Air filters collect particles by the mechanical collection mechanisms, namely, inertia, interception, gravitational settling, and Brownian diffusion. There exists the most penetrating particle size (MPPS) in submicron size range for which none of the collection mechanisms work effectively. In this study, we propose a new type of filter named as “centrifugal filter,” which collects aerosol particles by centrifugal force together with the conventional mechanical collection mechanisms. The centrifugal filter proposed in the present work may be rotated by a motor or compressed air. Air passes through the filter in the axial direction of filter rotation. The filter rotates so does the air embedded in the filter, and therefore centrifugal force exerts on particles. In addition to the mechanical collection mechanisms, small migration of particles due to the centrifugal force enhanced the collection efficiency of submicron particles significantly without increasing the pressure drop. The performance tests of centrifugal filter were conducted by changing the fiber diameter, the air flow velocity and the rotation speed. We found that the collection efficiency of filter is enhanced significantly by rotating the filter without increasing the pressure drop and that the filter efficiency is well predicted by the conventional filtration theory accounting for the centrifugal force.

Zeolite-supported highly dispersed Co sulfide clusters are synthesized using Co(CO)3 (NO) as a precursor. The amount of Co inCoSx/zeolite anchored by a CVD technique increases as the Al/Si ratio of the zeolite increases, whereas the activity per Co atomdecreases for thiophene hydrodesulfurization (HDS). When an Na-exchanged USY zeolite is used, the Co sulfide catalyst shows a muchhigher HDS activity than a conventional Co—Mo/Al2 O3 catalyst. It is considered from XAFS and NO adsorption techniques that thehigh HDS activity of CoSx/USY-Na is due to an extremely high dispersion of Co sulfide clusters. Recent studies indicate that Caenorhabditis elegans CED-4 interacts with and promotes the activation of the death protease CED-3, and that this activation is inhibited by CED-9. Here we show that a mammalian homolog of CED-4, Apaf-1, can associate with several death proteases, including caspase-4, caspase-8, caspase-9, and nematode CED-3 in mammalian cells. The interaction with caspase-9 was mediated by the N-terminal CED-4-like domain of Apaf-1. Expression of Apaf-1 enhanced the killing activity of caspase-9 that required the CED-4-like domain of Apaf-1. Furthermore, Apaf-1 promoted the processing and activation of caspase-9 in vivo. Bcl-XL, an antiapoptotic member of the Bcl-2 family, was shown to physically interact with Apaf-1 and caspase-9 in mammalian cells. The association of Apaf-1 with Bcl-XL was mediated through both its CED-4-like domain and the C-terminal domain containing WD-40 repeats. Expression of Bcl-XL inhibited the association of Apaf-1 with caspase-9 in mammalian cells. Significantly, recombinant Bcl-XL purified from Escherichia coli or insect cells inhibited Apaf-1-dependent processing of caspase-9. Furthermore, Bcl-XL failed to inhibit caspase-9 processing mediated by a constitutively active Apaf-1 mutant, suggesting that Bcl-XL regulates caspase-9 through Apaf-1. These experiments demonstrate that Bcl-XL associates with caspase-9 and Apaf-1, and show that Bcl-XL inhibits the maturation of caspase-9 mediated by Apaf-1, a process that is evolutionarily conserved from nematodes to humans.  相似文献

We have identified and characterized Diva, which is a novel regulator of apoptosis. Sequence analysis revealed that Diva is a member of the Bcl-2 family of proteins containing Bcl-2 homology domain 1, 2, 3, and 4 (BH1, BH2, BH3, and BH4) regions and a carboxyl-terminal hydrophobic domain. The expression of Diva mRNA was detected in multiple embryonic tissues but was restricted to the ovary and testis in adult mice. The expression of Diva promoted the death of 293T, Ramsey, and T47D cells as well as that of primary sensory neurons, indicating that Diva is a proapoptotic protein. Significantly, Diva lacks critical residues in the conserved BH3 region that mediate the interaction between BH3-containing proapoptotic Bcl-2 homologues and their prosurvival binding partners. Consistent with this, Diva did not bind to cellular Bcl-2 family members including Bcl-2, Bcl-XL, Bcl-w, Mcl-1, and A1/Bfl-1. Furthermore, mutants of Diva lacking the BH3 region fully retained their proapoptotic activity, confirming that Diva promotes apoptosis in a BH3-independent manner. Significantly, Diva interacted with a viral Bcl-2 homologue (vBcl-2) encoded by the Kaposi's sarcoma-associated herpesvirus. Consistent with these associations, apoptosis induced by Diva was inhibited by vBcl-2 but not by Bcl-XL. Importantly, Diva interacted with Apaf-1, an adapter molecule that activates caspase-9, a central death protease of the apoptotic pathway. The expression of Diva inhibited the binding of Bcl-XL to Apaf-1, as determined by immunoprecipitation assays. Thus, Diva represents a novel type of proapoptotic Bcl-2 homologue that promotes apoptosis independently of the BH3 region through direct binding to Apaf-1, thus preventing Bcl-XL from binding to the caspase-9 regulator Apaf-1.  相似文献

