Differential influence of a selective melanocortin MC4 receptor antagonist (HS014) on melanocortin-induced behavioral effects in rats 总被引:1,自引:0,他引:1

We injected i.c.v. the natural agonist alpha-MSH (melanocyte-stimulating hormone) and the first selective melanocortin MC4 receptor antagonist HS014 (cyclic [AcCys11, D-Nal14, Cys18, Asp-NH(2)22]-beta-MSH(11-22) in rats and scored a number of behavioral effects which have been related to the melanocortic peptides. The results showed that HS014 (5 microg/rat) completely blocked alpha-MSH (3 and 5 microg/rat)-induced grooming, yawning and stretching. Penile erections induced by alpha-MSH were, however, only partially blocked by HS014. Injections of alpha-MSH decreased food intake in food-deprived rats, whereas HS014 increased food intake. When the peptides were given together, the food intake was similar to that of saline treated controls. Locomotion/exploration and resting were not influenced by either peptide. Our data show that exogenous beta-MSH decreases food intake, and that an endogenous central melanocortinergic inhibitory tone on feeding prevails which can be blocked with HS014, leading to an increase in food intake. Our data also provide evidence that grooming, stretching and yawning in rats may be mediated by the melanocortin MC4 receptor, whereas penile erections might perhaps be mediated by some other melanocortin receptor. 相似文献

The French have one of the lowest incidences of coronary heart disease in the Western world despite a diet with a relatively high fat content. This phenomenon that has puzzled researchers worldwide for more than a decade is known as the 'French paradox' and has been linked to the high consumption of red wine in France. Red wine is rich in the complex polyphenols, the proanthocyanidins, and these compounds have recently attracted attention as potential cardiac-protective compounds. The present review summarizes the literature on proanthocyanidins with focus on their chemical structure, the occurrence, the daily intake from foods, the bioavailability and metabolism, and the evidence for a protective effect against cardiovascular diseases. 相似文献

The conditions affording biphasic competition curves in radioligand binding for ligands subjected to metabolic transformation was analyzed theoretically. It was shown that when a competing ligand was subjected to transformation to a ligand that showed higher affinity than the parent compound, biphasic competition curves, which might wrongly be interpreted as indicating the presence of two receptor sites, could be observed in binding assays containing a homogenous receptor population. Biphasic competition curves were seen if the conversion of the competitor occurred according to zero and second order kinetics, as well as by enzymatic catalytic processes. However, when the conversion occurred according to a first order kinetics, the competition curves were uniphasic and resolved only into one-site fits, with the apparent affinity of the competitor reflecting the degree of conversion of the competitor to its metabolite. When the metabolic conversion resulted in a metabolite that showed lower affinity for the receptor than the parent compound, the competition curves became supersteep for conversions according to zero and second order kinetics, as well as for conversion by enzymatic catalytic processes. 相似文献

In order to investigate a possible relationship between integrin degradation and water-holding capacity (WHC), integrin was quantified using western blot, and water mobility and distribution was measured by proton NMR T(2) relaxometry at 24h postmortem in pork (n=30) with a large variation in WHC (drip loss varying from 2.8% to 11.3%). Regression analyses revealed correlation coefficients of r=-0.32 (P=0.08) and r=0.40 (P=0.03) for the correlations between the content of integrin determined by western blot analysis and WHC determined as either drip loss or by NMR, respectively. Water mobility and distribution was also measured in 18 meat samples upon 7 days of aging, which revealed a correlation (r=0.54) between integrin content determined by western blot analysis 24h postmortem and the mobility of the myofibrillar water (T(21) relaxation time) at day 7. In contrast, no correlation could be established between integrin content 24h postmortem and WHC at day 7 determined by NMR as the amount of extramyofibrillar water (T(22) population) (r=-0.01). In conclusion, both visualisation by CLSM and quantification of integrin by western blot analyses of suggested that a strong link between integrin degradation and WHC in pork is questionable, whereas integrin degradation seems to have impact on the succeeding development in the mobility of the myofibrillar water. 相似文献

The effect of water on the activity and selectivity for a series of γ-Al2O3 supported cobalt Fischer–Tropsch catalysts has been studied in an isothermal fixed-bed reactor at T = 483 K, P = 20 bar, and H2/CO = 2.1. The catalysts were produced applying incipient wetness impregnation and consisted of 20 wt.% cobalt and 0.5 wt.%
rhenium deposited on γ-Al2O3 supports with different pore characteristics. For the narrow-pore catalysts, addition of water corresponding to an inlet
partial pressure ratio of PH2O/PH2 = 0.4 reduced the reaction rates. In contrast, for a catalyst with larger pores the same water pressure increased the reaction
rates. For all catalysts, water amounts equal to PH2O/PH2 = 0.7 at the reactor inlet suppressed the reaction rates and led to permanent deactivation. The addition of water increased

Conflicts over conservation are often regarded as dichotomies of diametrical opposites. When national parks are established in Norway, two stereotypes can be sketched: (1) the ecocentric position valuing 'pure, authentic nature', relying on the national government as the only acceptable decision and management level; and (2) a more anthropocentric position valuing cultural landscape elements and certain traditional and future uses of the area, favouring local management. A conflict over restoring a firing range to a state of 'authentic nature' for future designation as a national park is examined. The case has raised comprehensive local debate. This study presents the first application of a high-resolution magic angle spinning 1H NMR approach to elucidate the metabolic effects of a hypercholesterolemic condition and two high-fiber diets based on rye and wheat bread, respectively, in intact pig liver biopsy samples. Standard 1D and spin-echo 1H spectra were acquired on a total of 20 biopsy samples, and 2D total correlation spectroscopy experiments were carried out on selected samples for assignment of the observed resonances. Principal component analyses and partial least-squares regression discriminant analysis revealed differences in the hepatic lipid content and choline-containing compounds between normal and hypercholesterolemic pigs. In addition, the results demonstrated that the liver metabolite profile of hypercholesterolemic pigs fed a high-fiber rye bread differed from that of pigs fed high-fiber wheat bread with respect to both the lipoprotein fractions and the choline-containing compounds. These findings suggest that earlier reports on high-fiber rye diet-induced effects on plasma HDL/LDL content partially can be ascribed to effects on liver cholesterol metabolism and that the hepatic phospholipase pathways of phosphatidylcholine breakdown are affected by the high-fiber rye diet. 相似文献
