which plot the volume fraction of metal in the porous medium vs. the applied pressure. Mercury porosimetry is also used on similar preforms for comparison. The effect of volume fraction, particle geometry and capillary parameters on the drainage curve are studied and compared with the expression proposed by Brooks and Corey. The influence of the particle volume fraction and capillary parameters characterizing wetting in the two systems is discussed to derive an effective contact angle for wetting of alumina particles by molten copper.  相似文献

The objective of the study was to ascertain the state of the art with regard to the applicability of biobased packaging materials to foods and to identify potential food applications for biobased materials. The study revealed relatively few examples of biobased materials used as primary, secondary or tertiary packaging materials for foods. This is due to the fact that published investigations on the use of biobased materials are still scarce, and results obtained remain unpublished because of commercial pressures. The scientific literature contains numerous reports on applications of edible films and coatings to food but novel commercial applications of these are scarce. Based on information currently available on the properties of biobased packaging materials the study identified products in the fresh meat, dairy, ready meal, beverage, fruit and vegetable, snack, frozen food and dry food categories where conventional packaging may be replaced by biobased packaging.  相似文献

Wind power meteorology has evolved as an applied science firmly founded on boundary layer meteorology but with strong links to climatology and geography. It concerns itself with three main areas: siting of wind turbines, regional wind resource assessment and short-term prediction of the wind resource. The history, status and perspectives of wind power meteorology are presented, with emphasis on physical considerations and on its practical application. Following a global view of the wind resource, the elements of boundary layer meteorology which are most important for wind energy are reviewed: wind profiles and shear, turbulence and gust, and extreme winds.
