Total Synthesis and Structural Elucidation of Two Unusual Non-Methylene-Interrupted Fatty Acids in Ovaries of the Limpet <Emphasis Type="Italic">Cellana toreuma</Emphasis> 下载免费PDF全文
In our previous study, unusual odd‐numbered dienoic acids with a terminal olefin were found as minor components in ovaries of the Japanese limpet Cellana toreuma, and the synthetic interests have been focused onto their structural confirmation and the inspection into their potential biological activity. Here, we describe an efficient and stereoselective total synthesis of two new unusual dienoic acids, 19:2?7,18 and 21:2?7,20, through a common pathway involving the strategic combination of alkyne‐zipper reaction and Lindlar hydrogenation for the construction of their unique carbon chains. In our synthetic study, 2‐propyn‐1‐ol was at first subjected to alkylation and alkyne‐zipper reaction to form the two fragments, and the subsequent carbon chain elongation was achieved by the usual coupling reaction to obtain the C‐19 and C‐21 products bearing an internal acetylenic group. Then, the internal acetylenic group of these products was subjected to Lindlar hydrogenation to form a Z‐alkenyl moiety, and the subsequent deprotection of the products was carried out under an acidic condition without isomerization of the internal Z‐alkenyl group. Total synthesis of target fatty acids, 19:2?7,18 and 21:2?7,20, was finally accomplished by two‐step oxidation of the resulting alcohols into carboxylic acids in a highly chemoselective manner, and the structures of these unusual natural fatty acids were finally elucidated by identifying the GC–MS spectra of the methyl esters of authentic and synthetic fatty acids. 相似文献

This article describes the dewatering of sewage sludge using liquefied dimethyl ether (DME), which clarified the effect of sludge type and its particle size. Changes in the heating value of the dewatered sludge were also examined. In addition, the change in dewaterability upon reuse of the liquefied DME was investigated experimentally. Regardless of the nature of the sludge, its dewatering behavior was similar. The amount of dewatered water could be increased by decreasing the diameter of the sludge cake. Complete drying of the sludge cake via DME dewatering resulted in retention of approximately 96% of the heating value (17.7 MJ/kg-sludge) of the original sludge cake dry base (18.4 MJ/kg db). The DME could be recovered after extraction, and reuse experiments showed that liquefied DME dewatering performance was unchanged after five reuses. 相似文献

A method of reducing surface recombination velocities (SRVs) of crystalline silicon (c-Si) wafers is presented for the case when c-Si surfaces are passivated by amorphous Si (a-Si) thin films. It is demonstrated that the surface treatment of c-Si wafers prior to a-Si deposition, using hydrogen atoms generated by catalytic reaction of hydrogen gas with heated catalyzer, lowers SRV effectively, and that SRV is drastically reduced to less than 3 cm/s when a small amount of doping impurity is added during atomic hydrogen treatment. 相似文献

This study investigates the effect of microwaves on the dehydrogenation of decalin and tetralin. The reactions were studied in a fixed-bed reactor under microwave heating (MWH), and the results were compared with those under conventional heating (electrical heating) (CH). The result of this investigation showed that under the same operating conditions, increase in the conversion of tetralin in the microwave-heated system was more than that in the conventional heated system. This demonstrated the microwave effect appeared in the dehydrogenation of tetralin. In addition, the activity and stability of the catalyst also improved under MWH. The microwave effect resulted in increase in the conversion reaction of tetralin dehydrogenation by generation of a large temperature gradient from the catalyst surface to surrounding species and the subsequent mass transfer in which both vectors have direction inverted from that of CH. Such a phenomenon induced faster molecule desorption (product species) or enhancement of species transport in the system. In addition, the contiguous strong adsorption of hydrocarbon can be reduced, leading to decrease in coke deposition. Conditionally, this benefit issue particularly resulted in increase in the reaction rate in which species transport or mass diffusion is the rate-limiting step. 相似文献

Minimally invasive boron neutron capture therapy (BNCT) is an elegant approach for cancer treatment. The highly selective and efficient deliverability of boron agents to cancer cells is the key to maximizing the therapeutic benefits of BNCT. In addition, enhancement of the frequencies to achieve boron neutron capture reaction is also significant in improving therapeutic efficacy by providing a highly concentrated boron agent in each boron nanoparticle. As the density of the thermal neutron beam remains low, it is unable to induce high-efficiency cell destruction. Herein, we report phospholipid-coated boronic oxide nanoparticles as agents for BNCT that can provide a highly concentrated boron atom in each nanoparticle. The current system exhibited in vitro BNCT activity seven times higher than that of commercial boron agents. Furthermore, the system could penetrate cancer spheroids deeply, efficiently suppressing thermal neutron irradiation-induced growth. 相似文献

