Histogenetically, the thymus is the primary lymphopoietic organ and provides an optimal microenvironment for the differentiation of T lymphocytes, independently of the influence of foreign antigens. Lymphocytes with diverse potential are produced in a protective microenvironment optimal for their maturation, whose dual cellular network is provided by endodermally derived RE cells and numerous ectomesenchymal cells derived from the neural crest. The full development of intrathymic hematopoiesis depends upon the successful completion of a series of well coordinated cellular interactions between widely divergent (in terms of origin) cells [epithelium (primitive pharynx); ectomesenchyrne (neural crest); and PHSCs (yolk sac, fetal liver)]. The cells of the thymic epithelial primordium do not proliferate in the absence of "inductive" interactions with the ectomesenchyme. Moreover, the nature of the mesenchyme determines the behavior of the thymic epithelial anlagen. The ectomesenchymal origin of chemotactic stem cell factor secretion, responsible for hemopoietic stem cell immigration, is a distinct possibility. The human thymus is a generalized hematopoietic tissue with between 7 to 9 weeks of ontogenesis. In human and dog fetuses various elements of mammalian hematopoiesis were identified intrathymically: B lymphocytes, plasma cells, erythropoietic and granulocytopoietic (neutrophils and eosinophils) cells, antigen presenting dendritic cells, and mast cells. Our light and ultrastructural (transmission and scanning), as well as immunocytochemical observations have established that during the embryonal and fetal period, the thymus is seeded by pluripotent, yolk sac derived PHSCs characterized by the following immunophenotype CD34+CD43+CD38-Lin-HLA-DR+CD69+. Stem cell c-kit tyrosine kinase (also referred to as mast cell growth factor, stem cell factor, or steel factor) in combination with autocrine and paracrine growth factors and cytokines (IL-3, IL-4, IL-5, IL-6, IL-7, G-CSF, etc.) stimulates myelopoiesis, including erythropoiesis, as well as lymphopoiesis. These hematopoietic growth factors are produced by activated lymphoblastic cells and stromal RE cells under the influence of immunoneuroendocrine regulation, supported by the finding that experimental or spontaneous, in vivo neural crest ablation during early mammalian ontogenesis always results in an abnormal development of the thymus, as well as the heart and great vessels, thyroid, and parathyroid glands.  相似文献

The purpose of this study was to evaluate a non-flow related magnetic resonance imaging method to visualize small veins independent of arteries in the human brain. A long TE, high-resolution 3D gradient echo MR acquisition was used to highlight venous information. The method is based on the paramagnetic property of deoxyhemoglobin and the resulting phase difference between veins and brain parenchyma at long echo times. The MR magnitude images were masked with a phase mask filter to enhance small structure visibility.. Venous information down to sub-pixel vessel diameters of several hundred microns is visible. Venous data are displayed in an angiographic manner using a minimum intensity projection algorithm. Both superficial veins and deep white matter veins are visible. The method has been successfully applied in volunteers. Preliminary results in patients with cerebral arteriovenous malformations indicate its potential in clinical applications. The proposed method is easy to implement and does not require administration of a contrast agent or application of specially designed rf pulses to highlight the veins. Rather it exploits the intrinsic magnetic properties (BOLD-effect) and the prolonged T 2* of venous blood. The method may be of diagnostic potential in the assessment of arteriovenous malformations or other vascular venous lesions. © 1998 Elsevier Science B.V. All rights reserved.  相似文献

Single optically levitated microparticles were investigated by Raman spectroscopy. The particles were composed of di-octyl-phthalate (DOP) and glycerol; these substances are not mixable and form a two-phase droplet. Measurements of the Raman spectrum confirm the formation of droplets containing both chemical species. The spectra show strong input and output structural resonances as expected. If the particle is in resonance, the field inside the particle is enhanced, and most of the inelastically scattered light is emitted from molecules close to the droplet rim. If the particle does not fulfill the resonance condition, the contribution of an individual molecule to the Raman scattering does not depend strongly on the radial position of this molecule. On this basis, the radial distribution of the two components inside the evaporating droplet was determined by time-dependent measurements of the Raman spectrum. Furthermore, elastic-light scattering and the evaporation characteristics of the particles were investigated.  相似文献

A new measurement technique for the characterization of uniaxial as well as biaxial anisotropic surfaces and thin films is introduced. This technique is based on perpendicular-incidence photometric ellipsometry, in which a spectral-photometric dynamic ellipsometer with a rotating polarizer is used. This method is sensitive, contactless, nondestructive, and efficient for the estimation of anisotropic behavior. Furthermore, the spectroscopic measurement directly provides the anisotropy dispersion down to the UV wavelength range. Results on structurally anisotropic HfO(2) coatings are presented.  相似文献

