Proteomics has been widely used to study muscle biology and meat quality traits from different species including beef. Beef proteomics studies allow a better understanding of the biological processes related to meat quality trait determination. This study aimed to decipher by means of two-dimensional electrophoresis (2D-PAGE), mass spectrometry and bioinformatics the changes in post-mortem muscle with a focus on proteins differentially expressed in the Longissimus thoracis (LT) muscle of immunocastrated young heifers and steers. Carcass traits, chemical composition, pH, instrumental color (L*, a*, b*), cooking loss and Warner-Bratzler shear force (WBSF) of meat from F1 Montana-Nellore cattle were also evaluated. Backfat thickness (BFT) and intramuscular fat content (IMF) were 46.8% and 63.6% higher in heifers (p < 0.05), respectively, while evaporation losses (EL) were 10.22% lower compared to steers. No differences (p > 0.05) were observed for tenderness evaluated by WBSF (3, 10, and 17 days post-mortem), pH, and color traits (L*, a* and b*) between the experimental groups. The study revealed several proteins to be differentially expressed proteins in heifers compared steers (p < 0.05). In heifers, proteins involved in nutrient transport (TF, ALB, and MB), energy metabolism (ALDOA, GAPDH, and PKM), and oxidative stress and response to stress (HSPA8 and CA3) were associated with a greater BFT and IMF deposition. The higher expression of these proteins indicated greater oxidative capacity and lower glycolytic activity in the LT muscle of heifers. In steers, there was greater abundance of protein expression related to muscle contraction and proteins of structure (ACTA1, TPM2 and TNNT3), energy metabolism (ENO1, ENO3, PYGM, PGM1 and TPI1) and ATP metabolism (ATP5F1B, PEBP1 and AK1), indicating greater glycogenolysis in LT muscle, suggesting a shift in the glycolytic/oxidative fibers of steers.  相似文献

The 8-aminoquinoline primaquine (PQ) is the only antimalarial drug used as tissue schizonticide and relapsing malaria. Antichagasic activity was also reported. Nevertheless, as it also shows serious side effects, prodrugs such as succinyl and maleyl derivatives have been proposed to decrease its toxicity. Although PQ mechanism of action has not been completely elucidated, the promotion of oxidative stress is an advanced hypothesis that could explain its activity in both plasmodia and trypanosome parasites. The oxidation of PQ and its prodrugs, maleylprimaquine (MPQ) and succinylprimaquine (SPQ), was studied by cyclic voltammetry using glassy carbon electrode. All compounds were oxidized in aqueous medium, with the charge transfer process being pH-dependent in acidic medium and pH-independent in a weak basic medium, being the neutral form more easily oxidized. This indicated that the protonation of the nitrogen atoms displays a determinant role in the voltammetric oxidation, being both prodrugs more easily oxidized than PQ protonated forms, in the order: SPQ < MPQ < PQ. For a better understanding of this behavior, a molecular modeling study was performed using the AM1 semi-empirical method from Spartan 04 for Linux (v.119, Wavefunction Inc.). The medium pH showed to be fundamental not only to the electronic density of the quinoline ring but also to the rearrangement of the nitrogen side chain. The electronic density of primaquine non-protonated quinoline ring is higher than that in its protonated and diprotonated species. Also, the use of prodrugs and the degree of saturation of the carriers (maleic or succinic acid) interfere with this feature. SPQ and MPQ have a slight increase in the quinoline electronic density in comparison to PQ. Nevertheless, the carrier in the side chain of SPQ is closer to the quinoline ring than it is in MPQ, which accounts for the higher electronic density in the former. The most significant effect occurs in the correspondent protonated forms of the nitrogen quinoline. The application of molecular modeling study associated to voltammetric techniques showed to be an important way to understand the redox mechanism of electroactive drugs. These results may be related to a biological activity and can be useful to future primaquine derivatives design.  相似文献

The quality inspection of products poses a challenge towards existing production metrology techniques in terms of accuracy and speed, especially when it comes to inline or in-process measurements. In this terms, optical metrology proves its capabilities, in particular for sensible and fragile work pieces. For the inspection inside small spaces and cavities or for complex geometries, many optical metrology system are to inflexible or cannot be miniaturized enough. Within this work a set-up for a fiber-optic low-coherence interferometer system is described. Due to the high degree of miniaturization of the sensing probe, the system can be used for demanding measuring tasks like roundness of small drilling holes or the run-out of shafts. Different evaluation measurement are presented, which underline the potential of the described system for highly accurate, non-contact inspection of rotational parts.  相似文献

