A laboratory microwave oven was used to dry high-moisture corn samples. The microwave oven was operated in both continuous and pulsed modes at 250 W of magnetron power setting. In the pulsed mode, two magnetron power-on times of 10 s and 15 s were used each with different power-off times in the range of 20 s to 75 s. Drying was more rapid in the continuous mode than in the pulsed mode. But, the continuous mode required much higher total magnetron power-on times. In the pulsed mode, longer power-on times generally resulted in slightly faster drying; and the power-off times did not strongly influence the drying rate. Longer power-on times should be followed by relatively longer power-off times. For a given on-time, increase in power-off time helps to decrease the total power-on time required for drying. When the microwave oven was operated at 10 s of power-on and 75 s of power-off pulsing, it resulted in the lowest total power-on time. 相似文献

The direct slicing of CAD models created in CADDS V to generate geometric data for rapid prototyping using fused feposition modeling technique (FDM) is presented in this paper. The report file from an explicit model is accessed for obtaining model data. Algorithms have been developed for determining the volumes of model material as well as support materials. New algorithms have been developed for filling the sheet solid. A simulation module has been developed to verify whether the filling is correctly done. The two most important beta-turn features in peptides and proteins are the type I and type II turns, which differ mainly in the orientation of the central peptide unit. Facile conformational interconversion is possible, in principle, by a flip of the central peptide unit. Homologous crystal structures afford an opportunity to structurally characterize both possible conformational states, thus allowing identification of sites that are potentially stereochemically mobile. A representative data set of 250 high-resolution (=2.0 A), non-homologous protein crystal structures and corresponding variant and homologous entries, obtained from the Brookhaven Protein Data Bank, was examined to identify turns that are assigned different conformational types (type I/type II) in related structures. A total of 55 examples of beta-turns were identified as possible candidates for a stereochemically mobile site. Of the 55 examples, 45 could be classified as a potential site for interconversion between type I and type II beta-turns, while ten correspond to flips from type I' to type II' structures. As a further check, the temperature factors of the central peptide unit carbonyl oxygen atom of the 55 examples were examined. The analysis reveals that the turn assignments are indeed reliable. Examination of the secondary structures at the flanking positions of the flippable beta-turns reveals that seven examples occur in the loop region of beta-hairpins, indicating that the formation of ordered secondary structures on either side of the beta-turn does not preclude local conformational variations. In these beta-turns, Pro (11 examples), Lys (nine examples) and Ser (seven examples) were most often found at the i+1 position. Glycine was found to occur overwhelmingly at position i+2 (28 examples), while Ser (seven examples) and Asn (six examples) were amongst the most frequent residues. Activation energy barriers for the interconversion between type I and type II beta-turns were computed using the peptide models Ac-Pro-Aib-NHMe and Ac-Pro-Gly-NHMe within the framework of the AM1 semi-empirical molecular orbital procedure. In order to have a uniform basis for comparison and to eliminate the distracting influence of the deviation of backbone dihedral angles from that expected for ideal beta-turns, the dihedral angles phii+1 and psii+2 were fixed at the ideal values (phii+1=-60 degrees and psii+2=0 degrees). The other two angles (psii+1 and phii+2) were varied systematically to go from type II to type I beta-turn structures. The computational results suggest that there exists one stereospecific, concerted flip of the central peptide unit involving correlated single bond rotation that can occur with an activation barrier of the order of 3 kcal/mol. The results presented here suggest that conformational variations in beta-turns are observed in protein crystal structures and such changes may be an important dynamic feature in solution. 相似文献

Anemic conditions in women and children across the world are a serious cause for concern. The standard method for measuring hemoglobin (Hb) in human blood is the well-recognized cyanmethemoglobin method as recommended by the World Health Organization (WHO). There are a number of methods available that give very approximate results as compared to this method. The color-measurement technique is one of them and it is recommended by the WHO for adaptation in low-resource settings. Since human interpretation errors are likely to creep in during the subjective processes involved with this method, an artificial neural network (ANN) approach for the estimation of Hb count in human blood has been evaluated. The ANN used color-coded values of the samples as input and the Hb value, as obtained with the cyanmethemoglobin method, as desired output using 2007 samples. A fuzzy logic controller has been developed for interfacing PV array with utility grid through a three-phase line-commutated inverter for the first time. The controller tracks and feeds maximum power to the utility grid. The linguistic variables have been selected appropriately to modulate the firing angle of the inverter for tracking the maximum power. The simulink model of the proposed scheme employing fuzzy logic controller has been built using MATLAB/PSB. A PIC microcontroller has been programmed for generation of firing pulses to the thyristors in the inverter. Experimental setup of the proposed scheme has been built and the results obtained on a PV array of 54 V, 12 A rating are presented. Confocal laser scanning microscopy (CLSM) is a noninvasive technique for evaluating the microstructure of foods and other materials. CLSM provides several sequential subsurface layers of two-dimensional (2-D) images. An image processing algorithm was developed to reconstruct these 2-D layers into a three-dimensional (3-D) network. Microstructure of fat globules in cheese was used as an example application. The validity of the image reconstruction algorithm was evaluated by processing several layered digital images of known shape and size.
