Micrometer- to nanometer-scale structures of the cut surfaces of single- and polycrystalline diamonds by a pulsed ultraviolet laser have been thoroughly investigated by scanning and transmission electron microscopy. Within the laser-cut grooves, the processed diamond surfaces are extensively covered with laser-modified debris which consists of complex layered units of graphite with various crystallinities. The units consist of 1) highly oriented graphite, 2) corrugated graphite, and 3) nanocrystalline graphite, which are sequentially located from the surface of the underlying diamond substrate to the center of the grooves. Detailed textural examinations revealed that the highly oriented graphite unit is a product of the initial graphitization of diamond by a solid-state diffusion process, whereas the latter two units are deposition products from the liquid and/or vapor phases of carbon in the later stage. The present study demonstrates that the laser-cutting of diamonds proceeds in a two-step process: 1) extensive graphitization of laser-scanning path and 2) subsequent sublimation of the pre-formed graphite. These processes are basically identical among the three different types of diamonds (single crystal type Ib, single crystal type IIa and nano-polycrystalline aggregate) tested in this study. 相似文献

The Fushime geothermal field is located in a depression close to the coast line. The system is characterized by very high reservoir temperature (>350°C), and a high salinity production fluid. Geological analysis shows that the main reservoir in this field occurs in a fractured zone developed around a dacite intrusion located in the center of the field. High permeability zones recognized by drilling data are found to be associated with fault zones. One of these zones is clearly associated with a NW–SE trending andesite dike swarm which was encountered in some wells.Alteration in the system can be divided into four zones, in order of increasing temperature, based on calcium–magnesium aluminosilicate mineral assemblages: i.e., the smectite, transition, chlorite and epidote zones. The feed zone is located in the chlorite and epidote zones, which can be further divided into three sub-zones according to their potassium or sodium aluminosilicate mineralogy, from the center of the discharge zone: K-feldspar–quartz, sericite–quartz, and albite–chlorite zones.Chloride concentration of the sea-water is 19,800 mg/l, and Br/Cl mole ratio is 1.55. Based on geochemical information, the reservoir chloride concentration of this field ranges from 11,600 to 22,000 mg/kg. The Clres (Cl in reservoir), Br/Cl ratios and stable isotope data indicate that the Fushime geothermal fluid originated from sea-water and is diluted by ground water during its ascent. Some fluids produced from geothermal wells show low pH (about 4). It is thought that sulfide mineral (PbS, ZnS) precipitation during production produces this acidic fluid. 相似文献

Photoluminescence depending on nitrogen concentration was investigated using anatase-type TiO2 prepared by the calcination of a mixture of titanyl sulfate hydrate and urea. The substitutional ratio (x) of nitrogen in TiO2 was successfully varied from 0.004 to 0.022 by changing the molar ratio of the mixture. The absorbance at 380–560 nm due to the formation of mid-gap states was proportional to the substitutional ratio of nitrogen controlled by the preparation conditions. In contrast, the fluorescent intensity at 382 nm originating from the band-to-band transition monotonically decreased with an increase in the substitutional ratio with an expansion of the anatase lattice. On the other hand, the maximum intensity of photoluminescence at 560 nm excited at 350 nm, which could be associated with the transition from the conduction band to the mid-gap states, was observed at x = 0.01. The optimal substitutional ratio for the emission was almost agreed with that for the photocatalytic decomposition of methylene blue and acetaldehyde under visible-light illumination. The photoluminescence was fundamentally determined by the balance between photoexcitation originating from a sufficient number of mid-gap states and deactivation of excited electrons and holes due to lattice distortion or defective states induced with the nitrogen doping. 相似文献

