Reliable, rapid and inexpensive detection of cellulolytic enzymes that can be used for a wide variety of biological and environmental samples are currently in high demand. Here, a new cellulase detection protocol is described that circumvents problems observed with popular agar-based methods by exploiting the ability of carboxymethylcellulose (CMC) to form gel-like surfaces on its own. These pure CMC-layers are sensitive to cellulolytic degradation and stainable by Gram’s iodine without showing unwelcome reactions with other enzymes. The staining intensity negatively correlates with the enzyme activity and can be used for quantification. Cellulase activities are not obstructed by high sugar contents (e.g., in plant material) which limit the applicability of other quantification methods, making our new method particularly attractive for screening of plant extracts. A useful variant of this new method is its applicability to plant tissue prints for spatial mapping of the cellulolytic activity in a zymogram-like fashion.  相似文献

We present a study of the dissociative chemisorption of NO, O2, and N2 over close-packed, stepped, kinked, and open (fcc {111}, {211}, {311}, {532}, {100}, and {110}) transition metal facets using density functional theory (DFT). The offset of the Brønsted–Evans–Polanyi (BEP) relations suggest that the {111} surface is the least reactive, and that the {110} surface is the most reactive. This observation is verified by establishing volcano-relations based on mean-field microkinetic models for each facet, showing that the maximum rate over {110} is 4 orders of magnitude larger than the maximum {111} rate. The ordering of the maximum activity over the facets is: {110} > {100} ~ {532} > {311} ~ {211} > {111}, which is in general agreement with the offset in the BEP relations. We show that the top-point location and shape of the volcano relations are approximately independent of facet. This observation lends credibility to the approach of analyzing trends in catalytic reactivity over a single low-index facet, and assuming the experimentally observed activity trends are qualitatively well-described by such a single-facet analysis. Our study suggests that a key element for generally obtaining quantitative agreement between theory and experiments is for the simulations to address in detail the propensities of the various types of active sites. Finally, we show that the ordering of NO decomposition rates among metals and facets is essentially unaltered when using BEP- and scaling relations in the microkinetics instead of explicit DFT calculations for each elementary reaction step, and that using a “universal” BEP relation introduces no significant additional qualitative error in trend prediction.  相似文献

Conducted a mail survey of 141 human service workers (aged 25–65 yrs) to investigate the effects of coping on psychological strain and burnout produced by job stress. The survey assessed job stressors and coping strategies with open-ended questions and measured strain using closed-ended alienation, satisfaction, and symptom scales. Because previous research suggested that individual coping responses do not alleviate strain produced by job stress, the survey elicited information on group coping (social support) and on coping strategies initiated by agencies. Job stress was associated with high levels of strain, and group coping with low levels, but individual responses had little effect. Although Ss identified many strategies that agencies could use to reduce stress and strain, actual use of such strategies was slight. Because men and women worked in the same jobs, no sex differences in individual coping were predicted and none were found; women, however, reported more social support than men. There was no evidence for moderating (interaction) effects of stress and coping on strain. Results suggest that social service agencies should take actions to reduce stress among employees. This study was designed to evaluate the suitability of a novel bioabsorbable material in treating bone defects. A poly(desaminotyrosyl-tyrosine-ethyl ester carbonate) (PDTE carbonate) membrane (thickness 0.2-0.3 mm) was implanted into the mandibular angle of 20 New Zealand White rabbits to cover a through-and-through defect (12 x 6 mm). In group 1, the defects were left unfilled but covered with membrane and in group 2 the defects were filled with bioactive glass mesh and covered with membrane, too. Controls were left uncovered and unfilled. The animals were followed for 6, 12, 24 and 52 weeks, respectively. The material was evaluated by qualitative analysis of histological reactions and newly formed bone.We found that PDTE carbonate elicited a modest foreign body reaction in the tissues, which was uniform throughout the study. New bone formation was seen in all samples after six weeks. Group 1 had more new bone formation until 24 weeks and after this the difference settled. Based on findings of this study it was concluded that PDTE carbonate membranes have good biocompatibility and are sufficient to enhance bone growth without additional supportive matrix.  相似文献

Discrimination and recognition are often poorer for other-race than own-race faces. These other-race effects (OREs) have traditionally been attributed to reduced perceptual expertise, resulting from more limited experience, with other-race faces. However, recent findings suggest that sociocognitive factors, such as reduced motivation to individuate other-race faces, may also contribute. If the sociocognitive hypothesis is correct, then it should be possible to alter discrimination and memory performance for identical faces by altering their perceived race. We made identical ambiguous-race morphed faces look either Asian or Caucasian by presenting them in Caucasian or Asian face contexts, respectively. However, this perceived-race manipulation had no effect on either discrimination (Experiment 1) or memory (Experiment 2) for the ambiguous-race faces, despite the presence of the usual OREs in discrimination and recognition of unambiguous Asian and Caucasian faces in our participant population. Systems Biology aims for a holistic understanding of biological processes. In order to make this understanding operational and testable it can be recorded into formal process calculus models. This is a difficult task, however, because such formal models and their, often infinitely many, consequences are hard to enumerate and understand. In this paper we define a pathway analysis, based on static analysis techniques from programming languages, and show how it can be used to establish useful, finite, approximations to the set of causal consequences of models. The Pathway Analysis can be used to great advantage in all phases of the modelling approach – serving as the basis of debugging during model development, postdiction during model validation, and, finally, prediction during model guided drug design.  相似文献

This study for the first time reports the combined use of low-field nuclear magnetic resonance (LF-NMR) and confocal laser scanning microscopy (CLSM) in the study of the effects of salting on low and high dry matter (DM) potato tissue. The simultaneous use of CLSM and NMR resulted in important information in relation to the interpretation of the origin of the NMR water populations. Salting caused the raw potato cells to loose weight, which in the microscopic images was observed as loss of turgor pressure still further away from the edges of the samples with increased salting time. The total water loss after salting was lowest for high DM potatoes. The LF-NMR analyses revealed that this could be ascribed to a faster T2 relaxation time of the cytoplasmatic and extracellular water and thus to water being more restricted in high DM potatoes. In addition, a tendency to a faster T2 relaxation time was observed for both low and high DM potatoes with increasing salting time, which reveals a salt-induced restriction of the cytoplasmatic and extracellular water population. The paper illustrates the aptitude of NMR and CLSM to determine and elucidate structural changes and associated changes in water mobility in potato tissue.  相似文献

The formation of NOx from combustion of fossil and renewable fuels continues to be a dominant environmental issue. We take one step towards rationalizing trends in catalytic activity of transition metal catalysts for NO decomposition by combining microkinetic modelling with density functional theory calculations. We show specifically why the key problem in using transition metal surfaces to catalyze direct NO decomposition is their significant relative overbinding of atomic oxygen compared to atomic nitrogen.  相似文献