Amyotrophic lateral sclerosis (ALS) is the third most common neurodegenerative disorder and is sometimes associated with frontotemporal dementia. Charcot–Marie–Tooth disease (CMT) is one of the most commonly inherited peripheral neuropathies causing the slow progression of sensory and distal muscle defects. Of note, the severity and progression of CMT symptoms markedly vary. The phenotypic heterogeneity of ALS and CMT suggests the existence of modifiers that determine disease characteristics. Epigenetic regulation of biological functions via gene expression without alterations in the DNA sequence may be an important factor. The methylation of DNA, noncoding RNA, and post-translational modification of histones are the major epigenetic mechanisms. Currently, Drosophila is emerging as a useful ALS and CMT model. In this review, we summarize recent studies linking ALS and CMT to epigenetic regulation with a strong emphasis on approaches using Drosophila models. 相似文献

Porous diatomite ceramics were prepared by alkali treatment of the raw powder without any sintering procedure. The alkali treatment was carried out by dissolution of the surface by KOH solution to form solid crosslinking among the powder particles near room temperature. The effect of the amount and concentration of KOH solution on the properties of the porous diatomite ceramics was investigated. Variation of the treatment factors was found to be effective for controlling properties such as porosity, strength and thermal conductivity. Mixing a larger amount of high-concentration KOH solution with the raw powder could promote dissolution and the formation of thicker necks between particles by solid crosslinking. The resultant porous diatomite ceramics showed comparable compressive strength and thermal conductivity to those of sintered materials. The proposed method is therefore a useful tool for the preparation of high-performance porous ceramics near room temperature. 相似文献

We investigated the effects of esaxerenone, a novel, nonsteroidal, and selective mineralocorticoid receptor blocker, on cardiac function in Dahl salt-sensitive (DSS) rats. We provided 6-week-old DSS rats a high-salt diet (HSD, 8% NaCl). Following six weeks of HSD feeding (establishment of cardiac hypertrophy), we divided the animals into the following two groups: HSD or HSD + esaxerenone (0.001%, w/w). In survival study, all HSD-fed animals died by 24 weeks of age, whereas the esaxerenone-treated HSD-fed animals showed significantly improved survival. We used the same protocol with a separate set of animals to evaluate the cardiac function by echocardiography after four weeks of treatment. The results showed that HSD-fed animals developed cardiac dysfunction as evidenced by reduced stroke volume, ejection fraction, and cardiac output. Importantly, esaxerenone treatment decreased the worsening of cardiac dysfunction concomitant with a significantly reduced level of systolic blood pressure. In addition, treatment with esaxerenone in HSD-fed DSS rats caused a reduced level of cardiac remodeling as well as fibrosis. Furthermore, inflammation and oxidative stress were significantly reduced. These data indicate that esaxerenone has the potential to mitigate cardiac dysfunction in salt-induced myocardial injury in rats. 相似文献

We have found that antisense oligonucleotides and siRNA molecules modified with repeat structures of disulfide units can be directly introduced into the cytoplasm and exhibit a suppressive effect on gene expression. In this study, we analyzed the mechanism of cellular uptake of these membrane-permeable oligonucleotides (MPONs). Time-course analysis by confocal microscopy showed that the uptake of MPONs from the plasma membrane to the cytoplasm reached 50 % of the total uptake in about 5 min. In addition, analysis of the plasma membrane proteins to which MPONs bind, identified several proteins, including voltage-dependent anion channel. Next, we analyzed the behavior of MPONs in the cell and found them to be abundant in the nucleus as early as 24 h after addition with the amount increasing further after 48 and 72 h. The amount of MPONs was 2.5-fold higher than that of unmodified oligonucleotides in the nucleus after 72 h. We also designed antisense oligonucleotides and evaluated the effect of MPONs on mRNA exon skipping using DMD model cells; MPONs caused exon skipping with 69 % efficiency after 72 h, which was three times higher than the rate of the control. In summary, the high capacity for intracytoplasmic and nuclear translocation of MPONs is expected to be useful for therapeutic strategies targeting exon skipping. 相似文献

A mobile manipulator that consists of a robot manipulator and an Automated Guided Vehicle (AGV) was developed to automate transporting and exchanging workpieces for machine tools. Despite less accurate positioning of the AGV, positioning accuracy of 1 mm must be realized during attachment and removal of workpieces. To compensate for an error of the AGV, this paper proposes a novel method of high-accuracy pose estimation using a fiducial marker. Experimental results show that workpiece exchange can be automated with high reliability even though a clearance between a chuck and workpieces in diameter is as small as 1 mm. 相似文献