Fructooligosaccharides have been isolated from roots and leaves of Stevia rebaudiana, by hot aqueous extraction, followed by precipitation with ethanol. Their structure has been determined using methylation and NMR analysis, MALDI-TOF, and ESI-MS. Fructooligosaccharides contained almost exclusively (2→1)-linked β-fructofuranosyl, with terminal α-glucopyranosyl and β-fructofuranosyl units. MALDI-TOF and ESI-MS analyses showed the wide range of degree of polymerisation (DP) present in various extracts. From roots and leaves, three different fractions gave profiles of homologous series, with DPs ranging up to 17 with MALDI-TOF and 19 using ESI-MS. These inulin-type fructooligosaccharides were the major component of extracts from S. rebaudiana roots and significant components from the leaves.  相似文献

This paper presents a strategy for load distribution between the generating units in hydro power plants. The objective is to reach the maximum energy conversion efficiency for a given dispatched power. The developed tool employs a heuristic-based combinatorial optimization technique in conjunction with a set of system variables measurement allowing real-time load sharing. The developed equipment is used to give online energy conversion efficiency from each unit of the power plant. No specific previous information about the efficiency of system components is required. Simulation results of the proposed optimization technique when applied to typical hydro power plant data are presented.  相似文献

In this work, the synthesis of high‐performance, metal ion‐imprinted, mesoporous carbon electrocatalysts for hydrazine oxidation reaction (HzOR) using casein or a family of phosphoproteins derived from cow's milk as a precursor is shown. The synthesis is made possible by mixing trace amounts of non‐noble metal ions (Fe3+ or Co2+) with casein and then producing different metal ions‐functionalized casein intermediates, which upon carbonization, followed by acid treatment, lead to metal ion‐imprinted catalytically active sites on the materials. The materials effectively electrocatalyze HzOR with low overpotentials at neutral pH and exhibit among the highest electrocatalytic performances ever reported for carbon catalysts. Their catalytic activities are also better than the corresponding control material, synthesized by carbonization of pure casein and other materials previously reported for HzOR. This work demonstrates a novel synthetic route that transforms an inexpensive protein to highly active carbon‐based electrocatalysts by modifying its surfaces with trace amounts of non‐noble metals. The types of metal ions employed in the synthesis are found to dictate the electrocatalytic activities of the materials. Notably, Fe3+ is found to be more effective than Co2+ in helping the conversion of casein into more electrocatalytically active carbon materials for HzOR.  相似文献

Sustainable and safe energy sources combined with cost effectiveness are major goals for society when considering the current scenario of mass production of portable and Internet of Things (IoT) devices along with the huge amount of inevitable e‐waste. The conceptual design of a self‐powered “eco‐energy” smart card based on paper promotes green and clean energy, which will bring the zero e‐waste challenge one step closer to fruition. A commercial raw filter paper is modified through a fast in situ functionalization method, resulting in a conductive cellulose fiber/polyaniline composite, which is then applied as an energy harvester based on a mechano‐responsive charge transfer mechanism through a metal/conducting polymer interface. Different electrodes are studied to optimize charge transfer based on contact energy level differences. The highest power density and current density obtained from such a paper‐based “eco‐energy” smart card device are 1.75 W m?2 and 33.5 mA m?2 respectively. This self‐powered smart energy card is also able to light up several commercial light‐emitting diodes, power on electronic devices, and charge capacitors.  相似文献

Sociotechnical (ST) theory emphasizes the notion that the design and performance of new systems can be improved, and can only work satisfactorily within an organization if the social and the technical aspects are brought together and treated as interdependent aspects of a work system. The implementation of lean manufacturing (LM) principles and practices has become popular, despite increasing demand on the current and emerging workforce to achieve higher levels of quality and flexibility with lower costs. Exploratory studies that investigate how LM practices influence the effect of ST practices on performance are still scarce, and a holistic view of such a relationship is needed. In this context, this study aims at verifying the moderating effect of just‐in‐time (JIT) practices on the relationship between ST practices and the performance of quality and workers' health. We carried out a survey with 144 different companies from Southern Brazil that are undergoing a lean implementation. Results indicate that ST practices have a positive significant relationship with such performance, and the concurrent adoption of JIT does not undermine quality and workers' health.  相似文献

Tension‐compression, tension‐tension, torsional, and 90° out‐of‐phase axial‐torsional fatigue tests were performed on a quenched and tempered U2 steel. All tests were conducted under force/torque control because macroscopic plastic strains were insignificant in the life range of interest (from 104 to 2 × 106 loading cycles). Stress‐based versions of the Fatemi‐Socie critical plane parameter and of the Smith‐Watson‐Topper parameter with a critical plane interpretation were evaluated using the experimental data. The Smith‐Watson‐Topper parameter was not able to correlate the test data. The Fatemi‐Socie method correlated most of the test data within factor‐of‐three boundaries. A modified Crossland invariant‐based parameter made of two interaction rules between the shear stress amplitude and the maximum hydrostatic stress, and of a definition of shear stress amplitude based on the maximum prismatic hull method, yielded fatigue life estimates in reasonable agreement with the experimental observations.  相似文献