Summary 7,7-Dicyanoquinone methides (1a-b) copolymerized spontaneously with substituted -acetoxystyrenes (2a-b) in benzene at 60 °C to give alternating copolymers (poly(1a-b-co-2a-b)) with the molecular weights of 8–60×104 in 42–66% yields. When these copolymers were heated under nitrogen, they eliminated acetic acid quantitatively at around 200 °C to be converted to the copolymers (poly(3a-d)) with olefinic bonds, which were characterized by infrared and nuclear magnetic resonance spectroscopy. Alternating copolymers and the corresponding copolymers with olefinic bonds were amorphous, and capable of being cast from their chloroform solutions into transparent, tough films.  相似文献

In the preceding paper in this issue, we described the overproduction of one mutant chicken lysozyme in Escherichia coil.Since this lysozyme contained two amino acid substitutions (Ala31ValandAsn106Ser)in addition to an extra methionine residue at theNH2-terminus the substituted amino acid residues were convertedback to the original ones by means of oligonucleotide-directedsite-specific mutagenesis and in vitro recombination. Thus fourkinds of chicken lysozyme [Met–1 Val31Ser106-, Met–1Ser106-,Met–1 Val31-and Met–1 (wild type)] wereexpressed in E. coli. From the results of folding experimentsof the reduced lysozymes by sulfhydryl-disulfide interchangeat pH 8.0 and 38°C, follow ed by the specific activity measurementsof the folded en zymes, the following conclusions can be drawn:(i) an extra methionine residue at the NH2-terminus reducesthe folding rate but does not affect the lysozyme activity ofthe folded enzyme; (ii) the substitution of Asn106 by Ser decreasesthe activity to 58% of that of intact native lysozyme withoutchanging the folding rate; and (iii) the substitution of Ala31Val prohibits the correct folding of lysozyme. Since the wildtype enzyme (Met–1-lysozyme) was activated in vitro withoutloss of specific activity, the systems described in this study(mutagenesis, overproduction, purification and folding of inactivemutant lysozymes) may be useful in the study of folding pathways,expression of biological activity and stability of lysozyme.  相似文献

An atomic oxygen (AO) source has been redesigned to coordinate with a pulsed laser deposition system and used to grow nitrogen-doped TiO(2) films by deposition of TiN and simultaneous irradiation of the substrate with AO. The AO source uses an incandescently heated thin tube of zirconia as an oxygen permeation media to generate pure AO of low kinetic energy. The emission flux is calibrated using a silver-coated quartz crystal microbalance. The thin shape of the probe and transverse emission geometry of this emission device allow the emission area to be positioned close to the substrate surface, enhancing the irradiation flux at the substrate. AO irradiation is crucial for formation of TiO(2) phases via oxidation of the deposited TiN laser plume, and is effective for decrease of the substrate temperature for crystallization of anatase phase to as low as around 200 °C.  相似文献

This paper presents a new approach to the optimal placement of a step voltage regulator (SVR) taking into consideration the installation of distributed generators (DG) based on a genetic algorithm (GA). In this case, it is necessary to solve problems such as the upper and lower limits for the voltage at each node and line capacity. In addition, given that the load and output of a distributed generator change, it is important to solve more than one of these problems. In order to solve these problems, the conventional genetic algorithm was improved and the proposed method was applied to the complex operating conditions of a distribution power system. © 2012 Wiley Periodicals, Inc. Highly charged macromolecular ions exhibit various conformations in gas. Intramolecular charge-to-charge interaction induces a transformation from a globular structure into a stretched one. The change in the molecular conformation brings a complex dynamic behavior of ions under an electrostatic field. In the present study, we visualized the movement of a monovalent and multiply charged straight chain macromolecules, polyethylene glycol (PEG), by a molecular dynamics (MD) simulation. The simulation showed that a singly charged PEG ion (899?<?MW < 4,643) takes a globular conformation. The electrical mobilities of these ions determined from the migration distance under an electric field were compared with the experimental data and those determined by the classical Mason–Schamp theory under an assumption of spherical shape. As a result, we obtained a good agreement between the MD, theoretical, and experimental data for the monovalent ions. We also found that the MD simulation successfully predicts the electrical mobility of the multiply charged stretched PEG ions, but the classical theory fails. We were able to visualize the periodic bending and stretching motion by the MD simulation. This unique motion results from the localization of charges on the PEG molecule and may have a significant effect on the dynamic behavior of macromolecule ions in gas.