We have developed fluoride antireflection (AR) coatings on MgF(2) substrates for a wavelength of 248 nm by molecular-beam deposition. Transmission and laser-induced damage threshold of the samples were measured and atomic force microscope (AFM) investigations were carried out. We compare a 14-layer design for AR coatings with sublayer thicknesses of 12 nm with a conventional two-layer design with quarter-wavelength thicknesses. The laser-induced damage threshold of the 14-layer coating is slightly higher than that of the two-layer coating. The AFM surface images show that the 14-layer coating has a smoother surface than the two-layer coating.  相似文献

The numerical solution of fluid–structure-interaction problems poses a paradox in that most of the computational resources are consumed by the subsystem of least practical interest, viz., the fluid. Goal-oriented adaptive discretization methods provide a paradigm to bypass this paradox. Based on the solution of a dual problem, the contribution of local residuals to the error in a specific goal functional is estimated, and only the regions that yield a dominant contribution are refined. In the present work, we address a fundamental complication in the application of goal-oriented adaptivity to fluid–structure-interaction problems, namely, that the treatment of the interface conditions has nontrivial consequences for the properties of the dual problem. In the context of a linearized model problem, we consider two equivalent discretizations differing only on the formulation of the interface coupling terms. By means of an adjoint consistency analysis, we show that only one of these discretizations is adjoint consistent. Numerical experiments convey that the two discretizations behave very differently for the dual problem, and that the adjoint-consistent discretization yields more reliable error estimates. Based on the adjoint-consistent discretization, we finally present some h- and hp-adaptive results, confirming that tremendous savings in computational cost can be realized through the use of goal-oriented refinement strategies. The numerical experiments illustrate that the goal-oriented approach effectively equilibrates the error contributions of the fluid and structure subsystems, which is imperative for efficiently resolving the coupled fluid–structure-interaction problem, and which cannot be accomplished by uniform or residual-based refinement strategies.  相似文献

Epoxy resin based negative photo resists are showing interesting properties which are useful for a series of applications in electronic industries (Mark et al. in Encyclopedia of polymer science and engineering, Wiley, New York, 1986; Potter in Epoxide resins, Springer, New York, 1970; May and Tanka in Epoxy resin chemistry and technology, Marcel Dekker, New York, 1973; Bauer in Epoxy resin chemistry, vol 114, American Chemical Society, Washington, 1979; Hood in RAPRA Rev Rep 38:4, 1990). Especially in micro system technologies they promise a high potential. To adjust the properties for the particular application modification of the chemical composition and crosslinker system must be performed. An actual problem is the constancy in behaviour during the LIGA-process of the resist mixture. In this work analytical techniques are used to get a detailed insight into the chemical composition of a commercial available epoxy novolac resin. Methods like size exclusion chromatography (SEC), matrix assisted laser desorption-time of flight (MALDI-ToF) and fourier transform infrared spectroscopy (FTIR) offer information about the molecular weight and functionality. The results reveal a distribution in molecular weight and functionality. Due this the resin was chemically modified. After processing a dependency of molecular weight and therefore the amount of epoxy groups existent was obtained.  相似文献

In immersive virtual environments (IVEs), users can control their virtual viewpoint by moving their tracked head and walking through the real world. Usually, movements in the real world are mapped one-to-one to virtual camera motions. With redirection techniques, the virtual camera is manipulated by applying gains to user motion so that the virtual world moves differently than the real world. Thus, users can walk through large-scale IVEs while physically remaining in a reasonably small workspace. In psychophysical experiments with a two-alternative forced-choice task, we have quantified how much humans can unknowingly be redirected on physical paths that are different from the visually perceived paths. We tested 12 subjects in three different experiments: (E1) discrimination between virtual and physical rotations, (E2) discrimination between virtual and physical straightforward movements, and (E3) discrimination of path curvature. In experiment E1, subjects performed rotations with different gains, and then had to choose whether the visually perceived rotation was smaller or greater than the physical rotation. In experiment E2, subjects chose whether the physical walk was shorter or longer than the visually perceived scaled travel distance. In experiment E3, subjects estimate the path curvature when walking a curved path in the real world while the visual display shows a straight path in the virtual world. Our results show that users can be turned physically about 49 percent more or 20 percent less than the perceived virtual rotation, distances can be downscaled by 14 percent and upscaled by 26 percent, and users can be redirected on a circular arc with a radius greater than 22 m while they believe that they are walking straight.  相似文献
