We have developed a rapid, multi-compound analytical method for measuring residues of the pesticides thiamethoxam and its metabolite, clothianidin; fipronil and its three metabolites, fipronil sulfone, fipronil sulfide, and fipronil desulfinyl; and pyraclostrobin in unprocessed foods (rice, corn, cucumbers, tomatoes, apples, and bananas) by ultra-performance liquid chromatography coupled to tandem mass spectrometry. Acetonitrile was used as the extraction solvent, and an octadecylsilane-dispersive SPE was used to clean up the analytes, which were then separated through a UPLC HSS T3 column connected to a tandem mass spectrometer via an electrospray ionisation source. The linearity of this method for the target analytes was excellent (R2 ≥0.990) in the concentration range of 5–1000 μg kg–1. The average recoveries of the seven compounds at concentrations of 10, 100, and 1000 μg kg–1 from six spiked matrix samples ranged from 73.6 to 110.6%, all with RSD values of ≤19.7%. The limit of quantification was 10 μg kg–1. The method validated the effectiveness of the method for routine monitoring the residue of these pesticides and their metabolites in foods. 相似文献

TRPV4 (transient receptor potential vanilloid 4), a calcium permeable TRP ion channel, is known to play a key role in endocytosis. However, whether it contributes to exocytosis remains unclear. Here, we report that activation of TRPV4 induced massive exocytosis in both melanoma A375 cell and heterologous expression systems. We show here that, upon application of TRPV4-specific agonists, prominent vesicle priming from endoplasmic reticulum (ER) was observed, followed by morphological changes of mitochondrial crista may lead to cell ferroptosis. We further identified interactions between TRPV4 and folding/vesicle trafficking proteins, which were triggered by calcium entry through activated TRPV4. This interplay, in turn, enhanced TRPV4-mediated activation of folding and vesicle trafficking proteins to promote exocytosis. Our study revealed a signaling mechanism underlying stimulus-triggered exocytosis in melanoma and highlighted the role of cellular sensor TRPV4 ion channel in mediating ferroptosis. 相似文献

Iron powders with two different particle size distributions were compacted by high velocity compaction. The influences of
particle size distribution and impact velocity on green properties, including green density, springback, tensile strength
and bending strength etc., were studied with scanning electron microscopy (SEM) and a computer controlled universal testing
machine. The results show that the particle size distribution and the impact velocity strongly affect its properties. Wider
size distribution results in green compact with higher density and better strength. Furthermore, springback of compacts is
lower produced by the powder with wider size distribution, especially for radial springback. As impact velocity increases,
its green density and green strength gradually increases, but the increasing rate of density decreases gradually. No special
relation is found between springback and impact velocity. In addition, the axial springback and the bending strength are higher
than the radial springback and the tensile strength, respectively. 相似文献