We describe an investigation into the fatigue fracture behaviour under combined tension–torsion loading of a SiC whisker-reinforced A6061 aluminium alloy fabricated by a squeeze casting process. Special attention was paid to the environmental effects on fatigue fracture behaviour. Tests were conducted on both the composite and its unreinforced matrix material, A6061-T6, under load-controlled conditions with a constant value of the combined stress ratio, α = τmax /σmax in laboratory air or in a 3.5% NaCl solution at the free corrosion potential. The corrosion fatigue strength of both the matrix and composite was less in the solution than in air. The dominating mechanical factor that determined the fatigue strength in air was either the maximum principal stress or the von Mises-type equivalent stress, depending on the combined stress ratio. However, in the 3.5% NaCl solution, the corrosion fatigue strength of both materials was determined by the maximum principal stress, irrespective of the combined stress ratio. In the case of the matrix material, crack initiation occurred by a brittle facet normal to the principal stress due to hydrogen embrittlement. However, in the composite material, the crack was initiated not at the brittle facet, but at a corrosion pit formed on the specimen surface. At the bottom of the pit, a crack normal to the principal stress was nucleated and propagated, resulting in final failure. Pitting corrosion was nucleated at an early stage of fatigue life, i.e. about 1% of total fatigue life. However, crack initiation at the bottom of a pit was close to the terminal stage, i.e. about 70% or more of total fatigue life. The dominating factor which determined crack initiation at a pit was the Mode I stress intensity factor obtained by assuming the pit to be a sharp crack. Initiation and propagation due to pitting corrosion and crack growth were closely examined, and the fatigue fracture mechanisms and influence of the 3.5% NaCl solution on fatigue strength of the composite and matrix under combined tension–torsion loading were examined in detail. 相似文献

CRK is a human homolog of chichen v-Crk, which is an adaptor protein. The SH2 domain of CRK binds to several tyrosine-phosphorylated proteins, including the epidermal growth factor receptor, p130(Cas), Shc, and paxillin. The SH3 domain, in turn, binds to cytosolic proteins of 135-145, 160, 180, and 220 kDa. We screened expression libraries by Far Western blotting, using CRK SH3 as a probe, and identified partial cDNA sequences of four distinct proteins, including C3G, DOCK180, EPS15, and clone ST12. The consensus sequence of the CRK SH3 binding sites as deduced from their amino acid sequences was Pro+3-Pro+2-X+1-Leu0-Pro-1-X-2-Lys-3. The interaction of the CRK SH3 domain with the DOCK180 peptide was examined with an optical biosensor, based on the principles of surface plasmon resonance. A low dissociation constant of the order of 10(-7) resulted from a high association rate constant (kassoc = 3 x 10(4)) and low dissociation rate constant (kdiss = 3 x 10(-3)). All CRK-binding proteins except clone ST12 also bound to another adaptor protein, Grb2. Mutational analysis revealed that glycine at position +1 of ST12 inhibited the binding to Grb2 while retaining the high affinity binding to CRK SH3. The result suggests that the amino acid at position +1 also contributes to the high affinity binding of the peptides to the SH3 domain of Grb2, but not to that of CRK. 相似文献

cDNAs encoding the bovine immunodeficiency virus (BIV) transactivator gene (tat) were cloned from virally infected cells and characterized. BIV expresses two distinct tat mRNAs composed of three exons that are derived by alternative splicing. The BIV tat mRNA splice variants encode Tat proteins of 103 (Tat103) and 108 (Tat108) amino acids. The Tat103 coding region is specified only by exon 2, while that of Tat108 is specified by a truncated exon 2 and the first 30 nt of exon 3. Thus, the first 98 amino acids of each Tat are identical, and have amino terminal, cysteine-rich, conserved core, basic, and carboxyl-terminal domains similar to Tats encoded by primate lentiviruses. BIV-infected bovine cells express a 14-kDa phosphorylated Tat protein identical in size to recombinant Tat expressed in bacteria. BIV Tat was shown to localize exclusively in the nucleoli of virally infected and Tat-expressing cells. Reporter gene assays indicated that Tat103 and Tat108 can strongly transactivate the BIV long terminal repeat (LTR) in virally permissive canine Cf2Th and nonpermissive HeLa and mouse NIH 3T3 cells, but not in permissive lapine EREp cells. However, an intact BIV tat gene is required for viral replication in both Cf2Th and EREp cells. Strong LTR activation by BIV Tat requires a TAR (transactivation responsive) element delimited by viral nt +1 to +31 and the Tat basic domain. BIV Tat strongly cross-transactivates the HIV-1 LTR in a TAR-dependent manner in Cf2Th, but not in EREp, HeLa, or NIH 3T3 cells. In contrast, strong, TAR-dependent cross-transactivation of the BIV LTR by HIV-1 Tat could not be demonstrated in any of these cell types. In Cf2Th cells Tat108 effects a moderately stronger transactivation of the BIV LTR than Tat103, indicative of a functional difference in BIV Tat proteins encoded by the mRNA splice variants. The present studies demonstrate that BIV Tat parallels the primate lentiviral Tats in structure and biochemistry but is not interchangeable with the latter. 相似文献

