A relatively thermostable 22-kDa endoribonuclease (MAR1) was purified more than 10,000-fold from a mitochondrial extract of Leishmania tarentolae and the gene cloned. The purified nuclease has a Km of 100-145 +/- 33 nM and a Vmax of 1.8-2.9 +/- 2 nmol/min, depending on the RNA substrate, and yields a 3'-OH and a 5'-phosphate. Cleavage was limited to several specific sites in the substrate RNAs tested, but cleavage of pre-edited RNAs was generally independent of the addition of cognate guide RNA. The MAR1 gene was expressed in Escherichia coli or in L. tarentolae cells, and the recombinant protein was affinity-purified. The cleavage specificity of the recombinant enzyme from L. tarentolae was identical to that of the native enzyme. The single copy MAR1 gene maps to an 820-kilobase pair chromosome and contains an open reading frame of 579 nucleotides. The 18-amino acid N-terminal sequence shows characteristics of an uncleaved mitochondrial targeting sequence. Data base searching revealed two homologues of MAR1 corresponding to unidentified open reading frames in Caenorhabditis elegans (GenBankTM accession number Z69637) and Archaeoglobus fulgidus (GenBankTM accession number AE000943). The function of MAR1 in mitochondrial RNA metabolism in L. tarentolae remains to be determined.  相似文献

The pathogenic significance of cell adhesion molecules (CAMs) in ulcerative colitis (UC) is largely unknown. Colonic expression of E-selectin, sialyl Lewis X (sLe(x)), and macrophage inflammatory protein-1x (MIP-1alpha) as well as serum concentrations of E-selectin and MIP-1alpha in UC were studied. Thirty patients with UC, 10 patients with irritable bowel syndrome, and 10 healthy subjects were included. Colonic biopsies were stained immunohistochemically, and blood concentrations were measured with an ELISA technique. Soluble E-selectin did not correlate with diagnosis or disease activity. MIP-1alpha was below the detection limit. Epithelial cells expressed all three molecules, both on surface membranes and intracellularly. sLe(x) staining was weaker (P = 0.0002) and MIP-1alpha staining stronger (P = 0.014) in UC patients than in controls. Leukocyte MIP-alpha staining correlated with diagnosis (P = 0.021), sLe(x) staining (P = 0.023), and colonoscopy (P = 0.018). It is shown that E-selectin, sLe(x), and MIP-1alpha are synthesized and expressed by epithelial cells, indicating that CAMs are not only involved in leukocyte extravasation and migration, but also in the interaction between leukocytes and colonic epithelium. This knowledge might contribute to the development of improved treatments in UC.  相似文献

A procedure for subspace stacking is proposed, and it is proved that the technique is equally successful for both linear codes and anticodes. It is demonstrated that families of errorcorrecting codes may be constructed using the proposed technique for stacking linear codes. In December 1996, TIME magazine published an article titled "Finding God on the Web," which projected how the Internet would shape information seeking on faith and religion. A decade later, the Web has become a popular tool for gathering religious information as well as information and imagery related to religious travel. Yet there is little research on religious destination imagery online. Imagery—positive or negative, pictographic or narrative—influences the selection of tourism destinations. This study explored online Muslim images in Malaysia via interviews and content analyses of pictures and text on tourism destination websites' homepages. The results reveal minimal portrayal of Muslim images by Malaysian tourism destination organizations. This study adds to the small body of research, especially in Muslim countries, on online religious imagery and suggests avenues for tourism operators in Malaysia and elsewhere to improve their online image with both Muslim and non-Muslim travelers.  相似文献

Theoretical analysis of potential distribution in the interdigital-gated high electron mobility transistor (HEMT) plasma wave device was carried out. The dc IV characteristics of capacitively coupled interdigital structure showed that uniformity of electric field under the interdigital gates was improved compared to the dc-connected interdigital gate structure. Admittance measurements of capacitively coupled interdigital gate structure in the microwave region of 10–40 GHz showed the conductance modulation by drain–source voltage. These results indicate the existence of plasma wave interactions.  相似文献