The structural change of carbon aerogels at high temperatures up to 2800°C has been investigated. Change in microtexture of fine particles, which constitute carbon aerogels derived from phenolic resin, was of a typical non-graphitized carbon. The microporosity decreased with an increase of heat-treatment temperature, and disappeared at 2000°C. The mesoporosity still remained even after heat-treatment up to 2800°C, though 50% of mesopore volume was lost because of the fusion of the particles with the change of carbon microtexture. 相似文献

Human polyomavirus JC virus (JCV), the causative agent of progressive multifocal leukoencephalopathy, is ubiquitous in humans, infecting children asymptomatically then persisting in renal tissue. Since JCV DNA can readily be detected from urine, it should be a useful tool with which to study the mode of virus transmission in humans. Based on this notion, we examined the extent to which JCV was transmitted from the American to Japanese populations in Okinawa Island, Japan. (A population of about 50 000 American soldiers and families have been stationed in Okinawa since 1945.) Four JCV types (A to D) were identified in American populations in U.S.A., whereas only type B was prevalent in elder Japanese in Okinawa who had reached adulthood by 1945. Thus, types A, C, and D served as indicators of the transmission of JCV from American to Japanese populations. We then examined whether types A, C, and D were detectable in Japanese in Okinawa aged 30-50 years who may have been in contact with Americans during childhood. However, all the 125 isolates from the younger Japanese population were type B without exception. From this finding, we concluded that JCV is rarely transmitted between human populations. 相似文献

Preoperative evaluation of the operative indication of hepatic resection using the parameters obtained by 99mTc-GSA scintigraphy has been done. In particular, the accurate evaluation of the postoperative hepatic functional reserve essentially depends on these parameters. In the present study, a preoperative evaluation of postoperative hepatic functional reserve using 99mTc-GSA scintigraphy was performed in our operated cases retrospectively. PATIENTS AND METHODS: Thirty-eight patients who underwent hepatic resection were studied on 99mTc-GSA scintigraphy before and after operation. These patients were divided into two groups. Group A; had no postoperative complications (n = 31). Group B; had some postoperative complications (n = 7). Preoperative parameters of 99mTc-GSA liver scintigraphy (HH15, LU15) were calculated from the activities of liver and cardiac ROIs at 5 and 15 minutes after injection. The resection ratio (RR) was obtained by comparing the liver volumes which were calculated from the pre- and postoperative SPECT studies. The resectability indices (Res) were as follows: Res (LU15) = LU15 x (100-RR(%)/100, Res (HH15) = (1/HH15) x (100-RR (%)/100. RESULTS: There were statistically significant differences in the distribution of Res between A and B groups (p = 0.002, Mann-Whitney test). The values of Res, of which half of patients have complication, were 1.10 (Res (HH15)) and 16.4 (Res (LU15)). CONCLUSION: The resectability indices using 99mTc-GSA liver scintigraphy are useful for the preoperative evaluation of the limitation of hepatic resection. 相似文献

The various potential factors affecting the performance of nanopillar chips on DNA separation were investigated from the viewpoints of both numerical calculations and actual experiments. To yield higher performance and replace the conventional DNA separation techniques such as microchip electrophoresis, the phenomenon specific to the nanopillar chips should be deeply understood. In this paper, although various factors affecting the performance of the nanopillar chips are considered, we focused on the effect of electroosmotic flow, which is particularly noticeable in quartz-made nanopillar chips. High-resolution separation of DNA was realized when an electroosmotic flow was suppressed by simply using a higher concentration of buffer, but DNA separation failed in the presence of an electroosmotic flow. It was confirmed from the numerical simulations and the direct observations that the deformation of DNA band during the injection process was induced by electroosmotic flow and consequently led to a poor resolution. 相似文献