ABSTRACT

Magnesium layered hydroxide (MLH) intercalated with anionic 3-(4-methoxyphenyl)propionic acid (MPP) was synthesised by a direct reaction method using magnesium oxide and MPP as precursors. A further coating of chitosan was applied on the external surface of MLH–MPP nanocomposite to form a new material, named MLH–MPP/chitosan nanocomposite. The XRD pattern showed an intense and sharp peak at basal spacing 18.9 Å, proving that MPP anions were successfully intercalated into the interlayer gallery of MLH in a monolayer arrangement. The XRD pattern of the MLH–MPP/chitosan nanocomposite shows similar peaks with the MLH–MPP nanocomposite. The result was also supported by FTIR spectra and elemental analysis. TGA/DTG spectra showed that the thermal stabilities of the guest anion in the both nanocomposites were markedly enhanced. A controlled-release study of the MPP ion from the MLH–MPP/chitosan nanocomposite showed a slower release compared to MLH–MPP nanocomposite with an initial rapid release and slow release thereafter. Meanwhile, the release behaviours of MPP ions from both nanocomposites were governed by pseudo-second order kinetics. This result highlights the potential of the nanocomposite as an encapsulated material for the controlled-release formulation of MPP anions.  相似文献

This paper presents the current situation and projected planning of the electricity generation sector for Iskandar Malaysia by implementing a model to optimise the cost, utilise the usage of available renewable energy sources, and achieve carbon dioxide reduction targets. This Mixed Integer Linear Programming model was developed with the main objective of minimising the total cost of electricity generation, taking into consideration energy demand, reserve margin, electricity generation, peak and base generation, resource availability, and CO2 emission. Data for the year 2013 were forecasted until 2025 to illustrate the analysis for this study, and are represented via four scenarios. This optimal model is capable of balancing types of fuel and switching coal plants to natural gas power plants. It also enhances the use of renewable energy (RE) to meet CO2 emission targets. The model is further integrated with several other considerations related to energy systems, such as suitability of power plants as peak or base plants, RE resource availability, intermittency of solar power, losses during transmission, fuel selection for biomass, decision to retrofit existing coal power plant to NG power plant, and construction lead time of power plants. The results for this study determined that the optimal scenario is Scenario 3 (CS3). This research proves that Iskandar Malaysia can reduce CO2 emission by 2025 via utilisation of RE. This model is generic and can be applied to any case study, which would be useful for assisting government policy-making.  相似文献

In-stent restenosis is a common complication after stent surgery which leads to a dangerous wall narrowing of a blood vessel. Laser assisted patterning is one of the effective methods to modify the stent surface to control cell–surface interactions which play a major role in the restenosis. In this current study, 316LS stainless steel substrates are structured by focusing a femtosecond laser beam down to a spot size of 50 μm. By altering the laser induced spot density three distinct surfaces (low density (LD), medium density (MD) and high density (HD)) were prepared. While such surfaces are composed of primary microstructures, due to fast melting and re-solidification by ultra-short laser pulses, nanofeatures are also observed as secondary structures. Following a detailed surface characterization (chemical and physical properties of the surface), we used a well-established co-culture assay of human microvascular endothelial cells and human fibroblasts to check the cell compatibility of the prepared surfaces. The surfaces were analyzed in terms of cell adherence, proliferation, cell morphology and the differentiation of the fibroblast into the myofibroblast, which is a process indicating a general fibrotic shift within a certain tissue. It is observed that myofibroblast proliferation decreases significantly on laser treated samples in comparison to non-treated ones. On the other hand endothelial cell proliferation is not affected by the surface topography which is composed of micro- and nanostructures. Such surfaces may be used to modify stent surfaces for prevention or at least reduction of restenosis.  相似文献
